Stem cells of aquatic invertebrates as an advanced tool for assessing ecotoxicological impacts

Povzetek: Environmental stressors are assessed through methods that quantify their impacts on a wide range of metrics including species density, growth rates, reproduction, behaviour and physiology, as on host-pathogen interactions and immunocompetence. Environmental stress may induce additional sublethal effects, like mutations and epigenetic signatures affecting offspring via germline mediated transgenerational inheritance, shaping phenotypic plasticity, increasing disease susceptibility, tissue pathologies, changes in social behaviour and biological invasions. The growing diversity of pollutants released into aquatic environments requires the development of a reliable, standardised and 3R (replacement, reduction and refinement of animals in research) compliant in vitro toolbox. The tools have to be in line with REACH regulation 1907/2006/EC, aiming to improve strategies for potential ecotoxicological risks assessment and monitoring of chemicals threatening human health and aquatic environments. Aquatic invertebrates' adult stem cells (ASCs) are numerous and can be pluripotent, as illustrated by high regeneration ability documented in many of these taxa. This is of further importance as in many aquatic invertebrate taxa, ASCs are able to differentiate into germ cells. Here we propose that ASCs from key aquatic invertebrates may be harnessed for applicable and standardised new tests in ecotoxicology. As part of this approach, a battery of modern techniques and endpoints are proposed to be tested for their ability to correctly identify environmental stresses posed by emerging contaminants in aquatic environments. Consequently, we briefly describe the current status of the available toxicity testing and biota-based monitoring strategies in aquatic environmental ecotoxicology and highlight some of the associated open issues such as replicability, consistency and reliability in the outcomes, for understanding and assessing the impacts of various chemicals on organisms and on the entire aquatic environment. Following this, we describe the benefits of aquatic invertebrate ASC-based tools for better addressing ecotoxicological questions, along with the current obstacles and possible overhaul approaches.

Biomaterials and bioactive natural products from marine invertebrates

Povzetek: Aquatic invertebrates are a major source of biomaterials and bioactive natural products that can find applications as pharmaceutics, nutraceutics, cosmetics, antibiotics, antifouling products and biomaterials. Symbiotic microorganisms are often the real producers of many secondary metabolites initially isolated from marine invertebrates; however, a certain number of them are actually synthesized by the macro-organisms. In this review, we analysed the literature of the years 2010–2019 on natural products (bioactive molecules and biomaterials) from the main phyla of marine invertebrates explored so far, including sponges, cnidarians, molluscs, echinoderms and ascidians, and present relevant examples of natural products of interest to public and private stakeholders. We also describe omics tools that have been more relevant in identifying and understanding mechanisms and processes underlying the biosynthesis of secondary metabolites in marine invertebrates. Since there is increasing attention on finding new solutions for a sustainable large-scale supply of bioactive compounds, we propose that a possible improvement in the biodiscovery pipeline might also come from the study and utilization of aquatic invertebrate stem cells.

Povzetek: The “stem cells” discipline represents one of the most dynamic areas in biomedicine. While adult marine/aquatic invertebrate stem cell (MISC) biology is of prime research and medical interest, studies on stem cells from organisms outside the classical vertebrate (e.g., human, mouse, and zebrafish) and invertebrate (e.g., Drosophila, Caenorhabditis) models have not been pursued vigorously. Marine/aquatic invertebrates constitute the largest biodiversity and the widest phylogenetic radiation on Earth, from morphologically simple organisms (e.g., sponges, cnidarians), to the more complex mollusks, crustaceans, echinoderms, and protochordates. These organisms contain a kaleidoscope of MISC-types that allow the production of a large number of novel bioactive-molecules, many of which are of significant potential interest for human health. MISCs further participate in aging and regeneration phenomena, including whole-body regeneration. For years, the European MISC-community has been highly fragmented and has established scarce ties with biomedical industries in an attempt to harness MISCs for human welfare. Thus, it is important to (i) consolidate the European community of researchers working on MISCs; (ii) promote and coordinate European research on MISC biology; (iii) stimulate young researchers to embark on research in MISC-biology; (iv) develop, validate, and share novel MISC tools and methodologies; (v) establish the MISC discipline as a forefront interest of biomedical disciplines, including nanobiomedicine; and (vi) establish collaborations with industries to exploit MISCs as sources of bioactive molecules. In order to fill the recognized gaps, the EC-COST Action 16203 “MARISTEM” has recently been launched. At its initial stage, the consortium unites 26 scientists from EC countries, Cooperating countries, and Near Neighbor Countries.

