Exploring module interactions in modular CMA-ES across problem classes

Abstract: This study presents an in-depth analysis of module importance within the modular CMA-ES (modCMA-ES) algorithm using exploratory data analysis and large-scale benchmarking across the BBOB suite. Rather than introducing new algorithms, our contribution lies in uncovering how individual modules and their interactions influence optimization performance across diverse black-box problem classes. We evaluate 324 modCMA-ES variants across 24 problem classes using functional ANOVA (f-ANOVA) to quantify the variance in performance attributable to individual, pairwise, and triplet module interactions. Results reveal substantial variation in module importance across problem classes and highlight strong alignment between module interaction patterns and high-level landscape features, particularly multi-modality. Further, we demonstrate that configuring only the most important modules — identified via f-ANOVA — achieves performance comparable to or better than the single-best solver, especially in high-dimensional settings. This analysis, conducted at both low (5D) and high (30D) dimensions, offers actionable insights into module interactions within the mod-CMA-ES framework.

Thiol-reactive or redox-active : revising a repurposing screen led to a new invalidation pipeline and identified a true noncovalent inhibitor against papain-like protease from SARS-CoV-2

Abstract: The SARS-CoV-2 papain-like protease PLpro has multiple roles in the viral replication cycle, related to both its polypeptide cleavage function and its ability to antagonize the host immune response. Targeting the PLpro function is recognized as a promising mechanism to modulate viral replication, while supporting host immune responses. However, the development of PLpro-specific inhibitors remains challenging. Comprehensive investigations utilizing enzymatic, binding studies, and cellular assays revealed the previously reported inhibitors to act in an unspecific manner. At present, GRL-0617 and its derivatives remain the best-validated compounds with demonstrated antiviral activity in cells and in mouse models. In this study, we refer to the pitfalls of the redox sensitivity of PLpro. Using a screening-based approach to identify inhibitors of PLpro’s proteolytic activity, we made extensive efforts to validate active compounds over a range of conditions and readouts, emphasizing the need for comprehensive orthogonal data when profiling putative PLpro inhibitors. The remaining active compound, CPI-169, was shown to be a noncovalent inhibitor capable of competing with GRL-0617 in NMR-based experiments, suggesting that it occupied a similar binding site and inhibited viral replication in Vero-E6 cells, opening new design opportunities for further development as antiviral agents.

The electronic structure of 1/1 ZnMgHf and its consequences for the electronic transport of the quasicrystal

Abstract: This paper presents a detailed density functional theory study of the 1/1 periodic approximant Zn-Mg-Hf crystal structure, which is a close analog of the F-type Bergman-type quasicrystal. Two structural models, Bergman-like and Tsai-like, are examined to investigate their energetic stability, electronic structure, and transport properties. The Tsai-like model, characterized by additional multicenter bonds, higher Madelung stability and significant charge transfer, is found to be energetically more favorable by 10.9 meV per unit cell. Fermi surface nesting features may be related to quasicrystal stabilization mechanisms. Study utilizes Electron Localization Function, non-covalent bond analysis and Maximally Localized Wannier Functions. Experimental resistivity and magnetoresistance measurements confirm metallic behavior with nontrivial scattering mechanisms and subquadratic magnetoresistance. This study emphasizes the combined role of electronic sp-d hybridization, charge transfer, and multicenter bonds in the stabilization of Zn-based periodic approximant crystals. The present findings can be extended to the quasicrystalline phase.

Pomen tranzicijske pravičnosti za spoštovanje človekovega dostojanstva v povojnih razmerah

Abstract: Članek se osredotoča na pomen tranzicijske pravičnosti in narodne sprave za stabilizacijo demokracije v postavtoritarnih in postkonfliktnih družbah, pri čemer ima ključno vlogo uresničevanje človekovega dostojanstva. Članek poudarja, da stabilizacija demokracije zahteva odgovoren odnos do preteklosti, predvsem skozi prepoznavanje in popravo zlorab nedemokratičnih režimov. Tranzicijska pravičnost se pojavlja kot ključni mehanizem, ki ne zagotavlja zgolj pravnih sredstev za uveljavljanje odgovornosti, temveč tudi prispeva k popravi in obnovi družbenega tkiva, kar krepi realizirano dostojanstvo posameznikov, zlasti žrtev. Avtorja ugotavljata, da je uspeh tranzicijske pravičnosti pogosto omejen zaradi ideološke polarizacije, hegemonije elit in politizacije procesa, kar lahko ovira doseganje prave narodne sprave in stabilne demokracije.

Mariborska škofija in (de)nacionalizacija cerkvenega premoženja

Abstract: Avtor na podlagi preučevanja arhivskih virov in literature obravnava izvedbo nacionalizacije po drugi svetovni vojni, s katero je bilo komunistični oblasti omogočeno, da je uničila še tisto zasebno lastnino Cerkve, ki je takratna oblast ni uspela odvzeti z agrarno reformo. Tako je bila do leta 1960 približno polovica vseh nacionaliziranih verskih objektov v Sloveniji v tedanji mariborski škofiji. Ta odstotek nacionaliziranih cerkvenih objektov je vsekakor predstavljal hud udarec omenjeni škofiji tako na pastoralnem, izobraževalnem in nenazadnje tudi na ekonomskem področju. V drugem delu članka se avtor osredotoča na analizo postopka denacionalizacije premoženja, ki je bilo v preteklosti odvzeto Mariborski nadškofiji. Obravnava predvsem proces povračila tega premoženja na primeru bogoslovnega semenišča na Vrbanski 30 v Mariboru, ki je bilo v celoti vrnjeno v last nadškofije, s čimer je bila obnovljena pravna in gospodarska podlaga te cerkvene institucije. Pri tem izpostavi tudi zgodovinski kontekst ter pravne in družbene implikacije tega pomembnega koraka.
