Experimental investigation of the shear force capacity of prismatic cross laminated timber beams

Povzetek: Experimental tests of Cross Laminated Timber (CLT) under in-plane beam loading conditions are presented. The influence of the element layup, the individual lamination width, and the beam overhang at the supports on the shear force capacity was investigated. All the CLT beams had the same gross cross section, and a 4-point-bending test setup was used. The experimentally determined load-bearing capacities are compared with the load-bearing capacities resulting from analytical methods proposed for structural design, focusing on shear failure in the crossing areas of flatwise bonded laminations (shear failure mode III). The test results indicate no or very small influence of the element layup and the lamination width on the shear force capacity. These results partly contradict the predictions of the proposed design methods. Of the three studied beam geometry parameters, the beam overhang at the support had the greatest influence on the load-bearing capacity.

An experimental apparatus for bench-scale fire testing using electrical heating pads

Povzetek: This research study concerns an experimental, budget-friendly, electricity- powered apparatus for bench-scale fire testing. The apparatus consists of various elements, of which the most important are ceramic heating pads, used to impose heat fluxes on exposed surfaces of specimens. The test method allows to control the heating pads’ temperature and to adjust the distance between the heating pads and the specimen to obtain well-defined heat fluxes up to 50–60 kW/m2. Higher heat fluxes and temperatures can be obtained by setting the heating pads in full power mode, with or without the use of a thermal shield, which can lead to heat fluxes up to 150 kW/m2. The heating and thermal boundary conditions imposed by the apparatus are characterised and discussed, and the thickness of the convective boundary layer at the heating pads’ surface is estimated significantly lower than in the case of gas- fired radiant panels. The performance of the apparatus is analysed for various conditions: controlling the temperature of the heating pads, in an open environment or with the presence of thermal shields, and in full power mode. A few examples of application of the apparatus to fire test typical construction materials (steel and glass) are also presented. These results emphasise the well-defined heating conditions in temperature-controlled mode. The study finally discusses the advantages and limitations of the apparatus, as well as many possibilities of future applications and improvement for future research studies.

Additive manufacturing multi-material components of SAF 2507 duplex steel and 15-5 PH martensitic stainless steel

Povzetek: The study reports the successful deposition of 15–5 precipitation-hardened (PH) martensitic steel on SAF 2507 duplex stainless steel (and vice versa) to form functionally graded materials (FGMs) using powder-based, directed-energy-deposition (DED) technology. The evolution of the microstructure and the mechanical properties of the 15–5 PH/SAF 2507 functionally graded material in the as-built state were investigated systematically. The results proved that the microstructural transition zone (MTZ) is formed above the fusion line due to the dilution effect of both steels. The phase composition of the MTZ consists of ferrite and, in comparison to the base materials, an increased amount of austenite. The interface has the lowest hardness owing to the formation of a significant proportion of the austenitic phase. However, the tensile mechanical properties were not affected by the interface as failure occurred in both SAF 2507 and at the interface regions. The research presents a promising application of FGMs in a horizontal configuration to form a high - quality, metallurgical joint between heterogeneous materials. This study introduces a novel approach by additive manufacturing (AM) of heterogeneous multi-materials, merging the favorable properties of duplex SAF 2507 and martensitic 15-5 PH stainless steels through superior metallurgical bonding. The combination of SAF 2507 and 15-5 PH in a functionally graded material has not been previously explored, as the existing studies deal with the duplex stainless steels SAF 2507 and either martensitic stainless-steel 15-5 PH deposited separately by laser powder-bed-fusion (L-PBF) or DED methods. This research pioneers the investigation of these materials in tandem, paving the way for the development of novel FGMs with optimized properties for specific applications. The resulting FGM exhibits exceptional corrosion resistance and high strength, making it highly versatile for applications in diverse industries such as offshore oil and gas production, aerospace, aviation, and chemical processing equipment.

