Genetic aspects linked to production and use of forest reproductive material (FRM) : collecting scientific evidence for developing guidelines and decision support tools for effective FRM management

Povzetek: This report summarises the current state of knowledge in genetic aspects linked to the production and use of forest reproductive material (FRM). The first chapter describes the different categories of FRM, considers the strategic choice between artificial and natural regeneration in the face of climate change, and articulates the need for and purposes of FRM. Chapter 2 outlines the FRM production chain, in terms of: i) FRM approval, management, collection and certification (seed sources, stands, orchards, plus trees and clones others); ii) Testing standards and examples; iii) Effect of seed and seedling material not intended for use as FRM on forest genetic resources; iv) Breeding effects on basic material including conservation strategy; v) Harvesting, processing and storage of seeds; vi) Nursery practices; vii) Certification and Traceability; and viii) FRM Trading and Transport. Chapter 3 examines FRM with regard to: i) Forest regeneration strategies % especially considering climate change, and ii) Assisted migration and available decision support tools. Chapter 4 provides a detailed overview of forest establishment methods, and chapter 5 looks at keeping records of seed origin to improve forest management. The final chapter offers 38 recommendations arising from the review. This work is the result of an international collaboration rooted in EUFORGEN for more than two decades. It is built on the firm belief that the genetic element is decisive for the creation of a resilient forest capable of surviving threats and adapting to changes, thus enabling the evolution of ecosystems and the conservation of the productive landscape.

Immunotherapy for metastatic non-small cell lung cancer : real-world data from an academic Central and Eastern European center

Povzetek: Background. Immunotherapy with immune checkpoint inhibitors (ICIs) recently became the standard treatment for patients with advanced non-small cell lung cancer (NSCLC). Here, we present the first results of a real-world observational study on the effectiveness of ICI monotherapy in patients with advanced NSCLC treated at a single academic center in a Central and Eastern European (CEE) country. Materials and methods. Overall, 66 consecutive patients with advanced NSCLC treated with ICIs in everyday clinical practice, either with first-line pembrolizumab (26 patients) or second-line atezolizumab, nivolumab, or pembrolizumab (40 patients), from August 2015 to November 2018, were included. All data were retrieved from a hospital lung cancer registry, in which the data is collected prospectively. Results. Included patients had a median age of 64 years, most were male (55%), 6% were in performance status >/=2, and 18% had controlled central nervous system metastases at baseline. In first-line, the median progression-free survival (mPFS) was 9.3 months, while the median overall survival (mOS) was not reached. The 1-year overall survival (OS) was 62%. In second-line, the mPFS and mOS were 3.5 months and 9.9 months, respectively, with a 1-year OS of 35%. In the overall population, adverse events of any grade were recorded in 79% of patients and of severe grade (3-4) in 12% of patients. Conclusion. The first real-world outcomes of NSCLC immunotherapy from a CEE country suggest comparable effectiveness to those observed in clinical trials and other real-world series, mainly coming from North America and Western European countries. Further data to inform on the real-world effectiveness of immunotherapy worldwide are needed.

Priporočila Onkološkega inštituta za zdravljenje bolnikov z adenokarcinomom želodca

Povzetek: V Sloveniji vsako leto zboli zaradi raka želodca okrog 450 bolnikov, od tega ima tri četrtine bolnikov ob postavitvi diagnoze razširjeno ali razsejano bolezen. Zato je pomembno, da imamo razvito sodobno multidisciplinarno zdravljenje. Onkološki inštitut je na poti pridobitve evropske onkološke akreditacije, ki jo podeljuje priznana Evropska organizacija onkoloških inštitutov (OECI), katere član je tudi Onkološki inštitut Ljubljana. Namen tega projekta je razvoj kliničnih poti, ki bodo omogočile celostno obravnavo bolnikov z rakom. Slovenskih priporočil za zdravljenje adenokarcinoma želodca trenutno nimamo, zato smo bili primorani sprejeti te, ki jim bomo sledili na Onkološkem inštitutu. V teh priporočilih, zaradi drugačne narave bolezni, ni zajeto zdravljenje raka ezofagogastričnega stika.

