Cortico-muscular phase connectivity during an isometric knee extension task in people with early Parkinson’s disease

Abstract: — Introduction: Parkinson’s disease (PD) is characterized by enhanced beta-band activity (13–30 Hz) in the motor control regions. Simultaneously, corticomuscular (CM) connectivity in the beta-band during isometric contractions tends to decline with age, in various diseases, and under dual-task conditions. Objective: This study aimed to characterize electroencephalograph (EEG) and electromyograph (EMG) power spectra during a motor task, assess CM phase connectivity, and explore how these measures are modulated by an additional cognitive task. Specifically, we focused on the beta-band to explore the relationship between heightened beta amplitude and reduced beta CM connectivity. Methodology: Early-stage people with PD and age-matched controls performed an isometric knee extension task, a cognitive task, and a combined dual task, while EEG (128ch) and EMG (2x32ch) were recorded. CM phase connectivity was assessed through phase coherence and a phase dynamics model. Results: The EEG power spectrum revealed no cohort differences in the beta-band. EMG also showed no differences up to 80 Hz. However, the combined EEG-EMG analysis uncovered reduced beta phase coherence in people with early PD during the motor task. CM phase coherence exhibited distinct scalp topography and frequency ranges compared to the EEG power spectrum, suggesting different mechanisms for pathological beta increase and CM connectivity. Additionally, phase dynamics modelling indicated stronger directional coupling from the cortex to the active muscle and less prominent phase coupling across people with PD. Despite high inter-individual variability, these metrics may prove useful for personalized assessments, particularly in people with heightened CM connectivity.

Exposure to specific fungal lectins during adhesionimpairs biofilm formation of Listeria on polystyrene

Abstract: Listeria monocytogenes is a pathogenic bacterium that can form biofilms in food processing plants, allowing the bacteria to survive despite the control measures applied. As the surface of the bacteria is covered with versatile polysaccharides and proteins, these influence the interactions of the bacterium with any surface. The unique properties and high stability of fungal proteins make them good candidates for the control of bacteria by targeting surface structures. We screened a group of fungal lectins and protease inhibitors from different fungal species, protein folds and known targets for their antibacterial and antibiofilm activity against model strains of Listeria innocua and Listeria monocytogenes. Several of them significantly decreased the viability of biofilm bacteria, but had no effect on bacterial growth parameters at 37°C and thus had no antibacterial activity. Fungal lectins significantly impaired biofilm development even at room temperature, which was attributed to exposure to lectins during adhesion. The tested fungal proteins also reduced biofilm development on biological model surfaces. The observed antibiofilm activity of fungal proteins suggests that they have the potential to modulate interactions between bacteria and/or between bacteria and surfaces, which could be used in the future to reduce surface contamination by Listeria.

Chloroplast vesiculation and induced chloroplast vesiculation and senescence-associated gene 12 expression during tomato flower pedicel abscission

Abstract: Abscission is a tightly regulated process in which plants shed unnecessary, infected, damaged, or aging organs, as well as ripe fruits, through predetermined abscission zones in response to developmental, hormonal, and environmental signals. Despite its importance, the underlying mechanisms remain incompletely understood. This study highlights the deleterious effects of abscission on chloroplast ultrastructure in the cells of the tomato flower pedicel abscission zone, revealing spatiotemporal differential gene expression and key transcriptional networks involved in chloroplast vesiculation during abscission. Significant changes in chloroplast structure and vesicle formation were observed 8 and 14 h after abscission induction, coinciding with the differential expression of vesiculation-related genes, particularly with upregulation of Senescence-Associated Gene 12 (SAG12) and Chloroplast Vesiculation (CV). This suggests a possible vesicle transport of chloroplast degrading material for recycling by autophagy-independent senescence-associated vacuoles (SAVs) and CV-containing vesicles (CCVs). Ethylene signaling appears to be involved in the regulation of these processes, as treatment with a competitive inhibitor of ethylene action, 1-methylcyclopropene, delayed vesiculation, reduced the expression of SAG12, and increased expression of Curvature Thylakoid 1A (CURT1A). In addition, chloroplast vesiculation during abscission was associated with differential expression of photosynthesis-related genes, particularly those involved in light reactions, underscoring the possible functional impact of the observed structural changes. This work provides new insights into the molecular and ultrastructural mechanisms underlying abscission and offers potential new targets for agricultural or biotechnological applications.
