2. Cathepsin H is a cysteine protease considered to play a major role in tumor progression, however, its precise function in tumorigenesis is unclear. Cathepsin H was recently proposed to be involved in processing of bone morphogenetic protein 4 (BMP-4) in mice. In order to clarify whether cathepsin H also regulates BMP-4 in humans, its impact on BMP-4 expression, processing and degradation was investigated in prostate cancer (PC-3), osteosarcoma (HOS) and pro-monocytic (U937) human cell lines. Materials and methods. BMP-4 expression was founded to be regulated by cathepsin H using PCR array technology and confirmed by real time PCR. Immunoassays including Western blot and confocal microscopy were used to evaluate the influence of cathepsin H on BMP-4 processing. Results. In contrast to HOS, the expression of BMP-4 mRNA in U937 and PC3 cells was significantly decreased by cathepsin H. The different regulation of BMP-4 synthesis could be associated with the absence of the mature 28 kDa cathepsin H form in HOS cells, where only the intermediate 30 kDa form was observed. No co-localization of BMP-4 and cathepsin H was observed in human cell lines and the multistep processing of BMP-4 was not altered in the presence of specific cathepsin H inhibitor. Isolated cathepsin H does not cleave mature recombinant BMP-4, neither with its amino- nor its endopeptidase activity. Conclusions. Our results exclude direct proteolytic processing of BMP-4 by cathepsin H, however, they provide support for its involvement in the regulation of BMP-4 expression. O domorodnosti alpskega kozoroga v SlovenijiAndreja Nève Repe, Elena Bužan, Borut Toškan, Jernej Javornik, Boštjan Pokorny, Andrej Arih, Lars Zver, Maruša Prostor, Miha Krofel, Matija Stergar, Klemen Jerina, Hubert Potočnik, Rok Černe, Aleš Poljanec, 2023, strokovni članek Povzetek: Alpski kozorog (Capra ibex) je alpski endemit. Na območju Slovenije naj bi bila vrsta iztrebljena v drugi polovici 17. stoletja, globalno pa je bila zaradi prelova na robu izumrtja konec 19. stoletja. Preživela je le populacija na širšem območju parka Gran Paradiso na skrajnem zahodu Alp v Italiji. Zaradi naselitev in drugih varstvenih programov sedaj alpski kozorog živi v celotnih Alpah, vključno s Slovenijo. Vendar pri nas njegove populacije nazadujejo in so v zelo slabem stanju, kar je lahko rezultat več dejavnikov. Malo izvornih osebkov ob naselitvah, zgodovinska ozka grla in ločenost kolonij so povzročili parjenje v sorodstvu, kar je slabšalo genetsko stanje populacij in lahko negativno vpliva tudi na demografijo. K številčnemu zmanjševanju vrste so lahko prispevale tudi bolezni. Za dolgoročno ohranitev alpskega kozoroga v Sloveniji so nujni takojšnji aktivni ohranitveni ukrepi, pogoj pa je ustrezna opredelitev izvornosti vrste, saj je (bila) zaradi prejšnjih pomanjkljivih podatkov umeščena med tujerodne. V prispevku na podlagi arheo-zooloških, genetskih in preliminarnih habitatnih analiz utemeljujemo, da je v Sloveniji kozorog domorodna vrsta. V raziskavah smo pokazali, da je vrsta živela na ozemlju zdajšnje Slovenije v poznoantičnem in zgodnje srednjeveškem obdobju. Preliminarno smo določili tudi primernost in povezanost habitata kozoroga v slovenskem alpskem svetu ter nakazali verjetne potrebne ukrepe za ohranitev vrste v Sloveniji. Potresni odziv večetažnih strižnih zidanih sten in učinek utrjevanja s kompozitnimi oblogamiPetra Triller, Miha Tomaževič, Matija Gams, 2018, izvirni znanstveni članek Povzetek: večetažnih sten, sezidanih iz modularnih zidakov, s poudarkom na opazovanju mehanizmov odziva in porušitve, ki se razvijejo v večetažnih stenah z odprtinami. Druga tema je osredotočena na raziskave utrjevanja takšnega zidovja z malto, armirano z mrežicami iz steklenih vlaken. Za raziskave smo na Zavodu za gradbeništvo Slovenije zgradili in preiskali posamezne zidove in večji trietažni preizkušanec v naravni velikosti. V cikličnem strigu pri konstantnem tlaku smo preizkusili več referenčnih in utrjenih zidov ter trietažni model dimenzij 4 x 4 m v tlorisu in višine 6,7 m. Trietažni model smo najprej obremenili do večjih, a popravljivih poškodb. Zatem smo poškodovani preizkušanec utrdili s kompozitnimi oblogami iz malte, armirane s steklenimi vlakni (GFRP), ter ga ponovno preiskali, tokrat skoraj do porušitve. V obeh primerih je bil odziv modela na vodoravno obtežbo s t. i. etažnim mehanizmom, pri katerem se večina poškodb skoncentrira v zidovih najšibkejše etaže. V zidovih smo opazili tipične strižne poškodbe z diagonalnimi razpokami. Potresno obnašanje posameznih zidov je bilo zelo podobno odzivu zidnih slopov večetažnega preizkušanca. Rezultati kažejo, da je mogoče z ustreznim načinom utrjevanja zidov doseči občutno izboljšanje potresnega obnašanja.
