Assessment of differential expression of oncogenes in adenocarcinoma of stomach with fluorescent labeling and simultaneous amplification of gene transcripts

Abstract: Background. Gastric cancer is one of the leading malignancies with a poor prognosis and low survival rates. Although the mechanisms underlying its development are still unknown, there is a consensus that genetic instability, inactivation of tumor suppressor genes and over-expression of oncogenes are involved in the early and late stages of gastric carcinogenesis. In the present study we wanted to display differential expression of seven oncogenes,namely CCNE1, EGF, ERBB3, FGF4, HRG1, HGFR and TDGF1. Patients and methods. We employed a method based on the multiplex reverse transcription polymerase chain (RT-PCR) method with a fluorescence detection. Results. More than half of patients (74.3%) out of total 74 with gastric adenocarcinoma had over-expressed at least one oncogene, with the exception of FGF4, which was expressed in tumor tissue of less than one third of patients. 56.8% of the patients patients showed over-expression of two or more oncogenes. Conclusions. Patients with precancerous lesions had elevated levels of TDGF1 or cripto-1 (64.9%) and CCNE1 (57.1%), suggesting that they could be used as markers for an early detection of malignant changes in stomach. Finally, the fluorescent multiplex RT-PCR method could be of value for rapid assessment of oncogene mRNA levels in small samples of tumor or precancerous biopsies.

Abstract: Sorta ˈPicholineˈ je dobro prilagodljiva na različne rastne pogoje. Drevo je srednje bujno in razširjene rasti. Sorta je delno samooplodna. Uporabljamo jo lahko za predelavo v olje in za vlaganje, čemur prilagodimo tudi čas in način obiranja. Plodovi so nekoliko večji, srednje pozno se barvajo in vsebujejo srednjo količino olja z visoko vsebnostjo biofenolov in srednjo vsebnostjo tokoferolov. Olje velikokrat pri predelavi težko izločimo. Nizke temperature in sušo sorazmerno dobro prenaša. Proti pavjemu očesu je malo občutljiva, občutljiva pa je na oljčnega raka. Abstract: Sorta ˈPendolinoˈ je dober opraševalec sorte ‘Leccino’ in ‘Frantoio’. Sama je avtosterilna in za dobro oploditev potrebuje opraševalca ('Leccino', 'Moraiolo', 'Frantoio'). Drevo je močne rasti z izrazito povešajočimi tanjšimi rodnimi vejami in ozkimi ozkimi listi. V novejših nasadih se je izkazala kot sorta, ki zgodaj stopi v rodnost. Plodovi se zgodaj obarvajo v črno in dozorevajo v istem času kot 'Leccino’. Rodnost je dobra in redna, po vsebnosti olja pa je nekoliko nižja od sorte ’Leccino’. Olje ima srednjo vsebnost tokoferolov in biofenolov. Sorta je malo občutljiva za pavje oko in sajavost, odporna pa na napade oljčne muhe in oljčnega molja. Nizke temperature sorazmerno dobro prenaša. Abstract: Sorta ˈGrignanˈ je namenjena predelavi v olje. Zanjo je značilna počasna rast, krošnja pa je bolj pokončna in redka. To je sorta, ki zgodaj stopi v rodnost. Zaradi avtosterilnosti potrebuje za oploditev opraševalce. Dobri opraševalci naj bi bili 'Frantoio', 'Casaliva', 'Pendolino' in 'Leccino'. Rodnost je slabša do srednja, a konstantna. Plodovi se zgodaj začenjajo barvati, vendar neenakomerno. Vsebnost biofenolov v olju je srednja, tokoferolov pa nizka do srednja. Abstract: Sorta ˈFrantoioˈ je v svetu razširjena zaradi sposobnosti prilagajanja na različna okolja ter redne in dobre rodnosti ter kakovosti olja. Drevo je bujne rasti, mladi poganjki so dolgi in povešeni. Dozorevanje plodov je postopno in traja dalj časa. Plodovi vsebujejo več olja od sorte Leccino, olje pa ima visoko vsebnost oleinske kisline, nizko vsebnost tokoferolov in visoko vsebnost biofenolov. Na pavje oko občutljiva, na in oljčno muho je pa malosrednje občutljiva , slabše pa prenaša nizke temperature, vendar se hitro obnavlja. Znana je njena velika občutljivost na oljčnega raka, ki je pogost spremljevalec te sorte. Zelo dobro prenaša Verticillium dahliae - verticilijsko uvelost. Abstract: V okviru javne službe za izvajanje strokovnih nalog v oljkarstvu, so bile v letu 2021 izvedene naloge: selekcija, introdukcija, razmnoževanje sadilnega material, ugotavljanje ustreznih tehnologije za pridelavo oljk ter ugotavljanje vrednosti oljk za predelavo. V poročilu so zbrane morfološke in agronomske lastnosti ter značilnosti oljčnega olja sort 'Mata', 'Coratina', 'Frantoio', 'Oblica', 'Pendolino', 'Picholine' in 'Grignan' za opazovanja v letu 2021. Predstavljeni so tudi rezultati genotipizacije izbranih sort ter agronomskega vrednotenja in fenofaze različnih sort, ki so zasajene v introdukcijsko-kolekcijskem nasadu Purissima in Šempeter. V poročilu so zbrani podatki tehnoloških poskusov pridelave in predelave; parametri, ki karakterizirajo posamezna sortna olja ter parametri kakovosti oljčnega olja letnika 2021. Art v 1 IgE epitopes of patients and humanized mice are conformational

Abstract: Background: Worldwide, pollen of the weed mugwort (Artemisia vulgaris) is a major cause of severe respiratory allergy, with its major allergen, Art v 1, being the key pathogenic molecule for millions of patients. Humanized mice transgenic for a human T-cell receptor specific for the major Art v 1 T-cell epitope and the corresponding HLA have been made. Objective: We sought to characterize IgE epitopes of Art v 1–sensitized patients and humanized mice for molecular immunotherapy of mugwort allergy. Methods: Four overlapping peptides incorporating surface-exposed amino acids representing the full-length Art v 1 sequence were synthesized and used to search for IgE reactivity to sequential epitopes. For indirect mapping, peptide-specific rabbit antibodies were raised to block IgE against surface-exposed epitopes on folded Art v 1. IgE reactivity and basophil activation studies were performed in clinically defined mugwort-allergic patients. Secondary structure of recombinant (r) Art v 1 and peptides was determined by circular dichroism spectroscopy. Results: Mugwort-allergic patients and humanized mice sensitized by allergen inhalation showed IgE reactivity and/or basophil activation mainly to folded, complete Art v 1 but not to unfolded, sequential peptide epitopes. Blocking of allergic patients’ IgE with peptide-specific rabbit antisera identified a hitherto unknown major conformational IgE binding site in the C-terminal Art v 1 domain. Conclusions: Identification of the new major conformational IgE binding site on Art v 1, which can be blocked with IgG raised against non-IgE reactive Art v 1 peptides, is an important basis for the development of a hypoallergenic peptide vaccine for mugwort allergy.