Overview of global long-distance road Transportation of industrial roundwood

Povzetek: The aim of the study was to provide a comprehensive overview of global long-distance road transportation of industrial roundwood. The study focused on the maximum gross vehicle weight (GVW) limits allowed with different timber truck configurations, typical payloads in timber trucking, the road transportation share of the total industrial roundwood longdistance transportation volume, and the average long-distance transportation distances and costs of industrial roundwood. The study was carried out as a questionnaire survey. The questionnaire was sent to timber transportation logistics experts and research scientists in the 30 countries with the largest industrial roundwood removals in Europe, as well as selected major forestry countries in the world (Argentina, Australia, Brazil, Canada, Chile, China, Japan, New Zealand, South Africa, Türkiye, the United States of America and Uruguay) in February 2022, and closed in May 2022. A total of 31 countries took part in the survey. The survey illustrated that timber trucking was the main long-distance transportation method of industrial roundwood in almost every country surveyed. Road transportation averaged 89% of the total industrial roundwood long-distance transportation volume. Timber truck configurations of 4 to 9 axles with GVW limits of around 30 tonnes to over 70 tonnes were most commonly used. The results indicated that higher GVW limits allowed significantly higher payloads in timber trucking, with the lowest payloads at less than 25 tonnes, and the highest payloads more than 45 tonnes. The average road transportation distance with industrial roundwood was 128 km, and the average long-distance transportation cost in timber trucking was €11.1 per tonne of timber transported. In the entire survey material, there was a direct relationship between transportation distance and transportation costs and an inverse relationship between maximum GVW limits and transportation costs. Consequently, in order to reduce transportation costs, it is essential to maximise payloads (within legal limits) and minimise haul distances. Several measures to increase cost- and energy-efficiency, and to reduce greenhouse gas emissions in road transportation logistics, are discussed in the paper. On the basis of the survey, it is recommended that upto-date statistical data and novel research studies on the long-distance transportation of industrial roundwood be conducted in some countries in the future.

Diversity trapped in cages : revision of Blumenavia Möller (Clathraceae, Basidiomycota) reveals three hidden species

Povzetek: Basidiomata of Phallales have a diversified morphology with adhesive gleba that exudes an odor, usually unpleasant that attracts mainly insects, which disperse the basidiospores. The genus Blumenavia belongs to the family Clathraceae and, based on morphological features, only two species are currently recognized: B. rhacodes and B. angolensis. However, the morphological characters adopted in species delimitations within this genus are inconsistent, and molecular data are scarce. The present study aimed to review and identify informative characters that contribute to the delimitation of Blumenavia species. Exsiccates from America and Africa were analyzed morphologically, and molecularly, using ITS, LSU, ATP6, RPB2 and TEF-1% markers for Maximum Parsimony, Bayesian and Maximum likelihood analyses, and also for coalescent based species delimitations (BP&P), as well as for bPTP, PhyloMap, Topo-phylogenetic and Geophylogenetic reconstructions. According to our studies, seven species can be considered in the genus: B. rhacodes and B. angolensis are maintained, B. usambarensis and B. toribiotalpaensis are reassessed, and three new species are proposed, B. baturitensis Melanda, M.P. Martín & Baseia, sp. nov., B. crucis-hellenicae G. Coelho, Sulzbacher, Grebenc & Cortez, sp. nov., and B. heroica Melanda, Baseia & M.P. Martín, sp. nov. Blumenavia rhacodes is typified by selecting a lectotype and an epitype. Macromorphological characters considered informative to segregate and delimit the species through integrative taxonomy include length of the basidiomata, color, width and presence of grooves on each arm as well as the glebifer position and shape. These must be clearly observed while the basidiomata are still fresh. Since most materials are usually analyzed after dehydration and deposit in collections, field techniques and protocols to describe fugacious characters from fresh specimen are demanded, as well as the use of molecular analysis, in order to better assess recognition and delimitation of species in Blumenavia.

Brown rotting fungus closely related to Pseudomerulius curtisii (Boletales) recorded for the first time in South America

Povzetek: In the region of Santa Maria, Southern Brazil, we have analyzed morphologically and molecularly some interesting brown-rotting mushroom specimens closely related to Pseudomerulius curtisii. Except for minor differences in morphology and ITS sequence similarity, collections have corresponded to P. curtisii by basidiospore size and shape, the kind of hyphal system, the macromorphology, the slightly unpleasant pungent spicy smell turning stronger upon drying and, particularly, by the highly supported and closely related clade after phylogenetic analysis. Perhaps due the rarity in nature, morphological data are not abundant in literature and appears to be somewhat incomplete to discordant for the species, so we provide a more detailed description and illustrations from collected specimens.
