Povzetek: Za jutrišnjo hrano v svetu Čebele in drugi opraševalci so zelo pomembni za obstoj človeštva, saj bistveno prispevajo k varnosti preskrbe s hrano v svetu. Tretjina svetovne proizvodnje hrane oziroma vsaka tretja žlica hrane je odvisna od opraševanja, pri katerem imajo čebele najpomembnejšo vlogo. Visoko hranilni proizvodi, kot so med, matični mleček in cvetni prah, prav tako povečujejo hranilno vrednost živil. Živila, ki so odvisna od opraševalcev, prispevajo k zdravi prehrani, saj so bogat vir nujno potrebnih hranil.

Povzetek: A list of general minimum criteria was developed to be used in preparation of reference materials to be used in interlaboratory testing, including validations through test performance studies. The list was based on previously identified criteria in standards including ISO standards, EPPO guidelines, deliverables of relevant projects, related fields, and own experience of project partners. Several additional criteria were proposed: ‘availability’, ‘purity’ and ‘commutability’. These criteria result from a series of discussions, taking into account previous work and relevant international standards. Where relevant, each criterion was first defined as a series of levels from the highest to lowest ranking with the lowest ranking considered to be the minimum. Depending on the intended use, the reference material may need to fulfil higher levels of selected criteria or some criteria may not be relevant at all. The criteria were tested by the organizers of the TPS in round 1 of validation within VALITEST project which include bacteria, viruses, nematodes and fungi. The systematic and structured approach of describing RMs was found to be useful in promoting transparent descriptions of the RMs used and comparability of TPSs, as well as a step toward implementing FAIR data principles. The criteria defined here are applicable, potentially with minor modification, beyond the scope of this deliverable and project to other pests and other uses of RMs

Povzetek: A general standard operating procedure (SOP) for the production of reference material (RM) for use in plant health diagnostics was developed. The general SOP was designed based on (limited) information on existing SOPs and guidelines available with the consortium partners. The general SOP describes the different steps required in the production process, ranging from the different possible sources of the reference material, tests to confirm its identity, possibly required multiplication steps to the actual production process. For each step in the process, criteria and critical points are identified. The criteria that reference material has to meet, and their minimum required levels as identified and described in Deliverable 3.1 of WP3 form an import basis of the final production process.

Dynamic balance and gait impairments in Parkinson’s disease : novel cholinergic patterns

Povzetek: The cholinergic system has been implicated in postural deficits, in particular falls, in Parkinson’s disease. Falls and freezing of gait typically occur during dynamic and challenging balance and gait conditions, such as when initiating gait, experiencing postural perturbations, or making turns. However, the precise cholinergic neural substrate underlying dynamic postural and gait changes remains poorly understood. The aim of this study was to investigate whether brain vesicular acetylcholine transporter binding, as measured with [18F]-fluoroethoxybenzovesamicolbinding PET, correlates with dynamic gait and balance impairments in 125 patients with Parkinson’s disease (mean age 66.89±7.71 years) using the abbreviated Balance Evaluation Systems Test total and its four functional domain sub-scores (anticipatory postural control, reactive postural control, dynamic gait, and sensory integration). Whole brain false discoverycorrected (P < 0.05) correlations for total abbreviated Balance Evaluation Systems Test scores included the following bilateral or asymmetric hemispheric regions: gyrus rectus, orbitofrontal cortex, anterior part of the dorsomedial prefrontal cortex, dorsolateral prefrontal cortex, cingulum, frontotemporal opercula, insula, fimbria, right temporal pole, mesiotemporal, parietal and visual cortices, caudate nucleus, lateral and medial geniculate bodies, thalamus, lingual gyrus, cerebellar hemisphere lobule VI, left cerebellar crus I, superior cerebellar peduncles, flocculus, and nodulus. No significant correlations were found for the putamen or anteroventral putamen. The four domain-specific sub-scores demonstrated overlapping cholinergic topography in the metathalamus, fimbria, thalamus proper, and prefrontal cortices but also showed distinct topographic variations. For example, reactive postural control functions involved the right flocculus but not the upper brainstem regions. The anterior cingulum associated with reactive postural control whereas the posterior cingulum correlated with anticipatory control. The spatial extent of associated cholinergic system changes were least for dynamic gait and sensory orientation functional domains compared to the anticipatory and reactive postural control functions. We conclude that specific aspects of dynamic balance and gait deficits in Parkinson’s disease associate with overlapping but also distinct patterns of cerebral cholinergic system changes in numerous brain regions. Our study also presents novel evidence of cholinergic topography involved in dynamic balance and gait in Parkinson’s disease that have not been typically associated with mobility disturbances, such as the right anterior temporal pole, right anterior part of the dorsomedial prefrontal cortex, gyrus rectus, fimbria, lingual gyrus, flocculus, nodulus and right cerebellar hemisphere lobules VI and left crus I.

