The influence of storage conditions and DNA extraction protocol on the results of molecular analysis of the European spruce bark beetle (Ips typographus L.)

Povzetek: One of the key steps of the molecular identification of bark beetles is obtaining a sufficient quantity of high-quality DNA extract. In this study, we investigated the influence of different storage procedures for Ips typographus (L.) specimens and various DNA extraction protocols on the quantity and quality of DNA intended for use in molecular diagnostics. Adult beetles were frozen at -20 °C, either dry or in ethanol. We tested four different protocols for DNA extraction. We compared the quantity of extracted DNA and assessed its quality with PCR and Sanger sequencing. Different storage protocols had no significant effect on the quantity of DNA extracted. However, freezing specimens in ethanol provided higher-quality DNA for molecular applications. Only two of the extraction protocols produced sequenceable amplicons, and the difference in the amount of extracted DNA between them was not significant. We propose the optimal combination of storing specimens in ethanol at -20°C and using the Nucleospin Insect DNA extraction kit from Macherey Nagel, enabling a timeefficient identification process.

Citizen science is a vital partnership for invasive alien species management and research

Povzetek: Invasive alien species (IAS) adversely impact biodiversity, ecosystem functions and socio-economics. Citizen science can be an effective tool for IAS surveillance, management and research, providing large datasets over wide spatial extents and long time periods, with public participants generating knowledge that supports action. We demonstrate how citizen science has contributed knowledge across the biological invasion process, especially for early detection and distribution mapping. However, we recommend that citizen science could be used more for assessing impacts and evaluating the success of IAS management. Citizen science does have limitations and we explore solutions to two key challenges: ensuring data accuracy and dealing with uneven spatial coverage of potential recorders (which limits the dataset’s ‘fit for purpose’). Greater co-development of citizen science with public stakeholders will help us better realise its potential across the biological invasion process and across ecosystems globally while meeting the needs of participants, local communities, scientists and decision-makers.

Efficacy of trapping protocols for Agrilus jewel beetles : a multi-country assessment

Povzetek: The genus Agrilus is one of the most diverse insect genera worldwide. The larval feeding activity causes extensive damage in both forests and orchards. In addition, more than 30 species have been introduced outside their native range so far, including the emerald ash borer Agrilus planipennis Fairmaire. Thus, the availability of efficient trapping protocols for early detection of Agrilus species at entry points is of utmost importance. In this study we tested whether trapping protocols developed for surveillance of A. planipennis in North America were also effective for other Agrilus species. In particular, through a multi-country assessment we compared the efficacy of detecting Agrilus species on: (i) green glue-coated prism traps vs. green Fluon-coated multi-funnel traps when baited with the green leaf volatile (Z)-3-hexenol or left unbaited; and (ii) green multi-panel traps vs. green multi-panel traps baited with dead adult Agrilus beetles (decoys). A total of 23,481 individuals from 45 Agrilus species were caught. Trap design significantly affected both species richness and abundance of Agrilus species in several of the countries where the trapping experiments were carried out, and green prism traps outperformed green multi-funnel traps in most cases. On the contrary, the addition of a (Z)-3-hexenol lure or dead adult beetle decoys on to traps did not improve trap catches. Our study highlights that reliable trap models to survey Agrilus species are already available, but also that there is the clear need to further investigate chemical ecology of Agrilus species to develop semiochemical lures that can improve detection efficacy.

Razširjenost dvojnozobega smrekovega lubadarja (Ips duplicatus) v Sloveniji v letu 2023

Povzetek: Članek prikazuje rezultate raziskave razširjenosti vrste Ips duplicatus (Sahlberg, 1836) v Sloveniji v letu 2023. Uporabili smo 14 režastih pasti tipa Theysohn, ki so bile opremljene s feromonom Dupliwit. V raziskavo smo vključili tudi tri križne prestrezne pasti brez feromona, ki so bile postavljene v sklopu drugega projekta in smo v njih odkrili vrsto I. duplicatus. Prisotnost vrste smo potrdili na šestih lokacijah, skupno pa smo ujeli osem osebkov. Predstavljena je razprava o pomenu teh najdb v kontekstu razširjenosti, spremljanja in upravljanja dvojnozobega smrekovega lubadarja.

Preverjanje verjetnostne napovedi sanitarnega poseka smreke zaradi podlubnikov v Sloveniji v 2023

Povzetek: Preverili smo zanesljivost verjetnostne napovedi sanitarnega poseka smreke zaradi podlubnikov v Sloveniji v 2023. Verjetnostni model za napoved sanitarnega poseka smreke zaradi podlubnikov je potrdil visoko zanesljivost (AUC modela = 0,89, AUC napovedi = 0,84). Ugotovili smo optimalni prag za verjetnost sanitarnega poseka, ki ga bomo lahko uporabili pri naslednjih napovedih za bolj jasno določitev območij, kjer se bodo potencialno pojavila žarišča smrekovih podlubnikov. Napoved za leto 2023 smo naredili s pragom 0,30, ki pa se je izkazal za prenizkega, saj je bila kar tretjina modelskih celic lažno pozitivnih. Optimalen prag za verjetnostni model v letu 2023 je bil 0,40. Povprečen optimalen prag v obdobju 2020–2023 je bil 0,45, ki ga predlagamo za izdelavo verjetnostne napovedi v naslednjem letu.

Using the IUCN environmental impact classification for alien taxa to inform decision-making

Povzetek: The Environmental Impact Classification for Alien Taxa (EICAT) is an important tool for biological invasion policy and management and has been adopted as an International Union for Conservation of Nature (IUCN) standard to measure the severity of environmental impacts caused by organisms living outside their native ranges. EICAT has already been incorporated into some national and local decision-making procedures, making it a particularly relevant resource for addressing the impact of non-native species. Recently, some of the underlying conceptual principles of EICAT, particularly those related to the use of the precautionary approach, have been challenged. Although still relatively new, guidelines for the application and interpretation of EICAT will be periodically revisited by the IUCN community, based on scientific evidence, to improve the process. Some of the criticisms recently raised are based on subjectively selected assumptions that cannot be generalized and may harm global efforts to manage biological invasions. EICAT adopts a precautionary principle by considering a species’ impact history elsewhere because some taxa have traits that can make them inherently more harmful. Furthermore, non-native species are often important drivers of biodiversity loss even in the presence of other pressures. Ignoring the precautionary principle when tackling the impacts of non-native species has led to devastating consequences for human well-being, biodiversity, and ecosystems, as well as poor management outcomes, and thus to significant economic costs. EICAT is a relevant tool because it supports prioritization and management of non-native species and meeting and monitoring progress toward the Kunming–Montreal Global Biodiversity Framework (GBF) Target 6.
