Nature-inspired substituted 3-(imidazol-2-yl) morpholines targeting human topoisomerase IIα : dynophore-derived discovery

Abstract: The molecular nanomachine, human DNA topoisomerase IIα, plays a crucial role in replication, transcription, and recombination by catalyzing topological changes in the DNA, rendering it an optimal target for cancer chemotherapy. Current clinical topoisomerase II poisons often cause secondary tumors as side effects due to the accumulation of double-strand breaks in the DNA, spurring the development of catalytic inhibitors. Here, we used a dynamic pharmacophore approach to develop catalytic inhibitors targeting the ATP binding site of human DNA topoisomerase IIα. Our screening of a library of nature-inspired compounds led to the discovery of a class of 3-(imidazol-2-yl) morpholines as potent catalytic inhibitors that bind to the ATPase domain. Further experimental and computational studies identified hit compound 17, which exhibited selectivity against the human DNA topoisomerase IIα versus human protein kinases, cytotoxicity against several human cancer cells, and did not induce DNA double-strand breaks, making it distinct from clinical topoisomerase II poisons. This study integrates an innovative natural product-inspired chemistry and successful implementation of a molecular design strategy that incorporates a dynamic component of ligand-target molecular recognition, with comprehensive experimental characterization leading to hit compounds with potential impact on the development of more efficient chemotherapies.

Incidenca raka v prvem letu epidemije covida-19

Abstract: Uvod: Število novih diagnoz raka (incidenca) se v Sloveniji v zadnjem desetletju povečuje za povprečno 1,6 % na leto. V letu 2020, prvem letu omejitvenih ukrepov zaradi epidemije covida19, smo s platformo OnKOvid predvideli 3–8-odstotni padec novih diagnoz raka. Namen je predstaviti uradno incidenco raka za Slovenijo za leto 2020 in ugotoviti, kolikšen je bil dejansko manko novih diagnoz raka. Metode: V Registru raka Republike Slovenije smo v skladu z mednarodnimi pravili registrirali vse nove primere raka pri prebivalcih s stalnim prebivališčem v Sloveniji. Primerjali smo jih z registrirano incidenco za 2019, modelno incidenco za 2020, preliminarnimi rezultati OnKovida in dostopnimi rezultati incidence iz registrov raka iz drugih držav. Rezultati: V letu 2020 je za rakom zbolelo 15.096 oseb v Sloveniji (7.034 žensk, 8.002 moška). Glede na modelno oceno incidence za leto 2020 gre za manko 1.854 oseb (10,9 %; 11,6 % žensk, 10,4 % moških). Pri tem je za 10,5 % manj ugotovljenih rakov v omejenem stadiju, najbolj izrazito v starosti 50–69 let (za 13,2 %). Upad je primerljiv s poročili iz Anglije, ZDA in Kanade ter deloma večji kot na Škotskem in Švedskem. Najbolj izrazit upad novih diagnoz raka ugotavljamo pri nemelanomskem kožnem raku (23 %, najbolj po 50. letu), raku prostate (15,9 %, najbolj v omejenem stadiju), pljučnem raku (8,9 %, 60–64 let, v razširjenem stadiju), raku dojk (8,3 %, 45–64 let), ne-Hodgkinovem limfomu (9 %) in levkemijah (11,6 %). Pri kožnem melanomu, raku debelega črevesa in danke ni bilo primanjkljaja. Zaključek: Upad incidence raka v letu 2020 gre verjetno na račun omejitvenih ukrepov, reorganizacije zdravstva in z zdravjem povezanega vedenja med epidemijo covida-19, beležijo jo v več državah. Med epidemijami je potrebno nemoteno izvajanje onkologije.

Impact assessment of the Gajke and Brstje landfills on groundwater status using stable and radioactive isotopes

Abstract: Waste disposal in landfills represents a severe threat to aquatic environments on the local, regional, and global levels. In Slovenia, there are 69 registered landfills where groundwater is regularly monitored. However, isotope techniques are not regularly employed. Therefore, we employed isotope analysis of hydrogen, carbon, and oxygen in combination with total alkalinity to assess the impact of the selected landfill on groundwater and to evaluate the biogeochemical processes at work. The δ18O, δ2H, δ13CDIC, 3H activity and total alkalinity were determined in October 2020 at 12 sampling points from the surrounding area of the Gajke and Brstje landfills and leachate from the Gajke landfill. The δ18O (-9.24 ± 0.3 ‰) and δ2H (-64.9 ± 2.7 ‰) in groundwater indicate that the main water source consists in direct infiltration of precipitation, with no significant isotopic fractionation. Total alkalinity in the investigated area ranges from 5.45 to 73 mM and δ13CDIC from –14.9 to +6.1 ‰, respectively. Higher values of total alkalinity (up to 73 mM), δ13CDIC (up to +6.1 ‰), δ18O (-7.64 ‰) and 3H (209.8 TU) are detected in the leachate, indicating biogeochemical process related to CO2 reduction or methanogenesis. Methanogenesis could be present at locations GAP-10/13 (Brstje landfill) and G-2 (Gajke landfill) with δ13CDIC values ranging from –8.2 to –7.6 ‰ and with dissolved oxygen values around 0 % and elevated 3H values (from 16 to 18 TU). This study demonstrates the effectiveness of isotopic analysis as a valuable tool for monitoring landfills, revealing shifts in biogeochemical processes within the groundwater there.
