1. An outbreak of Verticillium dahliae on sycamore maple in a forest stand in SloveniaAna Brglez, Zina Devetak, Nikica Ogris, Sebastjan Radišek, Barbara Piškur, 2024, izvirni znanstveni članek Povzetek: Verticillium dahliae Kleb., the causal agent of Verticillium wilts, is a devastating plant disease affecting many plant species. Fungus V. dahliae was detected in a partially artificially established Acer pseudoplatanus L. forest stand in central Slovenia. This finding incited further study about the risk of different sources of V. dahliae isolates for maples in forests and the pathogenicity of three V. dahliae isolates of different origins was tested on saplings of A. pseudoplatanus, A. platanoides L., and A. campestre L. The inoculated saplings exhibited disease symptoms, i.e., leaf necrosis and wilting. At the end of the pathogenicity test, typical xylem browning was visible on the cross-sections, and the pathogen was successfully re-isolated. The isolates showed significant differences in their pathogenicity to specific maple hosts, with the agricultural isolate (originated from bell pepper) being the most aggressive on all three maple species. The disease severity index (DSI) and relative area under the disease progress curve (rAUDPC), as well as the success of re-isolation, indicate that A. platanoides is the most susceptible to inoculation with V. dahliae. In addition, significant differences in sapling biomass were observed between treated and control plants. These results suggest that maples in forest stands are threatened by V. dahliae, and biosecurity measures should be considered and implemented in forest management to reduce the transmission and potential spread of the pathogen. Candidate pathogenicity factor/effector proteins of ‘Candidatus Phytoplasma solani’ modulate plant carbohydrate metabolism, accelerate the ascorbate–glutathione cycle, and induce autophagosomesMarina Dermastia, Špela Tomaž, Rebeka Strah, Tjaša Lukan, Anna Coll Rius, Barbara Dušak, Timotej Čepin, Aleš Kladnik, Maja Zagorščak, Kristina Gruden, Maruša Pompe Novak, 2023, izvirni znanstveni članek Povzetek: The pathogenicity of intracellular plant pathogenic bacteria is associated with the action of pathogenicity factors/effectors, but their physiological roles for most phytoplasma species, including ‘Candidiatus Phytoplasma solani’ are unknown. Six putative pathogenicity factors/effectors from six different strains of ‘Ca. P. solani’ were selected by bioinformatic analysis. The way in which they manipulate the host cellular machinery was elucidated by analyzing Nicotiana benthamiana leaves after Agrobacterium-mediated transient transformation with the pathogenicity factor/effector constructs using confocal microscopy, pull-down, and co-immunoprecipitation, and enzyme assays. Candidate pathogenicity factors/effectors were shown to modulate plant carbohydrate metabolism and the ascorbate–glutathione cycle and to induce autophagosomes. PoStoSP06, PoStoSP13, and PoStoSP28 were localized in the nucleus and cytosol. The most active effector in the processes studied was PoStoSP06. PoStoSP18 was associated with an increase in phosphoglucomutase activity, whereas PoStoSP28, previously annotated as an antigenic membrane protein StAMP, specifically interacted with phosphoglucomutase. PoStoSP04 induced only the ascorbate–glutathione cycle along with other pathogenicity factors/effectors. Candidate pathogenicity factors/effectors were involved in reprogramming host carbohydrate metabolism in favor of phytoplasma own growth and infection. They were specifically associated with three distinct metabolic pathways leading to fructose-6-phosphate as an input substrate for glycolysis. The possible significance of autophagosome induction by PoStoSP28 is discussed. 