The influence of storage conditions and DNA extraction protocol on the results of molecular analysis of the European spruce bark beetle (Ips typographus L.)

Abstract: One of the key steps of the molecular identification of bark beetles is obtaining a sufficient quantity of high-quality DNA extract. In this study, we investigated the influence of different storage procedures for Ips typographus (L.) specimens and various DNA extraction protocols on the quantity and quality of DNA intended for use in molecular diagnostics. Adult beetles were frozen at -20 °C, either dry or in ethanol. We tested four different protocols for DNA extraction. We compared the quantity of extracted DNA and assessed its quality with PCR and Sanger sequencing. Different storage protocols had no significant effect on the quantity of DNA extracted. However, freezing specimens in ethanol provided higher-quality DNA for molecular applications. Only two of the extraction protocols produced sequenceable amplicons, and the difference in the amount of extracted DNA between them was not significant. We propose the optimal combination of storing specimens in ethanol at -20°C and using the Nucleospin Insect DNA extraction kit from Macherey Nagel, enabling a timeefficient identification process.

Tribological evaluation of vegetable ▫$oil/MoS_2$▫ nanotube-based lubrication of laser-textured stainless steel

Abstract: In the present work, the functionalisation of austenitic stainless steel, AISI 316L surfaces via nanosecond Nd:YAG laser texturing in order to modify the surface morphology with crosshatch and dimple patterns is presented. A tribological analysis under lubrication with sunflower and jojoba oil with and without the addition of a solid lubricant, MoS2 nanotubes, was performed. In conjunction with friction/wear response laser-textured surface wettability, oil spreadability and oil retention capacity were also analysed. It was shown that the crosshatch pattern generally exhibited lower friction than the dimple pattern, with the addition of MoS2 nanotubes not having any significant effect on the coefficient of friction under the investigated contact conditions. This was found in addition to the better oil spreadability and oil retention capacity results of the crosshatch-textured surface. Furthermore, texturing reduced the wear of the stainless-steel surfaces but led to an approximately one order of magnitude larger wear rate of the steel counter-body, primarily due to the presence of hard bulges around the textured patterns. Overall, the crosshatch pattern showed better oil retention capacity and lower friction in combination with different vegetable oils, thus making it a promising choice for improving tribological performance in various environmentally friendly applications.

Recovery study of gold nanoparticle markers from lateral flow immunoassays

Abstract: Lateral flow immunoassays (LFIAs) are a simple diagnostic device used to detect targeted analytes. Wasted and unused rapid antigen lateral flow immunoassays represent mass waste that needs to be broken down and recycled into new material components. The aim of this study was to recover gold nanoparticles that are used as markers in lateral flow immunoassays. For this purpose, a dissolution process with aqua regia was utilised, where gold nanoparticles were released from the lateral flow immunoassay conjugate pads. The obtained solution was then concentrated further with gold chloride salt (HAuCl4) so that it could be used for the synthesis of new gold nanoparticles in the process of ultrasonic spray pyrolysis (USP). Various characterisation methods including scanning electron microscopy, transmission electron microscopy, ultraviolet-visible spectroscopy and optical emission spectrometry with inductively coupled plasma were used during this study. The results of this study showed that the recovery of gold nanoparticles from lateral flow immunoassays is possible, and the newly synthesised gold nanoparticles represent the possibility for incorporation into new products.

Abstract: Lo scopo della conferenza di apertura del progetto ECO2SMART è promuovere gli approcci ecosistemici (EbA e Eco-DRR) per l’adattamento ai cambiamenti climatici e la riduzione del rischio di catastrofi. Intendiamo raggiungere questo obiettivo presentando buone pratiche di soluzioni sostenibili già implementate, condividendo conoscenze, creando connessioni e promuovendo il dialogo tra le parti interessate. Nel corso della conferenza verranno illustrate le opportunità offerte dal progetto ECO2SMART ed i principali risultati attesi alla fine di questo progetto.
