The Cancer Registry of Slovenia database was used to identify cases of laryngeal and pharyngeal NECs diagnosed between 1995%2020. Biopsies were analyzed for the expression of standard neuroendocrine markers (synaptophysin, chromogranin, CD56), INSM1, Ki-67, p16, and PD-L1 (using the combined positive score, CPS). In situ hybridization for human papillomavirus (HPV) and Epstein%Barr virus (EBV) was performed. Twenty patients (larynx, 12; pharynx, 8) were identified. One tumor was well differentiated (WD), five were moderately differentiated (MD), and 14 were poorly differentiated (PD). Disease control was achieved solely by surgery in 4/4 MD/PD T1-2N0-1 tumors. Eight patients died of the disease, seven of which were due to distant metastases. All three traditional markers were positive in 11/17 NECs and the INSM1 marker in all 20 tumors. Two of fourteen p16-positive tumors were HPV-positive, but all three nasopharyngeal NECs were EBV-negative. Three tumors had CPSs % 1. In conclusion, INSM1 was confirmed to be a reliable marker of neuroendocrine differentiation. Except in WD and early-stage MD/PD tumors, aggressive multimodal therapy is needed; the optimal systemic therapy remains to be determined. p16, HPV, and EBV seem to bear no prognostic information. Biomonitoring and assessment of toxic element contamination in floodplain sediments and soils using fluorescein diacetate (FDA) enzymatic activity measurements: evaluation of possibilities and limitations through the case study of the Drava River floodplainPéter Szabó, Gyozo Jordan, Tamás Kocsis, Katalin Posta, Levente Kardos, Robert Šajn, Jasminka Alijagić, 2022, izvirni znanstveni članek Povzetek: The EU Water Framework Directive requires the monitoring and evaluation of surface water sediment quality based on the assessment of risk posed by contamination on the biotic receptors. Floodplain sediments are important receptors of potentially toxic element (PTE) contamination from the upstream catchment areas, and floodplains host climate-sensitive riverine ecosystems and fertile agricultural areas at the same time. This study investigates the effect of PTE contamination on microbial communities in floodplain sediments and soils using the fast, inexpensive and reliable fluorescein diacetate (FDA) method in order to estimate its applicability for sediment quality monitoring and preliminary toxicity-based risk assessment. Sediment and soil samples were collected from the actively flooded alluvial plain and the river terrace areas along a 130-km stretch of the large Drava River floodplain known to be widely contaminated by historical mining, smelting and the associated industry in the upstream Alpine region. Results of detailed data analysis show that the total microbial activity represented by the measured FDA values is related to PTE (As, Cu, Zn, Cd, Pb) concentrations, but this relationship shows significant heterogeneity and depends on the spatial location and on the soil properties such as organic matter content, dissolved salt and nutrient content, and it is specific to the toxic elements. Results show that some microbe species appear to be able to adapt to the elevated PTE concentrations in toxic soil micro-environments, over time. Despite the observed heterogeneity of microbial activity, the results revealed a breakpoint in the FDA dataset around the FDA = 3 FC (fluorescein concentration) value suggesting that microbial activity is controlled by thresholds. Rast in kakovost metasekvoje (Metasequoia glyptostroboides Hu et Cheng) v živem arhivu pri LjubljaniIza Petek, Gregor Božič, Dušan Jurc, Kristjan Jarni, Robert Brus, 2022, izvirni znanstveni članek Povzetek: Analizirali smo živi arhiv metasekvoje (Metasequoia glyptostroboides Hu et Cheng) na obrečnem rastišču ob Savi pri Ljubljani. Osnovan je bil v letih 1993 in 1994 s sadikami, vzgojenimi iz originalnih semen z območja na Kitajskem, kjer je bila vrsta opisana (klasično nahajališče oz. locus classicus). Drevesa v nasadu so bila prvič izmerjena spomladi leta 2000 in nato še leta 2006, spomladi v letu 2020 pa smo meritve ponovili. V starosti 30 let je povprečno drevo merilo v višino 20,7 m, povprečni premer koreninskega vratu na višini debla 0,1 m je znašal 72,3 cm, povprečni premer debla na prsni višini pa 51,4 cm. Ugotovili smo, da je pogostnost reproduktivnih organov, torej storžev in moških cvetov, povezana s premerom in višino drevesa. Užlebljenost debla je zelo variabilna in povezana s premerom, zelo variabilno je tudi zmanjšanje premera z višino. Povezave med omenjenimi znaki in lego drevesa v nasadu nismo ugotovili. Svetloba oziroma lega v nasadu ni edini dejavnik, ki vpliva na lastnosti posameznega drevesa, vzrok je tudi genetska variabilnost med drevesi. Despite recent advances in our understanding of the physiological mechanisms leading to death, we still lack robust indicators of mortality risk that could be applied at the individual tree scale. Here, we build on a previous contribution exploring the differences in growth level between trees that died and survived a given mortality event to assess whether changes in temporal autocorrelation, variance, and synchrony in time-series of annual radial growth data can be used as early warning signals of mortality risk. Taking advantage of a unique global ring-width database of 3065 dead trees and 4389 living trees growing together at 198 sites (belonging to 36 gymnosperm and angiosperm species), we analyzed temporal changes in autocorrelation, variance, and synchrony before tree death (diachronic analysis), and also compared these metrics between trees that died and trees that survived a given mortality event (synchronic analysis). Changes in autocorrelation were a poor indicator of mortality risk. However, we found a gradual increase in interannual growth variability and a decrease in growth synchrony in the last %20 years before mortality of gymnosperms, irrespective of the cause of mortality. These changes could be associated with drought-induced alterations in carbon economy and allocation patterns. In angiosperms, we did not find any consistent changes in any metric. Such lack of any signal might be explained by the relatively high capacity of angiosperms to recover after a stress-induced growth decline. Our analysis provides a robust method for estimating early-warning signals of tree mortality based on annual growth data. In addition to the frequently reported decrease in growth rates, an increase in inter-annual growth variability and a decrease in growth synchrony may be powerful predictors of gymnosperm mortality risk, but not necessarily so for angiosperms.
