Histology basics and cell death detection in honeybee tissue

Povzetek: Honeybees (Apis mellifera L.) inside the hive (nurse workers and other hive bees) and outside the hive (foragers) are exposed to climate and weather changes, various pesticides, pathogens, and malnutrition, mainly entering through the mouth and primarily affecting the digestive tracts of adult bees. To understand and prevent the effects of such external and internal stressors on honeybees, one useful research method is the immunohistochemical method. A basic protocol is described to prepare the midgut (ventriculus) and hypopharyngeal glands (HPGs) of adult bees for histological analysis. A detailed methodology is described to assess the level of cell damage and distinguish necrosis from programmed cell death (apoptosis) as a natural process of tissue regeneration. The results of adult honeybee treatment with oxalic acid and pesticides (insecticide and acaricide) and the determination of cell death in the ventriculus and HPGs are presented. The pros and cons of the methodology are also discussed.

Irisin attenuates muscle impairment during bed rest through muscle-adipose tissue crosstalk

Povzetek: Simple Summary: Irisin is a known myokine secreted mainly by the muscle that is produced after physical activity. It induces browning in the adipose tissue with a consequent increase in mitochondrial oxidation of lipids and reduction of insulin resistance; thus, it has been hypothesized that irisin was the molecule mediating most of the beneficial effects related to exercise on adipose tissue and consequently on the whole organism. In our study we observed that extreme physical inactivity induces the loss of muscle mass and function, and an increase in the body adipose tissue as expected. However, of note, circulating irisin levels were increased secondary to enhanced irisin synthesis mainly from adipose tissue rather than muscle. In addition, subjects who produced more irisin had reduced muscle impairment. Therefore, our hypothesis is that there is negative feedback within the muscle-adipose tissue crosstalk, specifically not only does the muscle influence the adipose tissue through irisin during exercise, but also the adipose tissue protects the muscle during inactivity.Abstract: The detrimental effect of physical inactivity on muscle characteristics are well known. Irisin, an exercise-induced myokine cleaved from membrane protein fibronectin type III domain-containing protein-5 (FNDC5), mediates at least partially the metabolic benefits of exercise. This study aimed to assess the interplay between prolonged inactivity, circulating irisin, muscle performance, muscle fibers characteristics, as well as the FNDC5 gene expression (FNDC5ge) in muscle and adipose tissue among healthy subjects. Twenty-three healthy volunteers were tested before and after 14 days of Bed Rest, (BR). Post-BR circulating levels of irisin significantly increased, whereas body composition, muscle performance, and muscle fiber characteristics deteriorated. Among the subjects achieving the highest post-BR increase of irisin, the lowest reduction in maximal voluntary contraction and specific force of Fiber Slow/1, the highest increase of FNDC5ge in adipose tissue, and no variation of FNDC5ge in skeletal muscle were recorded. Subjects who had the highest FNDC5ge in adipose tissue but not in muscle tissue showed the highest circulating irisin levels and could better withstand the harmful effect of BR.

Investigating peatland stratigraphy and development of the Šijec bog (Slovenia) using near-surface geophysical methods

Povzetek: Owing to their anoxic environment, peatlands play an important role in the preservation of records documenting past atmospheric depositions. To determine past records, data on peat stratigraphy and bog development are needed. Ground penetrating radar (GPR) was used to determine the peat thickness and morphology of the Šijec bog on the Pokljuka plateau in Slovenia, which will serve as a basis for further geochemical studies. Information on the stratigraphy below the peat/clay boundary was acquired by applying electrical resistivity tomography (ERT). The GPR results reveal four depressions within the peat bog, which are separated by elevated ridges. Within the depressions the peat reaches a depth of 6–9 m. The edges of the bog are flat, with peat thickness ranging from 2 to 4 m. The reach of the GPR was complemented with manual peat probing. A comparison of the depths obtained using GPR and the peat probe reveals that the results of both methods correspond well in most locations. The ERT indicated similar peat depths; peat responds with high electrical resistivity. In contrast, clayey sediments with low resistivity are found below the peat. The peat depressions are underlain with larger clayey depressions reaching more than 20 m in thickness and represent lake sediments. The complementary geophysical methods proved to be an efficient approach with which we can delineate the peat morphology and the underlying stratigraphy. Both indicate bog formation from a lake with four deeper depressions, that are separated by glacial deposits. The results presented here show the potential for geophysical methods to infer formational processes in peatlands, showing the presence of a series of isolated basins that later coalesced into a single peat landform. This interpretation is consistent with previous conceptual models from studies in boreal regions.

