Preliminary experiments into colonization of microorganisms from activated sludge on different types of plastics

Povzetek: The presence of plastics in the environment is currently one of the most pressing global environmental problems. Microorganisms start to form biofilms on plastic surfaces when they first come in contact with the biosphere; however, these interactions and processes are little understood, especially in freshwaters. This study aimed to better understand the colonization process of microorganisms from activated sludge on plastic materials exhibiting different surface characteristics. We inoculated synthetic fabric (PET), water bottles (PET), and plastic bags for packing vegetables and fruits (HDPE) with microorganisms from activated sludge. Mixtures of plastics and activated sludge, as well as the control, were incubated at 22-24°C in Bushnell Haas (BH) liquid medium and shaken at 120 rpm for two months. The mixtures were sub-sampled weekly and seeded into fresh BH medium with test plastic materials to avoid feeding microorganisms on dead biomass. The colonization was followed by measuring optical density (OD600) of liquid medium, by measurements of isotopic composition of carbon (δ13C) in untreated and treated plastic materials and, with in-specting the plastics surface with scanning electron microscopy (SEM). Overall, the study confirmed differences between colonizing microorganisms on different plastic material when comparing SEM micrographs of materials from the flasks inoculated with activated sludge. The texture of the HDPE bag changed during the experiment in both, control and inoculated flasks, but it is not clear whether the observed changes were due to abiotic or biotic factors. We concluded that microorganisms from activated sludge are capable of colonizing both PET and HDPE materials, and biofilm formation is most probably influenced by the chemical composition of plastics and their surface characteristics.

Stress-tolerant yeasts : opportunistic pathogenicity versus biocontrol potential

Povzetek: Stress-tolerant fungi that can thrive under various environmental extremes are highly desirable for their application to biological control, as an alternative to chemicals for pest management. However, in fungi, the mechanisms of stress tolerance might also have roles in mammal opportunism. We tested five species with high biocontrol potential in agriculture (Aureobasidium pullulans, Debayomyces hansenii, Meyerozyma guilliermondii, Metschnikowia fructicola, Rhodotorula mucilaginosa) and two species recognized as emerging opportunistic human pathogens (Exophiala dermatitidis, Aureobasidium melanogenum) for growth under oligotrophic conditions and at 37 °C, and for tolerance to oxidative stress, formation of biofilms, production of hydrolytic enzymes and siderophores, and use of hydrocarbons as sole carbon source. The results show large overlap between traits desirable for biocontrol and traits linked to opportunism (growth under oligotrophic conditions, production of siderophores, high oxidative stress tolerance, and specific enzyme activities). Based on existing knowledge and these data, we suggest that oligotrophism and thermotolerance together with siderophore production at 37 °C, urease activity, melanization, and biofilm production are the main traits that increase the potential for fungi to cause opportunistic infections in mammals. These traits should be carefully considered when assessing safety of potential biocontrol agents.

Polysaccharide-based bilayer coatings for biofilm-inhibiting surfaces of medical devices

Povzetek: Chitosan (Chi) and 77KS, a lysine-derived surfactant, form polyelectrolyte complexes that reverse their charge from positive to negative at higher 77KS concentrations, forming aggregates that have been embedded with amoxicillin (AMOX). Dispersion of this complex was used to coat polydimethylsiloxane (PDMS) films, with an additional layer of anionic and hydrophilic hyaluronic acid (HA) as an outer adsorbate layer to enhance protein repulsion in addition to antimicrobial activity by forming a highly hydrated layer in combination with steric hindrance. The formed polysaccharide-based bilayer on PDMS was analyzed by water contact angle measurements, X-ray photoelectron spectroscopy (XPS), and surface zeta (ζ)-potential. All measurements show the existence and adhesion of the two layers on the PDMS surface. Part of this study was devoted to understanding the underlying protein adsorption phenomena and identifying the mechanisms associated with biofouling. Thus, the adsorption of a mixed-protein solution (bovine serum albumin, fibrinogen, γ-globulin) on PDMS surfaces was studied to test the antifouling properties. The adsorption experiments were performed using a quartz crystal microbalance with dissipation monitoring (QCM-D) and showed improved antifouling properties by these polysaccharide-based bilayer coatings compared to a reference or for only one layer, i.e., the complex. This proves the benefit of a second hyaluronic acid layer. Microbiological and biocompatibility tests were also performed on real samples, i.e., silicone discs, showing the perspective of the prepared bilayer coating for medical devices such as prostheses, catheters (balloon angioplasty, intravascular), delivery systems (sheaths, implants), and stents.
