Construction materials for a sustainable future : proceedings of the 2nd International Conference CoMS 2020/21
2020, proceedings of peer-reviewed scientific conference contributions (international and foreign conferences)

Genetic polymorphisms of xenobiotic metabolizing enzymes in human colorectal cancer
Vita Dolžan, Metka Ravnik-Glavač, Katja Breskvar, 1998, published scientific conference contribution

Abstract: It was proposed that both hereaditary and environmental factors contribute to the development of colorectal cancer (CRC). Carcinogenic polycyclic aromatic hydrocarbons (PAHs) from food or tobacco smoke can form DNA adducts and thus initiate carcinogenesis after metabolic activation via cytochrome P4501A1 (CYP1A1). Intermediate metabolites are detoxified by conjugation with glutathione S-transferases. Our aim was to look for inherited metabolic suceptibility to CRC. We used PCR-based genotyping approach to determine the frequencies of polymorphic alleles of two cytochromes P450 (CYP2D6 and CYP1A1)and two glutathione S-transferases (GSTM1 and GSTT1) in DNA samples from 31 sporadic, 25 familial CRC cases and 73 healthy controls. The difference in frequencies of poor metabolisers due to CYP2D6 gene polymorphismwas close to the limit of statistical significance between sporadiCRC and healthy control group (lambda 2=5.52, m=2, p=0.06) despite the small sample size. The frequencies of either CYP1A1 MspI, GST M1 or GST T1 genotypes were not significantly different in both CRC cases and in controls. Although our study suggests some difference in metabolic susceptibility between sporadic and familial CRC, further studies are needed to investigate the combined effect of polymorphic genes involved in carcinogen metabolism in a larger group of patients with defined exposure to dietary carcinogens and smoking.

Diagnostic value of planar myocardial perfusion scintigraphy in patients with coronary artery disease
Marko Klančič, Metka Milčinski, Darko Zorman, 1997, original scientific article

Abstract: Patients with suspected or proven coronary artery disease are investigated using noninvasive and invasive diagnostic methods. Noninvasive myocardial perfusion scintigraphy provides data on myocardial perfusion during stress andat rest. Coronary angiography is invasive morphologic method, performed before coronary artery dilatation or surgery. Aim of our retrograde analysis of planar thallium myocardial perfusion scintigrams and coronary angiograms was to assess sensitivity and specificity of rnyocardial perfusion planar scanning and to evaluate causes of possible disagreement. Original readings ofmyocardial perfusion scans and coronary angiograms of 156 patients with coronary artery disease were compared. When results of both investigations were partially concordant or discordant, the original studies were reviewed. Concordant results of both examinations were found in 62% of patients. In only3% (5 patierrts) the results were discordant and the reason for disagreement of results of both studies could not be detected. Most of the remaining 55 patients had more pronounced myocardial perfusion defects than was the estinrated severity of cororsary artery stenosis, attributed to the different nature of both irtvestigations. Anonralous coronary artery was foundirt 3% of all patients, tortuous coronary arteries with slow ftow of coritrast media in 9 patients (6% of all) and arterial hypertension with extreme left ventricular wall hypertrophy in one patient. Serrsitivity of the rnyocardial perfusion scintigraphy was l00% and specificity 50%. Positive predictive valrre for coronary artery disease was 96% and negative predictive value was 100 %. We cconclude that myocardial perfusion scintigraphy has a definite role in: diagnosis and follow-up of patients with suspected or provencoronary artery disease. Advantages and disadvantages of hormone replacement therapy
Sonja Bebar, Marjetka Uršič-Vrščaj, 1996, review article

Abstract: In the developed countries, more than 30% of female population is in the postmenopusal period of their life. Approximately one third of these present with severe clinical menopausal symptoms, such as hot flushes, sleeplessness, night sweats, and depression. Many women search medical help. By means of hormone replacement therapy (HRT), these problems can be either completely eliminated or alleviated in 90% of women. The suspected association between HRT and risk of carcinoma should be ignored, however. While estrogens in combination with progestogens exert a protective effec against ovarian and endometrial carcinomas, a possible correlation between HRT and breast cancer has not been fully explained yet. Nevertheless, some published reports have indicated a slightly increased risk of breast cancer after a prolonged use of HRT.
