Moss biomonitoring of the distribution of chemical elements in the air in the Prespa region, North Macedonia

Povzetek: A study was conducted to investigate atmospheric deposition and to explore the natural distribution and possible contamination with potentially toxic elements (PTEs) in the Prespa region, North Macedonia, using moss samples as biomonitors for air pollution. The distribution of 19 chemical elements (Ag, Al, Ba, Ca, Cr, Cu, Fe, K, Li, Mg, Mn, Mo, Na, Ni, P, Pb, Sr, V, and Zn) was detected in 11 moss samples from this area. The moss samples were analysed after microwave digestion using inductively coupled plasma – atomic emission spectrometry (ICP-AES). R-mode factor analysis was used to identify and characterise the elemental associations, and four associations of elements were identified. Four factors were separated from the group of elements: Factor 1 (Fe-Al-Cr-V-Ni), Factor 2 (Sr-Ba-Mg), Factor 3 (K-P-Zn) and Factor 4 (Pb-Cu). All element factors were found to be typical geochemical associations, with the exception of the distribution of K and P in the agricultural areas of the study area where fertilisers are used over a long period of time.

The importance of flaps in reconstruction of locoregionally advanced lateral skull-base cancer defects : a tertiary otorhinolaryngology referral centre experience

Povzetek: Background. The aim of the study was to identify the value of extensive resection and reconstruction with flaps in the treatment of locoregionally advanced lateral skull-base cancer.Patients and methods. The retrospective case review of patients with lateral skull-base cancer treated surgically with curative intent between 2011 and 2019 at a tertiary otorhinolaryngology referral centre was made. Results. Twelve patients with locoregionally advanced cancer were analysed. Lateral temporal bone resection was performed in nine (75.0%), partial parotidectomy in six (50.0%), total parotidectomy in one (8.3%), ipsilateral selective neck dissection in eight (66.7%) and ipsilateral modified radical neck dissection in one patient (8.3%). The defect was reconstructed with anterolateral thigh free flap, radial forearm free flap or pectoralis major myocutaneous flap in two patients (17.0%) each. Mean overall survival was 3.1 years (SD = 2.5) and cancer-free survival rate 100%. At the data collection cut-off, 83% of analysed patients and 100% of patients with flap reconstruction were alive. Conclusions. Favourable local control in lateral skull-base cancer, which mainly involves temporal bone is achieved with an extensive locoregional resection followed by free or regional flap reconstruction. Universal cancer registry should be considered in centres treating this rare disease to alleviate analysis and multicentric research.

Challenges in harmonized assessment of heavy metals in the Adriatic and Ionian Seas

Povzetek: The Adriatic-Ionian region (ADRION Region) shows strong development in terms of urban expansion in coastal and inland areas as well as increasing maritime traffic and offshore hydrocarbon extraction activities. A serious risk of pollution arises from hazardous substances requiring reliable and coherent monitoring and assessment programs. EU Directives (WFD – Water Framework Directive, MSFD – Marine Strategy Framework Directive) and Barcelona Convention protocols, aim to assess the level of pollution with the objective to implement measures to prevent and/or mitigate impacts on the marine environment. This high level integration process has to be based on common and agreed protocols for monitoring of contaminants. Aiming to share best practices to encourage a harmonized implementation of monitoring and assessment of contaminants, an extensive review of monitoring and analytical protocols adopted by six EU and non-EU countries along the Adriatic and Ionian seas was carried out in the framework of the Interreg Adrion project HarmoNIA (Interreg V-B Adriatic-Ionian (ADRION), 2018–2020). This paper presents a methodological proposal to define a common protocol for the evaluation of the metal contamination of seawater, sediment and biota. Contaminants have been chosen following preliminary consultations among countries of the ADRION area, considering objectives of WFD and MSFD, as well as Environmental Impact Assessment (EIA) procedures for offshore platforms. Information was gathered relative to matrix characteristics and quality assurance/quality control of the analytical performance (sample preservation, analytical methodology, reference materials, limit of detection, and limit of quantification, accuracy, reproducibility, etc.). The comparison of information provided by laboratories of nine institutions highlighted the request for harmonization in terms of sampling procedures, matrix characterization, preservation procedures, analytical methods and LOQ values. Although appropriate environmental quality standards for biota and sediment matrices should be established at national level and also through regional and sub-regional cooperation, as required by the WFD and MSFD, the proposed LOQ values, even if challenging, represent a benchmark and a stimulus to optimize analytical performance, to ensure the best level of protection to the coastal and offshore environment in the ADRION Region.