Empirical approach for modelling tree phenology in mixed forests using remote sensing

Povzetek: : Phenological events are good indicators of the effects of climate change, since phenological phases are sensitive to changes in environmental conditions. Although several national phenological networks monitor the phenology of different plant species, direct observations can only be conducted on individual trees, which cannot be easily extended over large and continuous areas. Remote sensing has often been applied to model phenology for large areas, focusing mostly on pure forests in which it is relatively easier to match vegetation indices with ground observations. In mixed forests, phenology modelling from remote sensing is often limited to land surface phenology, which consists of an overall phenology of all tree species present in a pixel. The potential of remote sensing for modelling the phenology of individual tree species in mixed forests remains underexplored. In this study, we applied the seasonal midpoint (SM) method with MODIS GPP to model the start of season (SOS) and the end of season (EOS) of six different tree species in Slovenian mixed forests. First, substitute locations were identified for each combination of observation station and plant species based on similar environmental conditions (aspect, slope, and altitude) and tree species of interest, and used to retrieve the remote sensing information used in the SM method after fitting the best of a Gaussian and two double logistic functions to each year of GPP time series. Then, the best thresholds were identified for SOS and EOS, and the results were validated using cross-validation. The results show clearly that the usual threshold of 0.5 is not best in most cases, especially for estimating the EOS. Despite the difficulty in modelling the phenology of different tree species in a mixed forest using remote sensing, it was possible to estimate SOS and EOS with moderate errors as low as <8 days (Fagus sylvatica and Tilia sp.) and <10 days (Fagus sylvatica and Populus tremula), respectively.

The influence of land use on the spatial distribution and intensity of heat islands in Slovenia

Povzetek: Heat islands (HI) are a common anthropogenic phenomenon and are defined as artificial surfaces (urban areas) that have a higher average temperature than their surroundings (rural areas). The aim of this work was to determine the influence of land use on the spatial distribution and intensity (HIi ) of HI in Slovenia. The MODIS Land Surface Temperature (LST) and Corine Land Cover (CLC) databases were used to perform the analysis. Within the identified HI, two HI levels were determined based on temperature difference. The results revealed a statistically significant negative correlation between HIi and both forest co%ver and forest fragmentation (forest edge density and ratio of mean forest patch size to HI size). Artificial surface was positively correlated with HIi . The results contribute to the understanding of the spatial distribution of HI and HIi and provide informati%on for spatial planning and policy-making to reduce the negative impact of HI.

Gene expression levels of the prolyl hydroxylase domain proteins PHD1 and PHD2 but not PHD3 are decreased in primary tumours and correlate with poor prognosis of patients with surgically resected non-small-cell lung cancer

Povzetek: Background: Hypoxia correlates with poor prognosis in several cancer types, including lung cancer. Prolyl hydroxylase domain proteins (PHDs) play a role in cell oxygen sensing, negatively regulating the hypoxia-inducible factor (HIF) pathway. Our study aim was to evaluate PHD1, PHD2 and PHD3 mRNA expression levels in primary tumours and normal lungs of non-small-cell lung cancer (NSCLC) patients and to correlate it with selected regulators of HIF signalling, with clinicopathological characteristics and overall survival (OS). Methods: Tumour tissue samples were obtained from 60 patients with surgically resected NSCLC who were treated with radical surgery. In 22 out of 60 cases, matching morphologically normal lung tissue was obtained. PHD1, PHD2 and PHD3 mRNA expressions were measured using RT-qPCR. Results: The PHD1 and PHD2 mRNA levels in primary tumours were significantly decreased compared to those in normal lungs (both p < 0.0001). PHD1 and PHD2 expression in tumours was positively correlated (rs = 0.82; p < 0.0001) and correlated well with HIF pathway downstream genes HIF1A, PKM2 and PDK1. Decreased PHD1 and PHD2 were associated with larger tumour size, higher tumour stage (PHD1 only) and squamous cell carcinoma. Patients with low PHD1 and patients with low PHD2 expression had shorter OS than patients with high PHD1 (p = 0.02) and PHD2 expression (p = 0.01). PHD1 showed borderline independent prognostic values in multivariate analysis (p = 0.06). In contrast, we found no associations between PHD3 expression and any of the observed parameters. Conclusions: Our results show that reduced expression of PHD1 and PHD2 is associated with the development and progression of NSCLC. PHD1 could be further assessed as a prognostic marker in NSCLC.
