Expression of FGFR1-4 in malignant pleural mesothelioma tissue and corresponding cell lines and its relationship to patient survival and FGFR inhibitor sensitivity

Povzetek: Malignant pleural mesothelioma (MPM) is a devastating malignancy with limited therapeutic options. Fibroblast growth factor receptors (FGFR) and their ligands were shown to contribute to MPM aggressiveness and it was suggested that subgroups of MPM patients could benefit from FGFR-targeted inhibitors. In the current investigation, we determined the expression of all four FGFRs (FGFR1-FGFR4) by immunohistochemistry in tissue samples from 94 MPM patients. From 13 of these patients, we were able to establish stable cell lines, which were subjected to FGFR1-4 staining, transcript analysis by quantitative RT-PCR, and treatment with the FGFR inhibitor infigratinib. While FGFR1 and FGFR2 were widely expressed in MPM tissue and cell lines, FGFR3 and FGFR4 showed more restricted expression. FGFR1 and FGFR2 showed no correlation with clinicopathologic data or patient survival, but presence of FGFR3 in 42% and of FGFR4 in 7% of patients correlated with shorter overall survival. Immunostaining in cell lines was more homogenous than in the corresponding tissue samples. Neither transcript nor protein expression of FGFR1-4 correlated with response to infigratinib treatment in MPM cell lines. We conclude that FGFR3 and FGFR4, but not FGFR1 or FGFR2, have prognostic significance in MPM and that FGFR expression is not suffcient to predict FGFR inhibitor response in MPM cell lines.

Analiza uspešnosti prodaje gozdnih lesnih sortimentov v gozdarski zadrugi lastnikov gozdov Pohorje - Kozjak

Povzetek: V prispevku je na primeru Gozdarske zadruge lastnikov gozdov Pohorje - Kozjak (zadruga) analizirana prodaja gozdnih lesnih sortimentov (GLS) za obdobje 2013%2017. S pomočjo obračunov prodaje GLS smo pridobili podatke o količini in cenah GLS, o prodajalcih GLS ter trgu. Za analizo uspešnosti prodaje GLS smo zadružne cene primerjali s povprečnimi slovenskimi cenami. S pomočjo anketnega vprašalnika smo pridobili podatke o zadovoljstvu članov zadruge z odkupnimi cenami ter samim delovanjem zadruge. Zadruga je večino lesa odkupila od nečlanov zadruge ter ga v veliki meri prodala v sosednjo Avstrijo. Odkupna cena za GLS je v zadrugi za 18,3 % višja od povprečne slovenske. Člani zadruge so v povprečju zadovoljni s cenami za odkupljene GLS, a se kljub temu 37,5 % anketiranih odloča za prodajo tudi drugim odkupovalcem, kar dolgoročno pomeni problem. Na podlagi rezultatov zaključimo, da se je zadružni model poslovnega povezovanja lastnikov gozdov izkazal kot uspešen, zato je nujno, da začne država prek gozdne politike intenzivneje pospeševati in finančno podpirati tovrstne modele poslovnega povezovanja.

Pulmonary pathology and COVID-19 : lessons from autopsy : the experience of European pulmonary pathologists

Povzetek: Since its initial recognition in December 2019, Coronavirus disease 19 (COVID-19) has quickly spread to a pandemic infectious disease. The causative agent has been recognized as a novel coronavirus, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), primarily affecting the respiratory tract. To date, no vaccines are available nor any specific treatment. To limit the number of infections, strict directives have been issued by governments that have been translated into equally rigorous guidelines notably for post-mortem examinations by international and national scientific societies. The recommendations for biosafety control required during specimen collection and handling have strongly limited the practice of autopsies of the COVID-19 patients to a few adequate laboratories. A full pathological examination has always been considered an important tool to better understand the pathophysiology of diseases, especially when the knowledge of an emerging disorder is limited and the impact on the healthcare system is significant. The first evidence of diffuse alveolar damage in the context of an acute respiratory distress syndrome has now been joined by the latest findings that report a more complex scenario in COVID-19, including a vascular involvement and a wide spectrum of associated pathologies. Ancillary tools such as electron microscopy and molecular biology used on autoptic tissue samples from autopsy are also significantly contributing to confirm and/or identify new aspects useful for a deeper knowledge of the pathogenetic mechanisms. This article will review and summarize the pathological findings described in COVID-19 until now, chiefly focusing on the respiratory tract, highlighting the importance of autopsy towards a better knowledge of this disease.
