112. Candidate pathogenicity factor/effector proteins of ‘Candidatus Phytoplasma solani’ modulate plant carbohydrate metabolism, accelerate the ascorbate–glutathione cycle, and induce autophagosomesMarina Dermastia, Špela Tomaž, Rebeka Strah, Tjaša Lukan, Anna Coll Rius, Barbara Dušak, Timotej Čepin, Aleš Kladnik, Maja Zagorščak, Kristina Gruden, Maruša Pompe Novak, 2023, original scientific article Abstract: The pathogenicity of intracellular plant pathogenic bacteria is associated with the action of pathogenicity factors/effectors, but their physiological roles for most phytoplasma species, including ‘Candidiatus Phytoplasma solani’ are unknown. Six putative pathogenicity factors/effectors from six different strains of ‘Ca. P. solani’ were selected by bioinformatic analysis. The way in which they manipulate the host cellular machinery was elucidated by analyzing Nicotiana benthamiana leaves after Agrobacterium-mediated transient transformation with the pathogenicity factor/effector constructs using confocal microscopy, pull-down, and co-immunoprecipitation, and enzyme assays. Candidate pathogenicity factors/effectors were shown to modulate plant carbohydrate metabolism and the ascorbate–glutathione cycle and to induce autophagosomes. PoStoSP06, PoStoSP13, and PoStoSP28 were localized in the nucleus and cytosol. The most active effector in the processes studied was PoStoSP06. PoStoSP18 was associated with an increase in phosphoglucomutase activity, whereas PoStoSP28, previously annotated as an antigenic membrane protein StAMP, specifically interacted with phosphoglucomutase. PoStoSP04 induced only the ascorbate–glutathione cycle along with other pathogenicity factors/effectors. Candidate pathogenicity factors/effectors were involved in reprogramming host carbohydrate metabolism in favor of phytoplasma own growth and infection. They were specifically associated with three distinct metabolic pathways leading to fructose-6-phosphate as an input substrate for glycolysis. The possible significance of autophagosome induction by PoStoSP28 is discussed. 113. Tribocorrosive study of new and in vivo exposed nickel titanium and stainless steel orthodontic archwiresTadeja Kosec, Petra Močnik, Uroš Mezeg, Andraž Legat, Maja Ovsenik, Monika Jenko, John T. Grant, Jasmina Primožič, 2020, original scientific article Abstract: The surface, corrosion and wear properties of new and in vivo exposed nickel titanium (NiTi) and stainless steel (SS) archwires used in orthodontic treatment were investigated. Electrochemical and tribo-electrochemical tests in artificial saliva were performed in order to define corrosion properties and to estimate wear rate of new and in vivo exposed NiTi and SS archwires. The surface chemical analysis of the passive film on the NiTi and SS archwires before and after tribocorrosion tests was performed by Auger Electron Spectroscopy (AES). In vivo exposed NiTi and SS archwires had better electrochemical properties than new archwires due to the protective nature of oral deposits. Total wear and coefficients of friction were higher among in vivo exposed archwires and higher in NiTi archwires in comparison to SS archwires. The estimated thickness of the TiO2 passive film on as-received NiTi is 8 nm, while the passive Cr2O3 film on as-received SS is just 1–2 nm. On in vivo exposed NiTi archwire, a 60–80 nm thick organic film/dental plaque was observed, and on SS, it was thinner, at about 60 nm. This research shows the importance of combining AES with electrochemical testing, to characterize tribocorrosive properties of NiTi and SS orthodontic archwires. 116. A red Roman column from Emona (Ljubljana, Slovenia)Andreja Maver, Bernarda Županek, Maja Gutman, Dragomir Skaberne, Sabina Dolenec, 2020, original scientific article Abstract: The excavation conducted in 1999 in Ljubljana (Slovenia), in Insula XXVII of the Roman colonia Iulia Emona, un-earthed the remains of a column shaft with an organic core set onto a stone block. The contribution presents this find, its context and likely parallels, the mineralogical-petrographic composition of the stucco, type of the pigments used, as well as the rock of the plinth and its provenance. Of the shaft, only the stucco survives, applied in four mortar layers and painted in red ochre, while its interior surface shows the impressions of reeds. The plinth is a rectangular block made of locally available Lower Jurassic oolitic limestone, presumably from Podpeč. The column may have formed part of a porticus or peristyle, possibly associated with a pool or a bath complex in this part of Late Roman Emona. 120. Measurements of bridge dynamic amplification factor using bridge weigh-in-motion dataJan Kalin, Aleš Žnidarič, Andrej Anžlin, Maja Kreslin, 2022, original scientific article Abstract: The dynamic component of bridge traffic loading is commonly taken into account with a Dynamic Amplification Factor (DAF)—the ratio between the dynamic and static load effects on a bridge. In the design codes, this factor is generally more conservative than in reality. Recently a new method of cal- culation of this factor had been developed. Data from 15 different bridges have been analysed since then and this paper presents the results of the analyses. The background for Bridge Weigh-in-Motion is given, and the most recent method for DAF calculation is described. The sites from which the data originated are presented, and the selection of data discussed. The results of the analyses are presented and discussed and some examples of DAF calculations are shown. Data from the considered sites have invariably demonstrated a DAF decrease with increasing axle load. This is a significant result, especially for assessment of existing structures, since it is beneficial to use measured structural param- eters to optimise structural analysis.