Virome analysis of irrigation water sources provides extensive insights into the diversity and distribution of plant viruses in agroecosystems

Povzetek: Plant viruses pose a significant threat to agriculture. Several are stable outside their hosts, can enter water bodies and remain infective for prolonged periods of time. Even though the quality of irrigation water is of increasing importance in the context of plant health, the presence of plant viruses in irrigation waters is understudied. In this study, we conducted a large-scale high-throughput sequencing (HTS)-based virome analysis of irrigation and surface water sources to obtain complete information about the abundance and diversity of plant viruses in such waters. We detected nucleic acids of plant viruses from 20 families, discovered several novel plant viruses from economically important taxa, like Tobamovirus and observed the influence of the water source on the present virome. By comparing viromes of water and surrounding plants, we observed presence of plant viruses in both compartments, especially in cases of large-scale outbreaks, such as that of tomato mosaic virus. Moreover, we demonstrated that water virome data can extensively inform us about the distribution and diversity of plant viruses for which only limited information is available from plants. Overall, the results of the study provided extensive insights into the virome of irrigation waters from the perspective of plant health. It also suggested that an HTS-based water virome surveillance system could be used to detect potential plant disease outbreaks and to survey the distribution and diversity of plant viruses in the ecosystem.

Characterization of hFOB 1.19 cell line for studying Zn-based degradable metallic biomaterials

Povzetek: In vitro testing is the first important step in the development of new biomaterials. The human fetal osteoblast cell line hFOB 1.19 is a very promising cell model; however, there are vast discrepancies in cultivation protocols, especially in the cultivation temperature and the presence of the selection reagent, geneticin (G418). We intended to use hFOB 1.19 for the testing of Zn-based degradable metallic materials. However, the sensitivity of hFOB 1.19 to zinc ions has not yet been studied. Therefore, we compared the toxicity of zinc towards hFOB 1.19 under different conditions and compared it with that of the L929 mouse fibroblast cell line. We also tested the cytotoxicity of three types of Zn-based biomaterials in two types of media. The presence of G418 used as a selection reagent decreased the sensitivity of hFOB 1.19 to Zn2+. hFOB 1.19 cell line was more sensitive to Zn2+ at elevated (restrictive) temperatures. hFOB 1.19 cell line was less sensitive to Zn2+ than L929 cell line (both as ZnCl2 and extracts of alloys). Therefore, the appropriate cultivation conditions of hFOB 1.19 during biomaterial testing should be chosen with caution.

Initial experiences of the first version of Slovene sustainable building Indicators that are based on Level(s)

Povzetek: To determine the possibilities for the implementation of sustainable building indicators in Slovenia, testing of the first version of the indicators, developed in the CARE4CLIMATE project and based on the EU Level(s) framework, was carried out in 2022. Invited and interested stakeholders of the construction process were provided with video content and instructions on the Slovenian e-platform of sustainable building indicators. In addition, workshops and lectures with individual subjects were also performed. The final phase of the training and testing procedure included a questionnaire, which was used to obtain information about the participants' opinions regarding the indicators. The analysis of the results of the testing, which was focused on level 2, confirmed the key preliminary finding of the development group, namely that currently, due to the lack of certain knowledge, data, and tools, all indicators for this level are not yet feasible in practice. The research also highlighted the greater need for training and specialization of experts in this field. At the same time, it showed that the testing of the first version itself was a big challenge: only 30 experts fully participated and filled out the online questionnaire. This number seems alarmingly low at first glance, but compared to level(s) testing in the EU member states, it is much more than 50 times higher. However, for the further execution of the indicators in Slovenia, it will therefore be necessary to invest a lot of effort and engagement. It is likely that state support will also be needed, for example, in the form of financial mechanisms or incentives and/or legislative background.

Depassivation and repassivation of stainless steels by stepwise pH change

Povzetek: Immersion tests with different stainless steels have been performed, while the pH was stepwise decreased and then increased again. During 8.5-day exposure, the depassivation and repassivation pH values as a function of pitting resistance equivalent number were determined. There is always a gap between both pH values (depassivation and repassivation), indicating that for every steel, there are conditions where an existing passive layer can be maintained but cannot be rebuilt after depassivation. In such environments, the passive layer is thicker, consisting mainly of molybdenum and iron rich oxides, while chromium is dissolved. Usually, depending on conditions, the passive layer is more chromium-rich, especially the inner layer. This is relevant, for example, for acidizing jobs in oil and gas industry, proving that repassivation after acidizing will happen promptly, when the pH is increased again.

