Characterization of hFOB 1.19 cell line for studying Zn-based degradable metallic biomaterials

Povzetek: In vitro testing is the first important step in the development of new biomaterials. The human fetal osteoblast cell line hFOB 1.19 is a very promising cell model; however, there are vast discrepancies in cultivation protocols, especially in the cultivation temperature and the presence of the selection reagent, geneticin (G418). We intended to use hFOB 1.19 for the testing of Zn-based degradable metallic materials. However, the sensitivity of hFOB 1.19 to zinc ions has not yet been studied. Therefore, we compared the toxicity of zinc towards hFOB 1.19 under different conditions and compared it with that of the L929 mouse fibroblast cell line. We also tested the cytotoxicity of three types of Zn-based biomaterials in two types of media. The presence of G418 used as a selection reagent decreased the sensitivity of hFOB 1.19 to Zn2+. hFOB 1.19 cell line was more sensitive to Zn2+ at elevated (restrictive) temperatures. hFOB 1.19 cell line was less sensitive to Zn2+ than L929 cell line (both as ZnCl2 and extracts of alloys). Therefore, the appropriate cultivation conditions of hFOB 1.19 during biomaterial testing should be chosen with caution.

Protibolečinsko obsevanje kostnih zasevkov

Povzetek: Kostni zasevki povzročajo bolečino, ki vpliva na kakovost življenja bolnika in lahko vodijo do zapletov, ki zahtevajo urgentno ukrepanje. Ko zaradi kostnih zasevkov grozi oziroma je prisoten patološki zlom ali maligna kompresija hrbtenjače, je nujno oceniti, ali bolnik potrebuje kirurško intervencijo, ki ji sledi pooperativno obsevanje. Kadar bolnik ni kandidat za operativno zdravljenje, se priporoča urgentno obsevanje. Pri bolečih kostnih zasevkih, brez grozečega oziroma prisotnega patološkega zloma ali maligne utesnitve hrbtenjače, ne glede na velikost samega zasevka, prvi pristop zdravljenja bolečine predstavlja farmakološko zdravljenje, ki ga podpre in dopolni protibolečinsko paliativno obsevanje. Pri paliativnem obsevanju so celokupne obsevalne doze nizke, obsevanje je izvedeno v enem odmerku ali v kratkih obsevalnih režimih. Bolniki, ki ne odgovorijo na prvo obsevanje, ali se jim bolečina ponovi, so lahko varno ponovno obsevani na istem mestu. Na Onkološkem inštitutu Ljubljana smo opravili retrospektivni pregled paliativnih obsevanj kosti med leti 2018 – 2021. Letno obsevamo 800 kostnih zasevkov, upažamo pa upad števila obsevanj. Najpogostejša frakcionacija je 5 x 4 Gy, samo 10% bolnikov pa prejme obsevanje v enem odmerku. Protibolečinsko obsevanje kosti je učinkovito, stroškovno učinkovito in varno. Pomembno je prepoznati bolnike, ki bi imeli korist od protibolečinskega obsevanja, in jih napotiti na konzilij. S poenostavitvijo radioterapevtskih postopkov in odločitvijo za obsevanje v enem odmerku, je lahko obsevalno zdravljenje bolniku prijaznejše in dostopnejše.

Strong edge geodetic problem on complete multipartite graphs and some extremal graphs for the problem

Povzetek: A set of vertices $X$ of a graph $G$ is a strong edge geodetic set if to any pair of vertices from $X$ we can assign one (or zero) shortest path between them such that every edge of $G$ is contained in at least one on these paths. The cardinality of a smallest strong edge geodetic set of $G$ is the strong edge geodetic number ${\rm sg_e}(G)$ of $G$. In this paper, the strong edge geodetic number of complete multipartite graphs is determined. Graphs $G$ with ${\rm sg_e}(G) = n(G)$ are characterized and ${\rm sg_e}$ is determined for Cartesian products $P_n\,\square\, K_m$. The latter result in particular corrects an error from the literature.

Efficacy of trapping protocols for Agrilus jewel beetles : a multi-country assessment

Povzetek: The genus Agrilus is one of the most diverse insect genera worldwide. The larval feeding activity causes extensive damage in both forests and orchards. In addition, more than 30 species have been introduced outside their native range so far, including the emerald ash borer Agrilus planipennis Fairmaire. Thus, the availability of efficient trapping protocols for early detection of Agrilus species at entry points is of utmost importance. In this study we tested whether trapping protocols developed for surveillance of A. planipennis in North America were also effective for other Agrilus species. In particular, through a multi-country assessment we compared the efficacy of detecting Agrilus species on: (i) green glue-coated prism traps vs. green Fluon-coated multi-funnel traps when baited with the green leaf volatile (Z)-3-hexenol or left unbaited; and (ii) green multi-panel traps vs. green multi-panel traps baited with dead adult Agrilus beetles (decoys). A total of 23,481 individuals from 45 Agrilus species were caught. Trap design significantly affected both species richness and abundance of Agrilus species in several of the countries where the trapping experiments were carried out, and green prism traps outperformed green multi-funnel traps in most cases. On the contrary, the addition of a (Z)-3-hexenol lure or dead adult beetle decoys on to traps did not improve trap catches. Our study highlights that reliable trap models to survey Agrilus species are already available, but also that there is the clear need to further investigate chemical ecology of Agrilus species to develop semiochemical lures that can improve detection efficacy.

First record of the northern spruce bark beetle, Ips duplicatus (Sahlberg, 1836), in Slovenia

Povzetek: The northern spruce bark beetle, Ips duplicatus (Sahlberg), has invaded Central Europe in the past century and continues to spread southwards and westwards. It has caused damage in some parts of Europe in recent decades and poses a serious threat to spruce forests across the continent. From 2018 to 2021, we conducted intensive monitoring to determine the status of I. duplicatus in Slovenia. We used different types of traps and pheromone lures that attract I. duplicatus beetles and took bark samples from bark beetle-infested trees. Of the 26 traps used during the four years of monitoring, I. duplicatus was found in 16 traps in the central part of Slovenia in 2020. Adult beetles were caught in traps containing both I. typographus and I. duplicatus lures. The identity of this species was confirmed using morphological keys and molecular methods. One to five I. duplicatus beetles were found per trap, totaling 25 specimens. However, we did not confirm any I. duplicatus in samples taken from trees in 2021. We conclude that I. duplicatus is present in Slovenia, but our results suggest that the species is only present locally and that the population is small.
