Title:Limfoblastoidne celične linije kot model in vitro za predklinično vrednotenje zdravilnih učinkovin

Abstract:Personalizirana medicina temelji na prilagajanju zdravljenja genotipu in fenotipu posameznika z namenom povečanja učinkovitosti terapije in zmanjšanja tveganja za neželene učinke. Pomembno vlogo pri njenem uveljavljanju imajo ustrezni predklinični modeli in vitro, ki omogočajo sistematično proučevanje interindividualnih razlik v odzivu na zdravilne učinkovine. Med pogosto uporabljenimi modeli so limfoblastoidne celične linije, pridobljene s transdukcijo človeških limfocitov B z virusom Epstein-Barr. Limfoblastoidne celične linije ohranjajo genetske značilnosti darovalca in predstavljajo uporaben model in vitro v genomskih, farmakogenomskih ter farmakoloških raziskavah. V članku so predstavljeni postopki priprave limfoblastoidnih celičnih linij, molekularni mehanizmi njihove imortalizacije ter njihova uporaba v predkliničnih študijah in personalizirani medicini. Obravnavane so tudi prednosti in omejitve uporabe limfoblastoidnih celičnih linij v predkliničnih študijah, zlasti vpliv transdukcije z virusom Epstein-Barr na celično fiziologijo, odzivnost na zdravila ter interpretacijo eksperimentalnih rezultatov. Limfoblastoidne celične linije predstavljajo pomemben podporni model in vitro, predvsem v zgodnjih fazah predkliničnih študij in kot dopolnilo naprednejšim celičnim sistemom v sodobni biomedicini.

Title:Lymphoblastoid cell lines as an in vitro model for preclinical drug evaluation
Abstract:Personalised medicine involves tailoring treatment to an individual’s genotype and phenotype, to increase therapeutic efficacy and reduce the risk of adverse effects. Its successful implementation depends on appropriate in vitro preclinical models that allow systematic investigation of interindividual differences in drug response. Among commonly used models are lymphoblastoid cell lines, generated by transforming of human B lymphocytes with Epstein–Barr virus. Lymphoblastoid cell lines retain the genetic characteristics of the donor and serve as a useful in vitro model in genomic, pharmacogenomic, and pharmacological research. This article presents methods for establishing of lymphoblastoid cell lines, the molecular mechanisms underlying their immortalisation, and their use in preclinical research and personalised medicine. The advantages and limitations of this model are also discussed, particularly the impact of Epstein–Barr virus transformation on cellular physiology and drug responsiveness. Lymphoblastoid cell lines are an important supportive in vitro model, especially in early research stages and as a complement to more advanced cellular systems.
