Local site conditions reduce interspecific differences in climate sensitivity between native and non-native pines

Abstract: Two European pine species, Pinus sylvestris and Pinus nigra, are experiencing dieback as a result of the increasing frequency and intensity of extreme climatic events. Recent species distribution models predicted shrinkage of ecological niches in the near future and shifted their habitat range towards the northeast. Consequently, P. sylvestris may contract its range and P. nigra may expand in Central Europe. To test whether native pine species have an advantage over introduced pine species in acclimation to a novel climate in Central Europe, we investigated the climate sensitivity and vitality of P. sylvestris, P. nigra and P. rigida. We sampled mature stands of each pine species at three sites in Central Europe, for which we determined climate–growth relationships: temporal stability of temperature and precipitation correlations with tree-ring width and resilience indices. Based on remote sensing data, we assessed differences in surface reflectance and photosynthetic activity obtained from the normalised difference vegetation index (NDVI). Our analyses revealed that the climate sensitivity and surface reflectance of pines are not due to their nativeness in Central Europe but better explained by local site conditions. The specificity and variability of drought events may determine both the homogeneous and diverse susceptibility of species to a negative water balance. Therefore, the character of future climatic extremes seems to be the key to understanding the acclimation of native and non-native pine species in Central Europe. Because our study do not provide evidence of the superiority of non-native pine species over P. sylvestris, and the potential impacts of introduced species on local habitats seem poorly understood in the face of climate change, we urge particular caution in introducing species with unrecognized invasive potential.

Pathophysiological mechanisms of reduced physical activity : insights from the human step reduction model and animal analogues

Abstract: Physical inactivity represents a heavy burden for modern societies and is spreading worldwide, it is a recognised pandemic and is the fourth cause of global mortality. Not surprisingly, there is an increasing interest in longitudinal studies on the impact of reduced physical activity on different physiological systems. This narrative review focuses on the pathophysiological mechanisms of step reduction (SR), an experimental paradigm that involves a sudden decrease in participants’ habitual daily steps to a lower level, mimicking the effects of a sedentary lifestyle. Analogous animal models of reduced physical activity, namely the “wheel-lock” and the “cage reduction” models, which can provide the foundation for human studies, are also discussed. The empirical evidence obtained thus far shows that even brief periods of reduced physical activity can lead to substantial alterations in skeletal muscle health and metabolic function. In particular, decrements in lean/muscle mass, muscle function, muscle protein synthesis, cardiorespiratory fitness, endothelial function and insulin sensitivity, together with an increased fat mass and inflammation, have been observed. Exercise interventions seem particularly effective for counteracting these pathophysiological alterations induced by periods of reduced physical activity. A direct comparison of SR with other human models of unloading, such as bed rest and lower limb suspension/immobilisation, is presented. In addition, we propose a conceptual framework aiming to unravel the mechanisms of muscle atrophy and insulin resistance in the specific context of reduced ambulatory activity. Finally, methodological considerations, knowledge gaps and future directions for both animal and human models are also discussed in the review.

Forest managers’ perspectives on environmental changes in the biosphere reserve Mura-Drava-Danube

Abstract: Riparian forests are particularly vulnerable to environmental change and anthropogenic influences because they are highly dynamic ecosystems, thus proper adaptation measures are crucial. The implementation of these measures, however, strongly depends on the actors’ perceptions of the specific problems occurring in such forests. For understanding the constraints of specific interest groups toward different adaptation activities, information in this field is essential. By conducting a questionnaire survey we explore how different types of forest managers, i.e., forestry professionals, forest owners, and conservation managers, perceive the effects of environmental change on forest management in the recently established Transboundary Biosphere Reserve Mura-Drava-Danube. We show that these forest managers are highly aware of ongoing environmental changes and appraise deteriorating forest conditions, especially after observing changes themselves. Abiotic damage is expected to increase the most, followed by biotic damage, the spread of non-native species, and tree dieback. Nearly 80% of the survey respondents expect further changes and almost all of them intend to adapt their management of forests to mitigate or prepare for these changes. Nevertheless, we show differences in sensitivity to change and willingness to initiate adaptation actions by assessing adaptation thresholds: conservation managers appear generally more tolerant to changes, which results in higher thresholds to initiate management adaptation than forestry professionals

Next-generation sequencing to characterize pyrazinamide resistance in Mycobacterium tuberculosis isolates from two Balkan countries

Abstract: Objectives. Next-generation sequencing (NGS) provide a comprehensive analysis of the genetic alterations that are most commonly linked with pyrazinamide (PZA) resistance. There are no studies reporting molecular background of PZA resistance in TB isolates from Balkan Peninsula. We aimed to examine the feasibility of full-length analysis of a gene linked with PZA resistance, pncA, using Ion Torrent technology in comparison to phenotypic BACTEC MGIT 960 DST in clinical TB isolates from two countries of the Balkan Peninsula. Methods. Between 1996 and 2017, we retrospectively selected 61 TB isolates. To identify gene variants related to drug resistance in genomic DNA extracted from TB isolates, AmpliSeq libraries were generated automatically using the AmpliSeq™ Kit for Chef DL8 and the Ion AmpliSeq TB Research Panel. Result.s Of all 61 TB isolates included, 56 TB were phenotypically resistant to any antibiotic. Among them, 38/56 (67.9%) TB isolates were phenotypically resistant to pyrazinamide and pncA mutations were detected in 33/38 cases (86.8%). A mutation in the pncA promoter region was the most prevalent genetic alteration, detected in eight TB isolates. Comparison of NGS to conventional BACTEC MGIT 960 DST revealed very strong agreement (90.2%) between the two methods in identifying PZA resistance, with high sensitivity (89.5%) and specificity (95.7%) for NGS. Conclusions. Detection of PZA resistance using NGS seems to be a valuable tool for surveillance of TB drug resistance also in the Balkan Peninsula, with great potential to provide useful information at least one weak earlier than is possible with phenotypic DST.