Elemental associations in stream and alluvial sediments of the Savinja and Voglajna Rivers (Slovenia, EU) as a result of natural processes and anthropogenic activities

Povzetek: Stream and alluvial sediments of the Savinja and Voglajna rivers were sampled, and sediment fractions <0.063 and 0.063–0.125 mm were analyzed on the content of 60 of the main and trace elements. The objective was to determine elemental associations and identify possible sources of these associations. Differences of Al/Ti oxides ratio (9.7–26) can be attributed to the variations in the source rocks, while the K/Al oxides ratio indicates erosional or depositional river regime and variation in source rocks. One anthropogenic and three natural associations of elements were identified. The anthropogenic association (Ag, In, Sb, Cu, As, Zn, Pb, Cd, Bi, Mo and Sn) is linked to historic Zn smelting in the Celje area, and the subsequent erosion of the material from inadequately managed pyrometallurgical waste deposit. The second association (Li, Sc, Al, V, Cs and Ga) is linked to clay minerals, the third one (Mg, Ca and Te) to carbonate rocks, and the fourth one (Hf, Zr) to the heavy mineral fraction.

Stališča onkologov o bolniški odsotnosti bolnic z rakom dojk, ki so v delovnem razmerju

Povzetek: Izhodišče: Optimizacija rehabilitacije onkoloških bolnikov in njihova vrnitev na delo sta pomembni tako zaradi izboljšanja počutja te ranljive skupine kot tudi zaradi zmanjšanja družbenih in finančnih vplivov na celotno družbo. Osebe, ki so prebolele raka, se poskušajo po zdravljenju vrniti na delo in znova vzpostaviti nekdanjo strukturo vsakdanjega življenja. Z raziskavo smo želeli razkriti stališča onkologov do bolniške odsotnosti bolnic z rakom dojk. Metode: Med slovenskimi onkologi, ki sodelujejo pri zdravljenju bolnic z rakom dojk, smo izvedli krajšo anonimno spletno anketo o stališčih do bolniške odsotnosti omenjenih bolnic. Rezultati: Na spletno anketo je od 52 povabljenih onkologov odgovorilo 24 (46,2 %), od tega 8 (33,3 %) specialistov internistične onkologije, 9 (37,5 %) specialistov onkologije z radioterapijo in 7 (29,2 %) specialistov kirurgov. Stališče onkologov, ocenjeno z 10-stopenjsko Likertovo lestvico, je, da sta pogovor z bolnico in obravnava vprašanj o vrnitvi na delovno mesto zelo pomemben del celostne obravnave (povprečna ocena 8,4/10). Hkrati pa ocenjujejo, da je razmerje med časom, ki ga imajo vsakodnevno na voljo za obravnavo bolnic z rakom dojk, in časom, ki ga lahko posvetijo vprašanjem o bolniški odsotnosti med zdravljenjem ali po zaključenem zdravljenju, slabo (povprečna ocena 3,6/10). Sorazmerno pogosto se srečujejo s t. i. pritiski bolnic zaradi podajanja mnenj v zvezi z bolniško odsotnostjo ali invalidsko upokojitvijo (povprečna ocena 6,5/10). V vprašalniku so bili prav tako zbrani predlogi za dolžino bolniškega staleža, upoštevajoč vrsto in zahtevnost zdravljenja ter individualne dejavnike. Predlagana je bila skupina strokovnjakov, ki bi sestavljala multidisciplinaren tim za obravnavo bolnic z bolniškim staležem
