Population differentiation in Acer platanoides L. at the regional scale—laying the basis for effective conservation of its genetic resources in Austria

Povzetek: Norway maple (Acer platanoides L.) is a widespread forest tree species in Central and Northern Europe but with a scattered distribution. In the debate on climate change driven changes in species selection in the forest, Norway maple has recently received raised interest because of its comparatively high drought resistance (higher than in sycamore maple). Therefore, it is an interesting species for sites high in carbonates and where other native tree species have become devastated by pathogens (e.g., elm, ash). In Austria, the demand on saplings is currently rising, while there is only very little domestic reproductive material available (on average more than 95% of saplings are imported from neighboring countries). This study was undertaken to identify genetic diversity and population structure of Norway maple in Austria to lay the foundation for the establishment of respective in situ and ex situ conservation measures. In addition, samples from planted stands and imported reproductive material from other countries were included to study the anthropogenic influence on the species in managed forests. We used 11 novel microsatellites to genotype 756 samples from 27 putatively natural Austrian populations, and 186 samples derived from two planted stands and five lots of forest reproductive material; in addition, 106 samples from other European populations were also genotyped. Cross species amplification of the new markers was tested in 19 Acer species from around the world. Population clustering by STRUCTURE analysis revealed a distinct pattern of population structure in Austria and Europe, but overall moderate differentiation. Sibship analysis identifies several populations with severe founding effects, highlighting the need for proper selection of seed sources of sufficient genetic diversity in the species.

Jet stream position explains regional anomalies in European beech forest productivity and tree growth

Povzetek: The mechanistic pathways connecting ocean-atmosphere variability and terrestrial productivity are well-established theoretically, but remain challenging to quantify empirically. Such quantification will greatly improve the assessment and prediction of changes in terrestrial carbon sequestration in response to dynamically induced climatic extremes. The jet stream latitude (JSL) over the North Atlantic-European domain provides a synthetic and robust physical framework that integrates climate variability not accounted for by atmospheric circulation patterns alone. Surface climate impacts of north-south summer JSL displacements are not uniform across Europe, but rather create a northwestern-southeastern dipole in forest productivity and radial-growth anomalies. Summer JSL variability over the eastern North Atlantic-European domain (5-40E) exerts the strongest impact on European beech, inducing anomalies of up to 30% in modelled gross primary productivity and 50% in radial tree growth. The net effects of JSL movements on terrestrial carbon fluxes depend on forest density, carbon stocks, and productivity imbalances across biogeographic regions.

A comparison of radial increment and wood density from beech provenance trials in Slovenia and Hungary

Povzetek: Provenance trials are a valuable source of information, especially in species such as European beech (Fagus sylvatica L.), which will likely increase its distribution due to global warming. The current study compares radial increment and wood density of beech provenances in the juvenile development stage from contrasting environments in Europe (Belgium, Slovenia, Czech Republic, Italy) planted at a mesic to wet site in Slovenia and a xeric site in Hungary. Existing data (past measurements of diameters and height) were combined with new measurements of tree height, diameter, dendrochronological and resistance drilling density measurements to assess differences in provenance radial growth. The wood density data were evaluated using a Bayesian general linear model. In order to study the differences in radial increment in more detail, two weather-wise contrasting years (2014 and 2017) were selected from the last decade, based on calculations of the 12-month Standardized Precipitation-Evapotranspiration Index. The differences in average tree-ring width among provenances at each sampled site appeared to be relatively small when averaged over a whole decade of data. However, according to year-to-year data, some provenances grew faster than others, especially in favorable weather conditions. In unfavorable conditions, the differences in tree-ring widths among provenances were smaller. For most provenances, variation in tree-ring widths within the same provenance increased in unfavorable conditions. The difference between the provenances with the highest and lowest wood densities at both locations did not exceed 5%. The model results indicate that the Idrija (Slovenia) provenance probably has a higher median wood density than other studied provenances at both sites. Although the current study confirmed some differences in wood density between provenances and trial locations, the differences are negligible in practice due to their low magnitude and the fact that the analyzed trees were still juvenile. As beech has a diffuse-porous wood, negligible differences in wood density would also be expected in adult trees. Beech provenances for planting in relation to changing weather should probably be chosen for their ability to survive more extreme weather events rather than to improve radial increment or wood density, especially as the differences in wood density of juvenile trees are relatively small.