Challenges in harmonized environmental impact assessment (EIA), monitoring and decommissioning procedures of offshore platforms in Adriatic-Ionian (ADRION) region

Povzetek: A harmonized and integrated approach for monitoring and assessment of contamination, including hydrocarbon exploitation one, is required both by Marine Strategy Framework Directive (MSFD) at EU level and by the Ecosystem Approach (EcAp) program of the Barcelona Convention at Mediterranean level. A broad review of protocols of environmental impact assessment (EIA) procedures, monitoring and decommissioning of offshore platforms adopted by EU and non-EU countries along the Adriatic-Ionian seas was carried out in the framework of the Interreg offshore platforms in Adriatic-Ionian (ADRION) project HarmoNIA (Harmonization and networking for contaminant assessment in the Ionian and Adriatic Seas). The comparison of information provided by six ADRION countries and the application of a harmonized and integrated approach has highlighted specific challenges for managing offshore platform impacts emerged at ADRION level: (i) need of the same legislative level (the Offshore Protocol of Barcelona Convention is not ratified by all countries); (ii) set up of a task force of ADRION experts for discussing critical issues related to impacts of offshore platforms; (iii) harmonization, at the regional level, of EIA procedures, monitoring and decommissioning; (iv) need of an agreed and common list of recommended parameters to monitor in water, sediment and biota for the assessment of impacts due to platform installations and PFW discharges.

Highly specific qPCR and amplicon sequencing method for detection of quarantine citrus pathogen Phyllosticta citricarpaapplicable for air samples

Povzetek: The fungus Phyllosticta citricarpa is a quarantine pathogen in the EU and is of high economic importance in many parts of the world where favourable climate conditions drive the development of citrus black spot (CBS) disease. Disease symptoms include necrotic lesions on leaves and fruits. Low disease pressure can reduce crop market-ability, while higher disease pressure can cause premature fruit drop, significantly increasing crop losses. The wind-dispersed spores of P. citricarpa are especially prob-lematic for rapid pathogen dispersal, but also provide an opportunity for early detec-tion of the disease spreading into a new area. In this study we have developed and validated a quantitative PCR (qPCR) assay based on the TEF1-α sequence. Specificity testing demonstrated that it is currently the only qPCR assay that does not cross- react with closely related Phyllosticta species. The assay is sensitive and can detect a single copy of the TEF1 gene in a reaction, it is highly repeatable and reproducible and can be used for testing of the sticky tapes from spore traps as well as citrus fruit sam-ples. High-throughput sequencing (HTS) of the DNA barcodes ITS1 and TEF1 was also explored for the detection and discrimination of P. citricarpa. The limit of detection of the HTS was 1000 spores on a daily spore trap tape. This study makes an important improvement to the diagnostics of the CBS and the methods developed can also be applied to improve the surveillance and early detection of the pathogen when linked to spore samplers in the field.

