Timeline of leaf and cambial phenology in relation to development of initial conduits in xylem and phloem in three coexisting sub-Mediterranean deciduous tree species

Povzetek: It is unclear how the anticipated climate change will affect the timing of phenology of different tree organs/tissues and thus the whole-tree functioning. We examined the timing of leaf phenology and secondary growth in three coexisting deciduous tree species (Quercus pubescens Willd., Fraxinus ornus L. and Ostrya carpinifolia Scop) from a sub-Mediterranean region in 2019. In addition, we investigated the relationship between leaf and cambial phenology and the onset of the potential functioning of initial conduits, as determined by the completed differentiation process (vessels) or final size (sieve tubes). For this purpose, leaf development was monitored and the microcores of cambium and the youngest phloem and xylem increments were repeatedly collected at 7-10-day intervals during the growing season. The results revealed differences in the timing of leaf development and seasonal radial growth patterns in spring among the studied tree species, depending on wood porosity. We found that cambial cell production started in all cases in the first half of March. However, in ring-porous Q. pubescens and F. ornus, radial growth in the stem occurred more than a month before buds were swollen, whereas in diffuse-porous O. carpinifolia, these two events were detected at almost the same time. The end of cambial cell production occurred earliest in F. ornus (mid-July) and two weeks later also in the other two species. The widest initial earlywood vessels and early phloem sieve tubes were found in Q. pubescens, the narrowest initial earlywood vessels in O. carpinifolia and the narrowest early phloem sieve tubes in F. ornus. This indicates differences in the e%ciency of conducting systems among the studied species. This novel approach of studying phloem phenology and anatomy in relation to leaf and xylem development contributes to a better understanding of how different tree species adapt their structure of secondary vascular tissues in response to environmental change.

Expression of FGFR1-4 in malignant pleural mesothelioma tissue and corresponding cell lines and its relationship to patient survival and FGFR inhibitor sensitivity

Povzetek: Malignant pleural mesothelioma (MPM) is a devastating malignancy with limited therapeutic options. Fibroblast growth factor receptors (FGFR) and their ligands were shown to contribute to MPM aggressiveness and it was suggested that subgroups of MPM patients could benefit from FGFR-targeted inhibitors. In the current investigation, we determined the expression of all four FGFRs (FGFR1-FGFR4) by immunohistochemistry in tissue samples from 94 MPM patients. From 13 of these patients, we were able to establish stable cell lines, which were subjected to FGFR1-4 staining, transcript analysis by quantitative RT-PCR, and treatment with the FGFR inhibitor infigratinib. While FGFR1 and FGFR2 were widely expressed in MPM tissue and cell lines, FGFR3 and FGFR4 showed more restricted expression. FGFR1 and FGFR2 showed no correlation with clinicopathologic data or patient survival, but presence of FGFR3 in 42% and of FGFR4 in 7% of patients correlated with shorter overall survival. Immunostaining in cell lines was more homogenous than in the corresponding tissue samples. Neither transcript nor protein expression of FGFR1-4 correlated with response to infigratinib treatment in MPM cell lines. We conclude that FGFR3 and FGFR4, but not FGFR1 or FGFR2, have prognostic significance in MPM and that FGFR expression is not suffcient to predict FGFR inhibitor response in MPM cell lines.

Multicenter evaluation of the fully automated PCR-based Idylla EGFR Mutation Assay on formalin-fixed, paraffin-embedded Q1 tissue of human lung cancer

Povzetek: Before initiating treatment of advanced nonesmall-cell lung cancer with tyrosine kinase inhibitors (eg, erlotinib, gefitinib, osimertinib, and afatinib), which inhibit the catalytic activity of epidermal growth factor receptor (EGFR), clinical guidelines require determining the EGFR mutational status for activating (EGFR exons 18, 19, 20, or 21) and resistance (EGFR exon 20) mutations. The EGFR resistance mutation T790M should be monitored at cancer progression. The Idylla EGFR Mutation Assay, performed on the Idylla molecular diagnostics platform, is a fully automated (<2.5 hours turnaround time) sample-to-result molecular test to qualitatively detect 51 EGFR oncogene point mutations, deletions, or insertions. In a 15- center evaluation, Idylla results on 449 archived formalin-fixed, paraffin-embedded tissue sections, originating from nonesmall-cell lung cancer biopsies and resection specimens, were compared with data obtained earlier with routine reference methods, including next-generation sequencing, Sanger sequencing, pyrosequencing, mass spectrometry, and PCR-based assays. When results were discordant, a third method of analysis was performed, when possible, to confirm test results. After confirmation testing and excluding invalids/errors and discordant results by design, a concordance of 97.6% was obtained between Idylla and routine test results. Even with <10 mm2 of tissue area, a valid Idylla result was obtained in 98.9% of the cases. The Idylla EGFR Mutation Assay enables sensitive detection of most relevant EGFR mutations in concordance with current guidelines, with minimal molecular expertise or infrastructure.