Stress corrosion crack initiation testing with tapered specimens in high-temperature water - results of a collaborative research project

Povzetek: The applicability of an accelerated test technique using tapered tensile specimens for investigating the stress corrosion crack (SCC) initiation behaviour of structural materials in high-temperature water was assessed in the framework of a European collaborative research project (MICRIN – MItigation of CRack INitiation). The main advantage of using a tapered geometry is, that in a single test a stress gradient is obtained through the gauge length, and therefore a stress threshold for SCC initiation can be determined in a reasonable timeframe. This method was used to investigate two different materials that were known to be susceptible to SCC in light water reactor environment: a high-Si stainless steel and a Ni-base weld metal (Alloy 182). The results of the international test programme confirmed that the tapered specimen test methodology could be used to identify a SCC initiation stress threshold, albeit that significant scatter was present in the data.

Non-native forest tree species in Europe : ǂthe ǂquestion of seed origin in afforestation

Povzetek: Non-native forest tree species have been introduced in Europe since the 16th century, but only in the second half of the 20th century the significance of the seed source origin for their economic use was recognized, resulting in the establishment of numerous provenance trials at a national, regional, European and International level, as those led by IUFRO. Breeding programs have also been launched in the continent for the most economically important species. Aim of this work is the formulation of provenance recommendations for planting of five non-native tree species in Europe (Douglas fir, grand fir, Sitka spruce, lodgepole pine and black locust), based on the information obtained from twenty countries, in the frame of the EU FP-1403 NNEXT Cost Action. The survey revealed that official and non-official national recommendations, based on provenance research results, have been elaborated and followed at a different level and extend for the above five species, but only for Douglas fir recommendations exist in almost all the participating to the survey countries. The compilation of provenance recommendations across Europe for each species is presented in the current work. Besides the recommended introduced seed sources, European seed sources are also preferred for planting, due to ease of access and high availability of forest reproductive material. European breeding programs yielding genetic material of high productivity and quality constitute currently the seed source of choice for several species and countries. Consolidation of trial data obtained across countries will allow the joint analysis that is urgently needed to draw solid conclusions, and will facilitate the development of ‘Universal-Response-Functions’ for the species of interest, rendering possible the identification of the genetic material suitable for global change. New provenance trial series that will test seed sources from the entire climatic range of the species, established in sites falling within and outside the environmental envelopes of their natural ranges, are urgently needed to pinpoint and understand the species-specific climate constraints, as well as to correlate functional traits to the seed origin and the environmental conditions of the test sites, so that the selection of suitable forest reproductive material of non-native tree species in the face of climate change can be feasible.

Next-generation sequencing to characterize pyrazinamide resistance in Mycobacterium tuberculosis isolates from two Balkan countries

Povzetek: Objectives. Next-generation sequencing (NGS) provide a comprehensive analysis of the genetic alterations that are most commonly linked with pyrazinamide (PZA) resistance. There are no studies reporting molecular background of PZA resistance in TB isolates from Balkan Peninsula. We aimed to examine the feasibility of full-length analysis of a gene linked with PZA resistance, pncA, using Ion Torrent technology in comparison to phenotypic BACTEC MGIT 960 DST in clinical TB isolates from two countries of the Balkan Peninsula. Methods. Between 1996 and 2017, we retrospectively selected 61 TB isolates. To identify gene variants related to drug resistance in genomic DNA extracted from TB isolates, AmpliSeq libraries were generated automatically using the AmpliSeq™ Kit for Chef DL8 and the Ion AmpliSeq TB Research Panel. Result.s Of all 61 TB isolates included, 56 TB were phenotypically resistant to any antibiotic. Among them, 38/56 (67.9%) TB isolates were phenotypically resistant to pyrazinamide and pncA mutations were detected in 33/38 cases (86.8%). A mutation in the pncA promoter region was the most prevalent genetic alteration, detected in eight TB isolates. Comparison of NGS to conventional BACTEC MGIT 960 DST revealed very strong agreement (90.2%) between the two methods in identifying PZA resistance, with high sensitivity (89.5%) and specificity (95.7%) for NGS. Conclusions. Detection of PZA resistance using NGS seems to be a valuable tool for surveillance of TB drug resistance also in the Balkan Peninsula, with great potential to provide useful information at least one weak earlier than is possible with phenotypic DST.
