Morphological and molecular characterization of Micromeria croatica (Lamiaceae), an endemic and potentially valuable horticultural species of the Dinaric Alps

Povzetek: Due to climate extremes and limited natural resources, especially water, we can expect increased demand in the future for species that can better tolerate climate extremes such as drought. One potentially valuable horticultural species is the endemic species of the Dinaride Mountains Micromeria croatica (Pers.) Schott (family Lamiaceae). It grows in the crevices of carbonate rocks, extending from an altitude of 150 m to more than 2000 m. This study aims to provide additional insight into the genetic and morphological diversity of this endemic species, focusing on valuable horticultural traits. To achieve this goal, morphological and molecular analyses were performed on ten natural populations. Through STRUCTURE and PCoA analyses, ten M. croatica populations were placed into western and eastern genetic groups, with several individuals from western populations assigned to the eastern group and vice versa. These atypical individuals assigned to the new genetic group by BAPS analysis indicate gene flow between western and eastern populations. Similarly, an analysis of molecular variance revealed fewer genetic differences than within studied populations. Both PCA and CANDISC analysis based on eleven morphological traits largely confirmed the existence of two slightly different genetic groups. Two populations containing plants with the most flowers per shoot, one with white-flowered individuals, one with the roundest leaves, and one with the narrowest leaves proved to be the most horticulturally valuable. The genetic and morphological variability found should be a sufficient basis for the potential selection of M. croatica populations and individuals for horticultural purposes.

Schedule-dependency of doxorubicin and vinblastine in EAT tumours in mice

Povzetek: Background. Antitumour schedule-dependency of the doxorubicin and vinblastine combination was explored. Materials and methods. Intraperitoneal Ehrlich ascites tumours (EAT) syngeneic to CBA mice were treated with vinblastine ar doxorubicin alone, or in combined treatment schedules. Results. Combinations of doxorubicin and vinblasfine administered at 48-h, but not at 24-h interval,regardless of the sequence of drugs, significantly reduced the numberof tumour cells in the ascites in corrtparison with all other treatments. In the combined treatment schedules, the predominant morphologicalchanges as well as DNA distribution pattern were dependent on thefirst drug applied. Regardless of the sequence of the drugs, median survival times of animals did not significantly differ between the treatment groups. Conclusions. The effect of combination of vinblastine and doxorubicin is schedule-dependent. The time interval, but not the sequence of drugs seems to be crucial for the observed effect. The data from preclinical studies are important for planning combined treatment schedules in clinical setting.

Antitumor effectiveness of bleomycin on SA-1 tumor after pretreatment with vinblastine

Povzetek: In our previous study, vinblastine (VLB) was shown to increase the plasma membrane fluidity. This effect of VLB might be exploited for better transport of drugs through the plasma membrane. The aim of the present study was to determine whether pretreatment with VLB can increase the cytotoxic effect of BLM on intraperitoneal SA-1 tumors in mice. Materials and methods. BLM and VLBwere used as single agents or in various combinations, i.e. BLM injected 24h before VLB or vice-versa, VLB injected 24 h before BLM. Cell and animal survival together with DNA histograms were the end-points used to determine the effect of these combined treatments. Results. Both drugs, either as singletreatment or in different combined therapy schedules reduced significantly the number of cells in peritoneal lavage, compared to control, saline treated animals. The combination of VLB, followed by BLM after 24 h reduced significantly the number of cells in peritoneal lavage, compared to the treatment in which BLM was followed by VLB or to the treatment with singledrugs alone. Median survival time of mice treated with VLB alone, BLM alone and combination of both drugs was significantly prolonged compared to the control untreated mice. When VLB and BLM were combined, both treatment combinations were more effective than monochemotherapies with VLB or BLM. The best results were obtained when VLB was followed by BLM after 24 h. The DNA histogram of cells treated with VLB showed a decreased number of cells in S phase and an increased number of cells with DNA values greater than in G2M compartment compared to the control untreated cells. BLM in the dosage used inthese experiments did not affect the progression of cells through cell cycle. Both combinations of VLB and BLM produced similar cell kinetic effect as VLB alone. Conclusion. Cathepsin D and plasminogen activator inhibitor type 1 in normal, benign and alignant ovarian tissues : a preliminary report