Timeline of autumn phenology in temperate deciduous trees

Povzetek: Cessation of xylem formation or wood growth (CWG) and onset of foliar senescence (OFS) are key autumn phenological events in temperate deciduous trees. Their timing is fundamental for development and survival of trees, ecosystem nutrient cycling, the seasonal exchange of matter and energy between the biosphere and atmosphere and affect the impact and feedback of forests to global change. A large-scale experimental effort and improved observational methods have allowed us to compare the timing of CWG and OFS for different deciduous tree species in Western Europe, in particularly silver birch, a pioneer species, and European beech, a late-succession species, at stands of different latitudes, of different levels of site fertility, and for two years with contrasting meteorological and drought conditions i.e., the low-moderately dry 2017 and the extremely dry 2018. Specifically, we tested whether foliar senescence started before, after or concurrently with CWG. OFS and CWG occurred generally between late September and early November, with larger differences across species and sites for OFS. Foliar senescence started concurrently with CWG in most cases, except for the drier 2018 and, for beech, at the coldest site, where OFS occurred significantly later than CWG. Behavior of beech in Spain, the southern edge of its European distribution, was unclear, with no CWG, but very low wood growth at the time of OFS. Our study suggests that OFS is generally triggered by the same drivers of CWG or when wood growth decreases in late summer, indicating an overarching mechanism of sink limitation as a possible regulator of the timing of foliar senescence.

Post-fire effects on development of leaves and secondary vascular tissues in Quercus pubescens

Povzetek: An increased frequency of fire events on the Slovenian Karst is in line with future climate-change scenarios for drought-prone environments worldwide. It is therefore of the utmost importance to better understand tree-fire-climate interactions for predicting the impact of changing environment on tree functioning. To this purpose, we studied the post-fire effects on leaf development, leaf carbon isotope composition (%13C), radial growth patterns and the xylem and phloem anatomy in undamaged (H-trees) and fire-damaged trees (F-trees) of Q. pubescens with good re-sprouting ability in spring 2017, the growing season after a rangeland fire in August 2016. We found that the fully developed canopy of F-trees reached only half of the LAI values measured in H-trees. Throughout the season, F-trees were characterised by higher water potential and stomatal conductivity and achieved higher photosynthetic rates compared to unburnt H-trees. The foliage of F-trees had more negative %13C values than those of H-trees. This reflects that F-trees less frequently meet stomatal limitations due to reduced transpirational area and more favourable leaf-to-root ratio. In addition, the growth of leaves in F-trees relied more on the recent photosynthates than on reserves due to the fire disturbed starch accumulation in the previous season. Cambial production stopped 3 weeks later in F-trees, resulting in 60% and 22% wider xylem and phloem increments, respectively. A novel approach by including phloem anatomy in the analyses revealed that fire caused changes in conduit dimensions in the early phloem but not in the earlywood. However, premature formation of the tyloses in the earlywood vessels of the youngest two xylem increments in F-trees implies that xylem hydraulic integrity was also affected by heat. Analyses of secondary tissues showed that although xylem and phloem tissues are interlinked changes in their transport systems due to heat damage are not necessarily coordinated.