Multivariate statistical methods in determining the spatial distribution of chemical elements in soil from the Mavrovo-Rostuše region, North Macedonia

Povzetek: In this work, the contents and spatial distributions of 19 elements (Ag, Al, B, Ba, Ca, Cr, Cu, Fe, K, Li, Mg, Mn, Na, Ni, P, Pb, Sr, V, Zn) in the soil of the western part of North Macedonia (Mavrovo-Rostuše region) are presented. For this purpose, a total of 66 soil samples were collected from 33 locations (33 samples of topsoil, 0-5 cm, and 33 samples of subsoil, 20-30 cm). All samples were analysed by inductively coupled plasma - atomic emission spectrometry (ICP-AES) after complete digestion with four acids (HNO3, HF, HClO4 and HCl). The obtained results were statistically evaluated and spatial distribution maps for all analysed elements were also prepared. Factor analysis was performed to reduce the number of data used and new synthetic variables (factors) were identified. Through the application of factor analysis, three geochemical associations were identified: Factor 1 (Zn, K, Cu, Fe, and Li), Factor 2 (Cr, Ni, and Mg) and Factor 3 (Ca and Al). From the obtained data and the maps of spatial distribution, it could be concluded that the distribution of the analysed elements is related to the lithology of the region. Thus, it was found that the higher content of elements of Factor 1 occurs in the eastern and southern part of the study area (middle and lower reaches of the Radika River and along the Mala River), where Mesozoic and Paleozoic carbonates as well as Paleozoic shales and Paleogene flysch prevail. Factor 2 (Cr, Ni and Mg) also represents a lithogenic association. The highest contents of the elements in both soil layers were found in the areas where Paleozoic sandstones and shales (village of Lazaropole and the area around the Mavrovo Lake) and Paleogene flysch (Rostuše village) predominate. Factor 3 (Ca and Al) also represents lithogenic association of elements. The highest content of these elements was found in the northwestern part of the study area (village of Žirovnica and along the Berička River) and in the northeastern region above the village of Brodec where Mesozoic carbonates and Paleogene flysch dominate.

Teaching and learning the language of the neighbour country : tools for mainstream primary education in the Slovenian-Italian border area

Povzetek: The article presents two central tools for teaching neighbour languages in border areas within the didactic framework of the Contact-Based-(Neighbour-)Language-Learning-andTeaching method (CoBLaLT, Cavaion 2015; 2016; 2019), namely a teacher’s guide to contact-based neighbour language teaching and the digital portfolio ‘My multimedia autobiography of crossborder contacts’ for pupils aged 11 to 15. They were developed as part of a postdoctoral project between 2017-2019 in the Italian-Slovenian border region. The article highlights the importance of making language teaching research a collaborative process between a strong scientific and professional community that thus fosters innovation and effectiveness of methods and contents in neighbour language learning.

PCR primers comparisons for a successful Tuber spp. DNA region amplification in routine identifications

Povzetek: Since late 20th century DNA sequencing became the method of choice method in precision species identification. The ITS region is one of the official fungal barcoding DNA markers, although in some cases sequencing of the ITS re-gion may, due to misidentification, mislabeling or nomen-clature errors in public databases, lead to incorrect or insuf-ficient identification, as is currently a case in the genus Tu b e r. The aim of this study was to test, which ITS primer pairs are most appropriate and optimal for Tu b e r species DNA region amplification. Thereby we (1) compared ampli-fication success for different Tu b e r species using fungal spe-cific primer pair ITS1f and ITS4 and (2) compared amplifi-cation success using different ITS primer pair combinations in amplifying DNA region an example species Tuber aesti-vum. Based on results, Tuber aestivum was one of the most reluctant Tu b e r species in this study and in most cases failed to amplify with the above primer pair. After comparing dif-ferent ITS primer pairs, we conclude that the primer pair ITS5 and ITS7 is the most appropriate primer pair for ampli-fication DNA region of T. ae stiv um as it resulted in high am-plification success from ectomycorrhizal root tips. Based on sequences, gained from public databases, we found that ITS1f and ITS6 primers have a mismatch in one base pair compared to the target sequence of Tuber aestivum, thus re-sulting in poor or no amplification success. Although prim-er pair ITS5 and ITS7 in our study was proven to be the most appropriate primer pair in amplifying DNA region Tu b e r aestivum species, further analysis about appropriateness of it for a general barcoding and identification of ectomycorrhiza in complex community samples is needed.