Relationships between wood-anatomical features and resistance drilling density in Norway spruce and European beech

Povzetek: Environmental conditions affect tree-ring width (TRW), wood structure, and, consequently, wood density, which is one of the main wood quality indicators. Although studies on inter- and intra-annual variability in tree-ring features or density exist, studies demonstrating a clear link between wood structure on a cellular level and its effect on wood density on a macroscopic level are rare. Norway spruce with its simple coniferous structure and European beech, a diffuse-porous angiosperm species were selected to analyze these relationships. Increment cores were collected from both species at four sites in Slovenia. In total, 24 European beech and 17 Norway spruce trees were sampled. In addition, resistance drilling measurements were performed just a few centimeters above the increment core sampling. TRW and quantitative wood anatomy measurements were performed on the collected cores. Resistance drilling density values, tree-ring (TRW, earlywood width–EWW, transition-TWW, and latewood width–LWW) and wood-anatomical features (vessel/tracheid area and diameter, cell density, relative conductive area, and cell wall thickness) were then averaged for the first 7 cm of measurements. We observed significant relationships between tree-ring and wood-anatomical features in both spruce and beech. In spruce, the highest correlation values were found between TRW and LWW. In beech, the highest correlations were observed between TRW and cell density. There were no significant relationships between wood-anatomical features and resistance drilling density in beech. However, in spruce, a significant negative correlation was found between resistance drilling density and tangential tracheid diameter, and a positive correlation between resistance drilling density and both TWW + LWW and LWW. Our findings suggest that resistance drilling measurements can be used to evaluate differences in density within and between species, but they should be improved in resolution to be able to detect changes in wood anatomy.

Dynamic contrast-enhanced MRI in malignant pleural mesothelioma : prediction of outcome based on DCE-MRI measurements in patients undergoing cytotoxic chemotherapy

Povzetek: The malignant pleural mesothelioma (MPM) response rate to chemotherapy is low. The identification of imaging biomarkers that could help guide the most effective therapy approach for individual patients is highly desirable. Our aim was to investigate the dynamic contrast-enhanced (DCE) MR parameters as predictors for progression-free (PFS) and overall survival (OS) in patients with MPM treated with cisplatin-based chemotherapy. Methods: Thirty-two consecutive patients with MPM were enrolled in this prospective study. Pretreatment and intratreatment DCE-MRI were scheduled in each patient. The DCE parameters were analyzed using the extended Tofts (ET) and the adiabatic approximation tissue homogeneity (AATH) model. Comparison analysis, logistic regression and ROC analysis were used to identify the predictors for the patient's outcome. Results: Patients with higher pretreatment ET and AATH-calculated Ktrans and ve values had longer OS (P≤.006). Patients with a more prominent reduction in ET-calculated Ktrans and kep values during the early phase of chemotherapy had longer PFS (P =.008). No parameter was identified to predict PFS. Pre-treatment ET-calculated Ktrans was found to be an independent predictive marker for longer OS (P=.02) demonstrating the most favourable discrimination performance compared to other DCE parameters with an estimated sensitivity of 89% and specificity of 78% (AUC 0.9, 95% CI 0.74-0.98, cut off > 0.08 min-1). Conclusions: In the present study, higher pre-treatment ET-calculated Ktrans values were associated with longer OS. The results suggest that DCE-MRI might provide additional information for identifying MPM patients that may respond to chemotherapy.

Establishment of a virtual transborder tumor board for cancer patients in Central and Southeastern Europe : an initiative of the Central European Cooperative Oncology Group (CECOG)

Povzetek: o establish a transborder virtual tumor board (VTB) fostering state-of-the-art management of cancer patients by exchanging knowledge and expertise among oncologists in Central and Southeastern Europe (CEE). Methods: We established and implemented a VTB based on the WebEx platform. This allowed for password-protected and secure upload of patient cases to be presented and discussed among colleagues from various oncology centers scattered throughout CEE in order to arrive at a recommendation for further diagnoses and/or treatment. Results: A total of 73 cases from 16 oncology centers located in 11 CEE countries were uploaded by 22 physicians; 71 were discussed over the course of 17 virtual meetings between June 2018 and May 2019 and 12 different kinds of malignant diseases were discussed with lung cancer (46.6%), melanoma (19.2%) and bladder cancer (13.6%) being the most commonly presented tumor entities. Of the discussed patients, 93.3% had stage IV disease at the time of presentation, 62.6% received chemotherapy or targeted treatment and 67.1% were treated with immune checkpoint inhibitors (ICPIs). The most common causes for presentation and discussion of patient cases were related to the use of ICPIs (80%). Conclusion: When the need for expertise exceeds locally available resources, web-based VTBs provide a feasible way to discuss patient cases and arrive at conclusions regarding diagnoses and/or treatment across large geographic distances. Moreover, VTBs provide an innovative way for proper, state-of-the-art management of patients with malignant diseases in times of social distancing and the resulting need for restricted interaction during the current SARS-CoV‑2 (severe acute respiratory syndrome coronavirus type 2) pandemic.
