Metabolic consequences of infection of grapevine (Vitis vinifera L.) cv. ʺModra frankinjaʺ with flavescence dorée phytoplasma

Povzetek: Flavescence dorée, caused by the quarantine phytoplasma FDp, represents the most devastating of the grapevine yellows diseases in Europe. In an integrated study we have explored the FDp–grapevine interaction in infected grapevines of cv. “Modra frankinja” under natural conditions in the vineyard. In FDp-infected leaf vein-enriched tissues, the seasonal transcriptional profiles of 14 genes selected from various metabolic pathways showed an FDp-specific plant response compared to other grapevine yellows and uncovered a new association of the SWEET17a vacuolar transporter of fructose with pathogens. Non-targeted metabolome analysis from leaf vein-enriched tissues identified 22 significantly changed compounds with increased levels during infection. Several metabolites corroborated the gene expression study. Detailed investigation of the dynamics of carbohydrate metabolism revealed significant accumulation of sucrose and starch in the mesophyll of FDp-infected leaves, as well as significant up-regulation of genes involved in their biosynthesis. In addition, infected leaves had high activities of ADP-glucose pyrophosphorylase and, more significantly, sucrose synthase. The data support the conclusion that FDp infection inhibits phloem transport, resulting in accumulation of carbohydrates and secondary metabolites that provoke a source-sink transition and defense response status.

Makroekonomski učinki proizvodnje in predelave lesa listavcev

Povzetek: As far as the utilization of the value-added potential of hardwood assortments is concerned, we have considerable reserves in Slovenia. These could be exploited mainly by increasing the economic use of roundwood in Slovenia and by developing more technologically advanced processing methods for roundwood. The aim of this paper is to assess the untapped potential of the forest-wood chain in Slovenia using input-output models, focusing on the use of hardwood. For the whole forest-wood chain, we assessed the existing situation and tested five scenarios for the improvement of the forest-wood chain in Slovenia. We developed a national input-output model to evaluate possible scenarios for the future development of the wood-based bioeconomy. The results of scenario analysis of the input-output model show that the Slovenian wood-based bioeconomy can achieve up to 20% higher production value, a 24% increase in employment and 19% increase in household income in the future through restructuring of economic activities. In the paper, we discuss how upgrading the forestry and wood-processing industry (according to the defined scenarios) could be achieved by increasing the processing capacity of hardwoods and provide recommendations for decision makers.

Antibacterial properties and cytotoxicity of 100% waste derived alkali activated materials : slags and stone wool-based binders

Povzetek: In this study we compare the leaching behavior and the antibacterial and cytotoxic properties of 100% slag or stone wool derived alkali activated materials. The antibacterial activity was measured as the inhibiting capacity against two Gram- negative bacterial strains, Escherichia coli and Pseudomonas aeruginosa and one Gram-positive bacterial strain: Enterococcus faecalis. The cytotoxicity properties were tested on mouse embryonic fibroblast NIH-3T3 cell-line. It was proved that the high quality of the 3D aluminosilicate network of the consolidated materials obtained from powders of CaO or MgO-rich slags or stone wool, opportunely activated with NaO and/or Na-silicate, was capable of stabilizing heavy metal cations. The concentrations of leachate heavy cations were lower than the European law limit when tested in water. The effect of additives in the composites, basal fibers or nanocellulose, did not reduce the chemical stability and slightly influenced the compressive strength. Weight loss in water increased by 20% with basalt fibers addition, while it remained almost constant when nanocellulose was added. All the consolidated materials, cement-like in appearance, exhibited limited antibacterial properties (viability from 50 to 80% depending on the bacterial colony and the amount of sample) and absence of cytotoxicity, envisaging good acceptance from part of the final consumer and zero ecological impact. CaO-rich formulations can replace ordinary Portland cement (showing bacterial viability at 100%) with a certain capability for preventing the reproduction of the E. coli and S. aureus bacteria with health and environmental protection results.
