For brown trout (Salmo trutta complex), estimating divergence times remains a challenge due to the lack of well-defined time calibration points and insufficient phylogeographic coverage in previous studies. The present work aims to improve molecular dating of mitochondrial control region sequences by using a multicalibration framework based on the latest paleogeological evidence for dating the origin of Lake Ohrid and two available Salmo fossils, including the overlooked Salmo immigratus. Our results clearly show that, contrary to common belief, the major divisions within the brown trout occurred in the Late Pliocene, not the Pleistocene. The Pliocene origin suggests that the brown trout lineages did not form because of geo(hydro)morphological changes during glaciation cycles but may be the result of orogeny and drainage evolution. In addition, increased sampling, particularly in Serbia, led to the identification of a new haplogroup (da-int) occupying an intermediate position with respect to da-es and da-bs haplogroups. While the control region can delineate brown trout lineages, its phylogenetic resolution is limited, so even extensive sampling could not further resolve the lineage level polytomies. A new cyclopoid genus (Copepoda, Crustacea) from a deep aquifer in northeastern Thailand with comments on peculiar sampling sites and local faunaAnton Brancelj, Chaichat Boonyanusith, La-orsri Sanoamuang, 2024, izvirni znanstveni članek Povzetek: Pseudohesperocyclops loeiensis, new genus, new species, was collected from plastic containers that receives water from a 120-meter-deep well drilled into Cretaceous formations composed of sandstone, claystone, siltstone, conglomerate, and gypsum in Loei province, northeast Thailand. The new genus differs from the related genera Metacyclops Kiefer, 1927 and Hesperocyclops Herbst, 1984, by the shape of the female genital double somite, the swimming leg armature formula, and the P4 Enp number of segments, as well as on the form of P5. The new species shares several characters with Metacyclops cushae Reid, 1991, and Metacyclops thailandicus Boonyanusith, Sanoamuang & Brancelj, 2018, which are moved to this new genus. All three members of the new genus differ in the shape of the female genital double somite, the anal operculum, the furcal rami, and the P1 Enp-2 apical spine. Detailed comments on the regional ecology and sampling methods are included to promote collecting samples from other, less common habitats. Băncilă, Teo Delić, Cene Fišer, Nataša Mori, Žiga Ogorelec, Maja Zagmajster, 2024, izvirni znanstveni članek Povzetek: Subterranean ecosystems (comprising terrestrial, semi-aquatic, and aquatic components) are increasingly threatened by human activities; however, the current network of surface-protected areas is inadequate to safeguard subterranean biodiversity. Establishing protected areas for subterranean ecosystems is challenging. First, there are technical obstacles in mapping three-dimensional ecosystems with uncertain boundaries. Second, the rarity and endemism of subterranean organisms, combined with a scarcity of taxonomists, delays the accumulation of essential biodiversity knowledge. Third, establishing agreements to preserve subterranean ecosystems requires collaboration among multiple actors with often competing interests. This perspective addresses the challenges of preserving subterranean biodiversity through protected areas. Even in the face of uncertainties, we suggest it is both timely and critical to assess general criteria for subterranean biodiversity protection and implement them based on precautionary principles. To this end, we examine the current status of European protected areas and discuss solutions to improve their coverage of subterranean ecosystems. Janisiewicz, 2020, izvirni znanstveni članek Povzetek: The "yeast-like" fungus, Aureobasidium pullulans, isolated from fruit and leaves exhibits strong biocontrol activity against postharvest decays on various fruit. Some strains were even developed into commercial products. We obtained 20 of these strains and investigated their characteristics related to biocontrol. Phylogenetic analyses based on internal transcribed spacer (ITS) and the D1/D2 domains of rRNA 28S gene regions confirmed that all the strains are most closely related to A. pullulans species. All strains grew at 0°C, which is very important to control decay at low storage temperature, and none grew at 37°C, which eliminates concern for human safety. Eighteen strains survived 2 hrs exposures to 50°C and two strains even survived for 24 hrs. Salt-tolerance varied; however, all strains grew on medium with 14% NaCl and 14 even with 18% NaCl. Such tolerances to high temperature and elevated salinity enable compatibility with postharvest practices. Substantial differences were observed in enzymatic activity, especially with respect to production of chitinases, xylanase, or urease. Siderophore production was detected and the ability to form biofilm varied widely between the strains. Knowledge of common characteristics of these strains may be very useful in future selection of the best antagonists within this species. 40. One-step RT-droplet digital PCR : a breakthrough in the quantification of waterborne RNA virusesNejc Rački, Dany Morisset, Ion Gutiérrez-Aguirre, Maja Ravnikar, 2014, izvirni znanstveni članek Povzetek: Water contamination by viruses has an increasing worldwide impact on human health, and has led to requirements for accurate and quantitative molecular tools. Here, we report the first one-step reverse-transcription droplet digital PCR-based absolute quantification of a RNA virus (rotavirus) in different types of surface water samples. This quantification method proved to be more precise and more tolerant to inhibitory substances than the benchmarking reverse-transcription real-time PCR (RT-qPCR), and needs no standard curve. This new tool is fully amenable for the quantification of viruses in the particularly low concentrations usually found in water samples.
