Chloroplast redox state changes mark cell-to-cell signaling in the hypersensitive response

Povzetek: Bisphenol A (BPA) is one of the most commonly used substances in the manufacture ofvarious everyday products. Growing concerns about its hazardous properties, including endocrinedisruption and genotoxicity, have led to its gradual replacement by presumably safer analogues inmanufacturing plastics. The widespread use of BPA and, more recently, its analogues has increasedtheir residues in the environment. However, our knowledge of their toxicological profiles is limitedand their combined effects are unknown. In the present study, we investigated the toxic effectscaused by single bisphenols and by the combined exposure of BPA and its two analogues, BPAP andBPC, after short (24-h) and prolonged (96-h) exposure in HepG2 spheroids. The results showed thatBPA did not reduce cell viability in HepG2 spheroids after 24-h exposure. In contrast, BPAP andBPC affected cell viability in HepG2 spheroids. Both binary mixtures (BPA/BPAP and BPA/BPC)decreased cell viability in a dose-dependent manner, but the significant difference was only observedfor the combination of BPA/BPC (both at 40μM). After 96-h exposure, none of the BPs studiedaffected cell viability in HepG2 spheroids. Only the combination of BPA/BPAP decreased cellviability in a dose-dependent manner that was significant for the combination of 4μM BPA and 4μMBPAP. None of the BPs and their binary mixtures studied affected the surface area and growth ofspheroids as measured by planimetry. In addition, all BPs and their binary mixtures studied triggeredoxidative stress, as measured by the production of reactive oxygen species and malondialdehyde,at both exposure times. Overall, the results suggest that it is important to study the effects of BPsas single compounds. It is even more important to study the effects of combined exposures, as thecombined effects may differ from those induced by single compounds.

The perception of minorities toward the Turkish state : ǂthe ǂcase of ethno-religious communities

Povzetek: The relationship between state and non-Muslim communities has been a de-licate issue since the founding of the Turkish Republic despite the principle of secularism stated in its constitution. Against this background, the association of national identity with Sunni-Islam has been the main marker of inclusion/exclusi-on for national identity. Especially since 2002 when the Justice and Development Party (JDP) came to power, the debate with regard to freedom of religion and the rights of religious minorities came to the fore. Over the course of decades there have been numerous studies approaching the state’s perspective towards religious minorities. However, there is a paucity of academic studies that focuses on citizen-ship experiences of the members of these communities through the course of their daily and social lives. In this article, we first provide a historical perspective of the state towards religious minorities from the establishment of the Republic until to-day, including the JDP period. In the second part of this study we aim to explore recasting perspectives of the non-Muslim minorities over the previous decade by taking the standpoint of the members of Greek Orthodox, Jews and Armenian communities. To this end, we conduct in-depth interviews with the members of these communities who reside in Istanbul. Finally, new negotiation fields which have been flourishing among these communities will be addressed.

The crumbling touchstone of the Vatican's ostpolitik : relations between the Holy see and Yugoslavia, 1970-1989

Povzetek: Based on Yugoslav archival sources, the paper analyses the relationship between the Holy See and Yugoslavia as the only Eastern European socialist state with which the former had official diplomatic relations. The relationship between the Holy See and socialist Yugoslavia provides insight into the precarious position of the Catholic Church in Yugoslavia, but also into the issues and dilemmas that the Holy See faced in terms of its Ostpolitik towards Eastern European socialist regimes. The article initially centres on the parallels between the Holy See and the foreign policy agenda of non-aligned Yugoslavia, especially during the papacy of Paul VI, when both actors shared an understanding of the acute problems of the Global South. It then analyses the stance of the Holy See towards the Yugoslav domestic policy of self-management that claimed that this system could present a viable environment for the life of believers in modern socialist society. The analysis closes with an in-depth examination of the role of the Holy See in the process of convergence between religion and nationalism during the 1980s, in which the Vatican did not play as clear-cut a role as it has generally been ascribed and proved unable to tackle this formidable phenomenon.

Comparative study of interactions of aliskiren and AT [sub] 1 receptor antagonists with lipid bilayers

Povzetek: The renin-angiotensin-aldosterone system (RAAS) plays a key role in the regulation of blood pressure. Renin is the rate limiting enzyme of the RAAS and aliskiren is a highly potent and selective inhibitor of the human renin. Renin is known to be active both in the circulating blood stream as well as locally, when bound to the (pro)-renin receptor ((P)RR). In this study we have investigated a possible mechanism of action of aliskiren, in which its accumulation in the plasma membrane is considered as an essential step for effective inhibition. Aliskiren's interactions with model membranes (cholesterol rich and poor) have been investigated by applying different complementary techniques: differential scanning calorimetry (DSC), Raman spectroscopy, magic angle spinning (MAS) nuclear magnetic resonance (NMR) spectroscopy and small- and wide-angle X-ray scattering (SAXS and WAXS). In addition, in silico molecular dynamics (MD) calculations were applied for further confirmation of the experimental data. Aliskiren's thermal effects on the pre- and main transition of dipalmitoyl-phosphatidylcholine (DPPC) membranes as well as its topographical position in the bilayer show striking similarities to those of angiotens.in II type 1 receptor (AT1R) antagonists. Moreover, at higher cholesterol concentrations aliskiren gets expelled from the membrane just as it has been recently demonstrated for the angiotensin receptor blocker (ARB) losartan. Thus, we propose that both the AT1R and the (P)RR-bound renin active sites can be efficiently blocked by membrane-bound ARBs and aliskiren when cholesterol rich membrane rafts/caveolae are formed in the vicinity of the receptors.