4. First report of Diplodia fraxini and Diplodia subglobosa causing canker and dieback of Fraxinus excelsior in SloveniaBenedetto Teodoro Linaldeddu, Carlo Bregant, Lucio Montecchio, Ana Brglez, Barbara Piškur, Nikica Ogris, 2022, drugi znanstveni članki Povzetek: Over the last decades the vitality and productivity of European ash trees in Slovenia has been seriously impacted by the onset of canker and dieback disease symptoms on young and old trees, primarily identified as ash dieback caused by Hymenoscyphus fraxineus. Given the limited information available about the aetiology of this emerging disease, a study was carried out to isolate, identify and characterize the fungal species involved in the observed ash symptoms. Field surveys were conducted in five forest sites where 50 symptomatic branch samples were collected. All samples were inspected and used for fungal isolation. Based on morphology, colony appearance and DNA sequence data of the internal transcribed spacer region (ITS), 125 fungal colonies belonging to five species were isolated and identified. Only a few symptomatic ash samples yielded colonies of H. fraxineus, whereas Botryosphaeriaceae species were isolated with a high frequency, with Diplodia fraxini as the dominant species. A pathogenicity test proved that all isolated species were pathogenic on European ash, causing bark lesions and wood discoloration. All Botryosphaeriaceae species isolated in this study are reported for the first time on European ash in Slovenia. 5. Drought stress can induce the pathogenicity of Cryptostroma corticale, the causal agent of sooty bark disease of sycamore mapleNikica Ogris, Ana Brglez, Barbara Piškur, 2021, izvirni znanstveni članek Povzetek: Reports of sooty bark disease of maples caused by the fungus Cryptostroma corticale have recently been emerging from across Europe. The aims of our study were to describe the first report of sooty bark disease in Slovenia, to determine the pathogenicity of C. corticale, to confirm the optimum temperature for the growth of the fungus, and to determine the mass loss of Acer pseudoplatanus wood inoculated by C. corticale. We confirmed the presence of C. corticale on A. pseudoplatanus via morphological and molecular analysis. The optimal growth of C. corticale was measured in vitro on potato dextrose agar and was determined to occur at 25 °C. Pathogenicity tests were performed on 30 saplings of A. pseudoplatanus under two treatments, humid and drought stress, and the fungus was pathogenic in both treatments. The mean length of bark lesions and wood discoloration of the drought-stressed saplings was significantly greater than that in the humid treatment. Re-isolations of C. corticale were successful from all inoculated saplings, and thus Koch%s postulates were confirmed. The mass loss of A. pseudoplatanus wood was determined by mini-block test in a period of 10 weeks and was observed as minimal. Based on the results, we conclude that C. corticale is a weak and opportunistic pathogen that most likely expresses itself intensively under hot and dry conditions. 6. Pseudodidymella fagi in Slovenia : first report and expansion of host rangeNikica Ogris, Ana Brglez, Barbara Piškur, 2019, izvirni znanstveni članek Povzetek: The fungus Pseudodidymella fagi is spreading in Europe and causing leaf blotch of European beech, Fagus sylvatica. Between 2008 and 2017, outbreaks of P. fagi were observed on European beech in Switzerland, Germany (also on F. orientalis), Austria, and Slovakia. In Slovenia, leaf blotch symptoms were first observed on F. sylvatica in 2018. P. fagi was identified as the causal agent of the observed symptoms in Slovenia by morphological examinations together with sequencing of the internal transcribed spacer (ITS) region of the rDNA. This study links the fungus to the expansion of the known distribution of the disease to Slovenia, and based on in vitro pathogenicity trials, also to a new potential host, Quercus petraea. The pathogenicity tests confirmed F. sylvatica and F. orientalis as hosts for P. fagi, but not Castanea sativa, where pathogenicity to F. orientalis was proved for first time in vitro. Although Koch%s postulates could not be proven for C. sativa, it seems to be partially susceptible in vitro because some of the inoculation points developed lesions. Additionally, damage to Carpinus betulus related to P. fagi near heavily infected beech trees was observed in vivo but was not tested in laboratory trials. Based on the results and our observations in the field, it is likely that P. fagi has a wider host range than previously thought and that we might be witnessing host switching.