Seasonal radial growth of Black pine (Pinus nigra Arnold) from Bosnia and Herzegovina, monitored by the pinning method and manual band dendrometers

Povzetek: Despite numerous dendroclimatological investigations into different tree species from Bosnia and Herzegovina, information is lacking on intra-annual wood formation patterns, which would help us to interpret the climate signal in tree rings better. Using the pinning method and manual band dendrometers, we investigated the seasonal dynamics of radial growth of black pine (Pinus nigra Arnold) trees in two successive growing seasons: 2011 and 2012. The up to 60-year-old trees grew in a stand at the base of a hill in the western, mountainous part of the Balkan Peninsula. The seasonal dynamics of wood formation and final number of cells differed between the studied years. Wood formation started in both years in early to mid-March. Differences were noticed in the wood production culmination; in 2011 it occurred at the end of May and beginning of June in 2012 and 2011, respectively. Xylem growth finished in 2012 in the middle of August and in 2011 in the middle of September. Based on the first derivative of the Gompertz function calculated rate of xylem growth was lowest in 2011. The dendrometers recorded a slow increment rate in spring, higher in summer and a decreasing rate again in the late summer in both growing seasons. In comparison with pinning, dendrometers showed a delay in the start of radial growth of up to 20 days in 2012. Additionally, dendrometers showed an increase in stem girth after the end of both growing seasons, when wood formation was already completed. Deviations between the two methods could be ascribed to the influence of water storage dynamics in the main stem and numerous structural processes in bark tissue, which are captured in dendrometer data. The influence of weather conditions on xylem phenology is also indicated by differences between the two studied years, although it is difficult to identify the influence of particular short-term weather events.

Daily climate data reveal stronger climate-growth relationships for an extended European tree-ring network

Povzetek: An extended European tree-ring network was compiled from various sources of tree-ring data from Europe, northern Africa and western Asia. A total of 1860 tree-ring chronologies were used to compare correlation coefficients calculated with aggregated day-wise and month-wise mean temperature, sums of precipitation and standardised precipitation-evapotranspiration index (SPEI). For the daily approach, climate data were aggregated over periods ranging from 21 to 365 days. Absolute correlations calculated with day-wise aggregated climate data were on average higher by 0.060 (temperature data), 0.076 (precipitation data) and 0.075 (SPEI data). Bootstrapped correlations are computationally expensive and were therefore calculated on a 69.4% subset of the data. Bootstrapped correlations indicated statistically significant differences between the daily and monthly approach in approximately 1% of examples. A comparison of time windows used for calculations of correlations revealed slightly later onset and earlier ending day of the year for the daily approach, while the largest differences between the two approaches arise from window lengths: Correlations calculated with day-wise aggregated climate data were calculated using fewer days than the monthly approach. Differences in the onset and ending dates of periods for the daily and monthly approaches were greater for precipitation and SPEI data than for temperature data.

Patterns of tree microhabitats across a gradient of managed to old-growth conditions : a case study from beech dominated forests of South-Eastern Slovenia

Povzetek: An inventory of tree microhabitats was done in two unmanaged forests (Kobile and Ravna gora forest reserves) and one managed beech forest in SE Slovenia. The purpose of this study was to determine the influence of forest management, natural disturbances, and tree characteristics on microhabitat patterns. Forest structure and microhabitats were recorded in systematically placed plots (500 m2 in size) across each area. In total, we inventoried 849 trees on 54 plots and 1833 tree microhabitats. The results showed that forest management had no significant influence on the abundance of microhabitats per tree, but there were differences regarding microhabitat type between managed and unmanaged sites. There were substantially more microhabitats related to standing dead and live habitat trees in unmanaged forest (e.g. woodpecker cavities, insect galleries and bore holes, branch holes, dead branches and fruiting bodies of fungi), whereas in managed forests there were more tree microhabitats related to management (e.g. exposed heartwood, coarse bark, and epiphytic plants). The results also indicate that disturbance, tree diameter, vitality, and species influence the density, diversity, and occurrence of tree microhabitats.

O rastnih funkcijah

Povzetek: Članek ima namen z matematičnega stališča osvetliti lastnosti funkcij, ki jih gozdarji, agronomi in biologi uporabljajo za opis rasti nekaterih količin, kot višinska, debelinska, prostorninska rast dreves v odvisnosti od časa in podobno. Navadno uporabljajo za ponazoritev teh procesov elementarne, odsekoma analitične funkcije, ker na takih lahko uporabimo metode aproksimacije eksperimentalnih podatkov. Vendar take funkcije le grobo kažejo dejanska dogajanja v majhnih intervalih neodvisne spremenljivke. Zato bi bilo treba postaviti trdnejše temelje in kriterije za določevanje tipov funkcij, ki bi bile primerne za opisovanje katerih koli pojavov te vrste. Ta študija je poskus v tej smeri.
