The Carniolan honeybee from Slovenia : a complete and annotated mitochondrial genome with comparisons to closely related Apis mellifera subspecies

Povzetek: The complete mitochondrial genome of the Carniolan honeybee (Apis mellifera carnica) from Slovenia, a homeland of this subspecies, was acquired in two contigs from WGS data and annotated. The newly obtained mitochondrial genome is a circular closed loop of 16,447 bp. It comprises 37 genes (13 protein coding genes, 22 tRNA genes, and 2 rRNA genes) and an AT-rich control region. The order of the tRNA genes resembles the order characteristic of A. mellifera. The mitogenomic sequence of A. m. carnica from Slovenia contains 44 uniquely coded sites in comparison to the closely related subspecies A. m. ligustica and to A. m. carnica from Austria. Furthermore, 24 differences were recognised in comparison between A. m. carnica and A. m. ligustica subspecies. Among them, there are three SNPs that affect translation in the nd2, nd4, and cox2 genes, respectively. The phylogenetic placement of A. m. carnica from Slovenia within C lineage deviates from the expected position and changes the perspective on relationship between C and O lineages. The results of this study represent a valuable addition to the information available in the phylogenomic studies of A. mellifera—a pollinator species of worldwide importance. Such genomic information is essential for this local subspecies’ conservation and preservation as well as its breeding and selection.

A methodological proposal for the climate change risk assessment of coastal habitats based on the evaluation of ecosystem services : lessons learnt from the INTERREG project ECO-SMART

Povzetek: Climate change is seriously impacting coastal biodiversity and the benefits it provides to humans. This issue is particularly relevant in the case of the European Union’s Natura 2000 network of areas for nature protection, where the sensitivity of local ecosystems calls for intervention to increase resistance and resilience to climate-related risks. Given the complex ways in which climate can influence conservation hotspot areas, there is a need to develop effective strategic approaches and general operational models to identify priorities for management and inform adaptation and mitigation measures. Here, a novel methodological proposal to perform climate risk assessment in Natura 2000 sites is presented that implements the systematic approach of ISO 14090 in combination with the theoretical framework of ecosystem services assessment and local stakeholder participation to identify climate-related issues for local protected habitats and improve the knowledge base needed to plan sustainable conservation and restoration measures. The methodology was applied to five Natura 2000 sites located along the Adriatic coast of Italy and Slovenia. Results show that each of the assessed sites, despite being along the coast of the same sea, is affected by different climate-related issues, impacting different habitats and corresponding ecosystem services. This novel methodology enables a simple and rapid screening for the prioritization of conservation actions and of the possible further investigations needed to support decision making, and was found to be robust and of general applicability. These findings highlight the importance of designing site-specific adaptation measures, tailored to address the peculiar response to climate change of each site in terms of biodiversity and ecosystem services.

Genomics and adaptation in forest ecosystems

Povzetek: Rapid human-induced environmental changes like climate warming represent a challenge for forest ecosystems. Due to their biological complexity and the long generation time of their keystone tree species, genetic adaptation in these ecosystems might not be fast enough to keep track with conditions changing at such a fast pace. The study of adaptation to environmental change and its genetic mechanisms is therefore key for ensuring a sustainable support and management of forests. The 4-day conference of the European Research Group EvolTree (https://www.evoltree.eu) on the topic of “Genomics and Adaptation in Forest Ecosystems” brought together over 130 scientists to present and discuss the latest developments and findings in forest evolutionary research. Genomic studies in forest trees have long been hampered by the lack of high-quality genomics resources and affordable genotyping methods. This has dramatically changed in the last few years; the conference impressively showed how such tools are now being applied to study past demography, adaptation and interactions with associated organisms. Moreover, genomic studies are now finally also entering the world of conservation and forest management, for example by measuring the value or cost of interspecific hybridization and introgression, assessing the vulnerability of species and populations to future change, or accurately delineating evolutionary significant units. The newly launched conference series of EvolTree will hopefully play a key role in the exchange and synthesis of such important investigations.