The StPti5 ethylene response factor acts as a susceptibility factorby negatively regulating the potato immune responseto pathogens

Povzetek: Ethylene response factors (ERFs) have been associated with biotic stress in Arabidopsis, while their function in non-model plants is still poorly understood. Here we investigated the role of potato ERF StPti5 in plant immunity. We show that StPti5 acts as a susceptibility factor. It negatively regulates potato immunity against potato virus Y and Ralstonia solanacearum, pathogens with completely different modes of action, and thereby has a different role than its orthologue in tomato. Remarkably, StPti5 is destabilised in healthy plants via the autophagy pathway and accumulates exclusively in the nucleus upon infection. We demonstrate that StEIN3 and StEIL1 directly bind the StPti5 promoter and activate its expression, while synergistic activity of the ethylene and salicylic acid pathways is required for regulated StPti expression. To gain further insight into the mode of StPti5 action in attenuating potato defence responses, we investigated transcriptional changes in salicylic acid deficient potato lines with silenced StPti5 expression. We show that StPti5 regulates the expression of other ERFs and downregulates the ubiquitin-proteasome pathway as well as several proteases involved in directed proteolysis. This study adds a novel element to the complex puzzle of immune regulation, by deciphering a two-level regulation of ERF transcription factor activity in response to pathogens.

Comparison of the mineralogy and microstructure of EAF stainless steel slags with reference to the cooling treatment

Povzetek: TIn the present study the differences in the mineralogical composition and microstructure of various types of EAF stainless steel (EAF S) slag with regard to the cooling treatment, the operation practice in an EAF (electric arc furnace) and environmental ageing reactions were evaluated. It was shown that the mineralogy of the investigated EAF S slags varied from one slag to another, depending on the quality of the produced stainless steel. The production process of the treated steel also has a strong influence on the mineralogy of the slags. The conditions during water cooling treatment were not sufficient to prevent the crystallization of primary mineral phases, which occurs predominantly in air-cooled EAF S slags, probably due to the high basicity of the investigated slags. However, the water cooling treatment of hot slag leads to the absence of γ-CaSiO 4 and the formation of secondary mineral phases predominantly calcite, portlandite, ettringite, calcium aluminate hydrate and calcium silicate hydrate. It has been shown that during the environmental ageing test (down-flow column test) secondary mineral phases were formed, which were the same as those formed during the water cooling treatment.

Tailoring the crystalline and amorphous phase ratios of TiO2 through the use of organic additives during hydrothermal synthesis

Povzetek: The photocatalytic properties of TiO2 are primarily determined by its crystallinity and crystalline phase ratios. To improve the photocatalytic properties of TiO2, greater control over the formation of crystalline and amorphous phases during synthesis is therefore required. In this study, we demonstrate how the addition of minute amounts of three organic compounds (isopropanol, acetone and acetic acid) during hydrothermal treatment affects the amorphous and crystalline phase ratios: the addition of isopropanol or acetone accelerates the phase transition from anatase and brookite to rutile, whereas the addition of acetic acid inhibits the transformation of anatase to rutile, increasing the content of amorphous phase compared to samples where no organic compound was added. We show that the combination of the organic compound added, along with the duration of the hydrothermal treatment, can be used to tailor the phase composition of TiO2, so as to obtain either: i) TiO2 with a high content of both rutile and amorphous phase, ii) TiO2 with a high rutile content and iii) TiO2 with different ratios of all four phases, when the duration of synthesis is short (2–4 h). The materials synthesized exhibited high photocatalytic activity (in most cases higher than P25), which is attributed to the beneficial phase composition and high specific surface area.
