Effects of electrochemotherapy on immunologically important modifications in tumor cells

Povzetek: Electrochemotherapy (ECT) is a clinically acknowledged method that combines the use of anticancer drugs and electrical pulses. Electrochemotherapy with bleomycin (BLM) can induce immunogenic cell death (ICD) in certain settings. However, whether this is ubiquitous over different cancer types and for other clinically relevant chemotherapeutics used with electrochemotherapy is unknown. Here, we evaluated in vitro in the B16-F10, 4T1 and CT26 murine tumor cell lines, the electrochemotherapy triggered changes in the ICD-associated damage-associated molecular patterns (DAMPs): Calreticulin (CRT), ATP, High Mobility Group Box 1 (HMGB1), and four immunologically important cellular markers: MHCI, MHC II, PD-L1 and CD40. The changes in these markers were investigated in time up to 48 h after ECT. We showed that electrochemotherapy with all three tested chemotherapeutics induced ICD-associated DAMPs, but the induced DAMP signature was cell line and chemotherapeutic concentration specific. Similarly, electrochemotherapy with CDDP, OXA or BLM modified the expression of MHC I, MHC II, PD-L1 and CD40. The potential of electrochemotherapy to change their expression was also cell line and chemotherapeutic concentration specific. Our results thus put the electrochemotherapy with clinically relevant chemotherapeutics CDDP, OXA and BLM on the map of ICD inducing therapies.

Low but significant evolutionary potential for growth, phenology and reproduction traits in European beech

Povzetek: Local survival of forest tree populations under climate change depends on existing genetic variation and their adaptability to changing environments. Responses to selection were studied in European beech (Fagus sylvatica) under field conditions. A total of 1087 adult trees, seeds, 1-year-old seedlings and established multiyear saplings were genotyped with 16 nuSSRs. Adult trees were assessed for phenotypic traits related to growth, phenology and reproduction. Parentage and paternity analyses were used to estimate effective female and male fecundity as a proxy of fitness and showed that few parents contributed to successful regeneration. Selection gradients were estimated from the relationship between traits and fecundity, while heritability and evolvability were estimated using mixed models and the breeder's equation. Larger trees bearing more fruit and early male flowering had higher total fecundity, while trees with longer growth season had lower total fecundity (directional selection). Stabilizing selection on spring phenology was found for female fecundity, highlighting the role of late frosts as a selection driver. Selection gradients for other traits varied between measurement years and the offspring cohort used to estimate parental fecundity. Compared to other studies in natural populations, we found low to moderate heritability and evolvability for most traits. Response to selection was higher for growth than for budburst, leaf senescence or reproduction traits, reflecting more consistent selection gradients across years and sex functions, and higher phenotypic variability in the population. Our study provides empirical evidence suggesting that populations of long-lived organisms such as forest trees can adapt locally, even at short-time scales.

Using bridge weigh-in-motion systems to monitor single-span bridge influence lines

Povzetek: Bridge weigh-in-motion systems use instrumented bridges or culverts to weigh vehicles as they pass over the structures. They also provide data to allow the calculation of several bridge performance indicators. The article starts with the basics of a bridge weigh-in-motion system and briefly describes two key bridge performance indicators, girder distribution factor and dynamic amplification factor, which are also derived from B-WIM measurements. The central part of the article focuses on monitoring of influence lines, the third key parameter that characterises the bridge performance under traffic loads. First, the method of calculating the bending moment influence lines from random heavy traffic is described. A coefficient of rotational stiffness is introduced, which defines the shape of influence lines around the supports as a linear combination of the ideal simply supported and fixed supported influence lines, to allow quantifying the influence line changes. Then the long-term monitoring of influence lines is investigated on four different single-span test bridges. The initial focus is given on the examination of the effect of temperature on the shape of influence lines. Finally, two sets of influence lines are compared on one test bridge, one from before and the other from after replacing the expansion joints and bearings. The work done so far confirms that calculating of influence lines from random vehicles with a B-WIM system is entirely feasible and that differences in their shape can be detected on single-span bridges. What remains to be investigated is the comparison of these differences to the actual damages and under which circumstances the proposed procedure can compete with or better the routine bridge inspection and the conventional monitoring techniques.

Growth response of European beech (Fagus sylvatica L.) and Silver Fir (Abies alba Mill.) to climate factors along the Carpathian massive

Povzetek: European forests are becoming increasingly threatened by climate change and more frequent droughts. The likely responses of species to climate change will vary, affecting their competitiveness, their existence, and consequently, forest management decisions and measures. We determined the influence of climate on the radial growth of European beech and silver fir along the Carpathians to find similarities between the two species and the main differences. Along the Carpathian Mountains, seven sites with mature fir–beech stands above 800 m above sea level were selected and analyzed. Our study confirmed different responses depending on species and location. A more pronounced response of tree growth to climate was observed on the eastern side of the Carpathians, while it was less expressed or even absent on the southern sites. Both beech and fir show better radial growth with higher precipitation in July and slower growth with higher average and maximum temperatures in June of the current year. Fir demonstrates a positive correlation between radial growth and temperature in winter, while beech demonstrates a negative correlation between radial growth and temperature in summer. In the 1951–1960 decade, the average tree ring widths in fir and beech were largest at the southern sites compared to the other sites, but since 2011, the southern sites have had the lowest increase while northern sites have had the largest. Both species respond differently to climate and are likely to follow different competitive paths in the future.

Chloroplast redox state changes mark cell-to-cell signaling in the hypersensitive response

Povzetek: Bisphenol A (BPA) is one of the most commonly used substances in the manufacture ofvarious everyday products. Growing concerns about its hazardous properties, including endocrinedisruption and genotoxicity, have led to its gradual replacement by presumably safer analogues inmanufacturing plastics. The widespread use of BPA and, more recently, its analogues has increasedtheir residues in the environment. However, our knowledge of their toxicological profiles is limitedand their combined effects are unknown. In the present study, we investigated the toxic effectscaused by single bisphenols and by the combined exposure of BPA and its two analogues, BPAP andBPC, after short (24-h) and prolonged (96-h) exposure in HepG2 spheroids. The results showed thatBPA did not reduce cell viability in HepG2 spheroids after 24-h exposure. In contrast, BPAP andBPC affected cell viability in HepG2 spheroids. Both binary mixtures (BPA/BPAP and BPA/BPC)decreased cell viability in a dose-dependent manner, but the significant difference was only observedfor the combination of BPA/BPC (both at 40μM). After 96-h exposure, none of the BPs studiedaffected cell viability in HepG2 spheroids. Only the combination of BPA/BPAP decreased cellviability in a dose-dependent manner that was significant for the combination of 4μM BPA and 4μMBPAP. None of the BPs and their binary mixtures studied affected the surface area and growth ofspheroids as measured by planimetry. In addition, all BPs and their binary mixtures studied triggeredoxidative stress, as measured by the production of reactive oxygen species and malondialdehyde,at both exposure times. Overall, the results suggest that it is important to study the effects of BPsas single compounds. It is even more important to study the effects of combined exposures, as thecombined effects may differ from those induced by single compounds.