Povzetek: Background. The aim of the present study was to determine the concentration ofcathepsin D (Cath D) and plasminogen activator inhibitor type 1 (PAI-1) in normal ovarian tissues, benign and malignant ovarian tumor tissues, and to asses relationship between Cath D and PAI-1 content, and some clinical and pathohistological parameters. Materials and methods. Cath D contents and PAI-1concentrations were determined (using immunoradiometric ELSA-Cath D assayand commercial IMUDIND R ELISA immunoassay, respectively) in 35 samples: 10 normal ovarii, 10 benign, 10 primary malignant and 5 metastatic ovarian tumors. Results. The concentrations of Cath D were significantly higher in malignant (32.89+-14.26 pmol/mg protein ) and metastatic (31.42+-10.24 pmol/mgprotein), than in normal (13.68+-4.03 pmol/mg protein) and benign (17.89+-13.13 pmol/mg protein) ovarian tissues. There was no statistical differences in the concentrations of PAI-1 between normal, benign, malignant and metastatic tumor specimens. The concentrations of Cath D as well as PAI-1 did not correlate to the age of patients, menopausal status, parity, GOG risk group, clinical stage or pathohistological grading. Conclusion. Concentrationsof Cath D (but not PAI-1) were significantly increased in malignant and metastatic ovarian tumor tissues when compared to normal and benign ovarian tumor samples; they were independent from pathohistological andclinical parameters.

Image cytometry analysis of normal buccal mucosa smears: influence of smoking and sex-related differences

Povzetek: To get more information about the influence of smoking on chromatin pattern ofnormal buccal mucosa cells and to assess sex-related differences in nuclear features, quantitative analysis of normal buccal mucosa smears was performed. In this study, buccal smears were collected from 78 healthy subjects. Image cytometry analysis was performed on Feulgen-thionin stained smears. probability distributions of 78 nuclear features were calculated for both, cell-by-cell and slide-by-slide classifications. Seven nuclear features showeddiscriminative ability between smokers and non-smokers; most of them were nuclear texture features. Statistical analysis of nuclear features in groups of females and males hoswed that only two nuclear features were different. It is concluded that smoking should be considered in image cytometry analysis of lesions in oral cavity.

Longitudinal study of malignancy associated changes in progressive cervical dysplasia

Povzetek: Eight of 29 patients with progressive CIN were followed for 2 to 10 years. Their consecutive Pap smears were destained and stained according to Feulgen thionin method. Cyto-Savant/TM high resolution image cytometer (Oncometrics Technol. Corp., Vancouver, Canaderage values of muclear texture features and their probability distributions for consecutive Pap smears from each patient were calculated. Three out of 5 discriminant MAC, highDNAamount, highDNAcomp and highDNAarea, were to increase as a function of time in 5 out of 8 patients. A preliminary analysis which was performed on non-standardized archival material demonstrated a monotonous increase of discrete texture features as a function of time in patients with progressive CIN.

Vinblastine increases antitumor effectiveness of bleomycin

Povzetek: In our previous vinblastine (VELBE) was shown to increase the plasma membrane fluidity. This effect of VELBE might be expolited for better transport of drugs through the plasma membrane. Bleomycin (BLM) is a highly cytotoxic drug when present inside the cells but has a hampered transport through the plasma membrane. The aim of the present study was to determine whether pretreatment with VELBE can increase the effect of BLM on intraperitoneal SA-1 tumors in mice. BLM and VELBE were used as single agents or in various combinations, i.e. VELBE and BLM injected simultanously, BLM injected 24 h before VELBE or VELBE injected 24 h before BLM. Mice survival was the end-point used for determining the effect of this combined treatments. VELBE and BLM as a single treatment significantly prolonged median survival time of study animals compared to controls. Furthermore, when VELBE and BLM were combined, all threetested combinations were more effective than VELBE or BLM as single treatments. The effect on animal survival was equal when VELBE was given 24 h after or simultanously with BLM. The longest survival, however was obtained when VELBE was injected 24 h before BLM. From these results we can conclude that the underlying mechanisms for more than additive effect of VELBE and BLM when VELBE was given 24 h before BLM could be attributed to an increased membrane fluidity, possibly in combination with a cell kinetic effect.