Farmers' preferences for result-based schemes for grassland conservation in Slovenia

Povzetek: Government-funded payments for ecosystem services (PES) have increasingly been used to facilitate transactions between users of environmental services and their providers. In order to improve the link between payments and the service provided, some countries in the EU have promoted result-based schemes (RBS), which remunerate farmers for ecological results, as part of their agricultural policy. Since PES programs are voluntary, it is important to understand farmers’ responses before more large-scale implementations of RBS are initiated. Using a choice experiment and a mixed logit model, we elicited the preferences of farmers in two Natura 2000 sites in Slovenia for different design elements of a hypothetical scheme for dry grassland conservation. We found that the majority of farmers preferred the result-based approach over the management-based scheme both in terms of payment conditions and monitoring; one group of farmers preferred the RBS very strongly (average WTA of more than 500 EUR/ha/yr) and another group less strongly (average WTA about 200 EUR/ha/yr). Farmers also showed a higher preference for on-farm advise and training in small groups than for lectures, which would be offered to a larger audience. A collective bonus, which would incentivise coordination and could potentially increase participation rates in the scheme, significantly influenced the farmers’ willingness to adopt the scheme. However, the estimated average WTA was comparable or lower than the 40 EUR/ha annual bonus payment. Older farmers and those who managed small and semi-subsistent farms were significantly more likely to be highly resistant to scheme adoption no matter its design.

The era of reference genomes in conservation genomics

Using genomic information for management planning of an endangered perennial, Viola uliginosa

Povzetek: Species occupying habitats subjected to frequent natural and/or anthropogenic changes are a challenge for conservation management. We studied one such species, Viola uliginosa, an endangered perennial wetland species typically inhabiting sporadi-cally flooded meadows alongside rivers/lakes. In order to estimate genomic diversity, population structure, and history, we sampled five sites in Finland, three in Estonia, and one each in Slovenia, Belarus, and Poland using genomic SNP data with double-digest restriction site-associated DNA sequencing (ddRAD-seq). We found mono-phyletic populations, high levels of inbreeding (mean population FSNP = 0.407-0.945), low effective population sizes (Ne = 0.8-50.9), indications of past demographic ex-pansion, and rare long-distance dispersal. Our results are important in implementing conservation strategies for V. uliginosa, which should include founding of seed banks, ex situ cultivations, and reintroductions with individuals of proper origin, combined with continuous population monitoring and habitat management.

Occurrence of invasive alien plant species in the floodplain forests along the Mura River in Slovenia

Povzetek: Background and purpose: The objectives of our study were to identify invasive alien plant species (IAS) in the main Natura 2000 forest habitat types (FHT) along the Mura River in Slovenia, and to estimate their abundance and cover. The aim of our study was to find out a) Which IAS appear in the research forests? b) What is their frequency and cover percentage? c) Whether individual IAS prosper better in some FHT than others? d) What is the correlation between the cover of IAS and the tree layer cover? Materials and methods: We analysed the fidelity of invasive plant species to individual FHT. The studied FHTs along the Mura River were the following: 91E0* (Alluvial forests with Alnus glutinosa and Fraxinus excelsior), 91F0 (Riparian mixed forests of Quercus robur, Ulmus laevis and Ulmus minor, Fraxinus excelsior or Fraxinus angustifolia, along the great rivers) and 91L0 (Illyrian oak-hornbeam forests). Two forest areas of about 600 ha were studied in total. With the intention to calculate number and cover of IAS some statistical analysis was made. In addition, correlations between the abundances of the most present IAS and cover of upper tree layer were carried out. Results: In total, 15 IAS were recorded in studied FHTs. Some species, like Robinia pseudoacacia, Impatiens glandulifera, I. parviflora, Fallopia japonica (incl. F. x bohemica), Erigeron annuus, Ambrosia artemisiifolia, Amorpha fruticosa, Conyza canadensis and Juncus tenuis occur only in one or two FHTs, while some species can be found in all studied FHTs (e.g. Solidago sp.). We found out that the most threatened forests are those with prevailing Salix alba, Alnus glutinosa, Fraxinus angustifolia and Ulmus laevis tree species. Those are the forests of FHT 91E0 which have less dense tree canopies, grow closest to the river and on the wettest sites. We found a statistically significant higher number and cover of IAS in the FHT 91E0 and the lowest number and cover in FHT 91L0. Conclusions: Alluvial forests with Alnus glutinosa and Fraxinus excelsior (FHT 91E0) along the Mura River are most prone to invasion of IAS. The increasing presence of IAS in the study areas seriously affects natural regeneration, stability, and continuity of floodplain forests in all other FHTs in the study area. At the same time the amount of IAS in these forests also depends on management measures and their intensities which accelerate light availability. Some measures and guidelines for managing of these forests with the purpose of reducing IAS impacts are suggested in this study.
