Circulating serum sVCAM-1 concentration inadvanced ovarian cancer patients : correlation with concentration in ascites

Abstract: Background. Vascular cell adhesion molecule-1 (VCAM-1) is associated with ovarian cancer progression but theorigin of its soluble form (sVCAM-1) in serum is not well investigated. The purpose of this study was to elucidate whetherthe concentration of sVCAM-1 in serum correlates with the concentration in ascites, that represents local tumour environment,and with systemic inflammation, various clinicopathological characteristics, and patient outcome.Patients and methods. Thirty-six patients with advanced ovarian cancer were included in the study. Serum forsVCAM-1 analysis was obtained prior to surgery. Ascites samples were collected at the beginning of the operation.Clinical data were collected from patients medical records. sVCAM-1 in samples was analysed by flow cytometricbead-based assay. The mean follow-up period was 11 months (range 0-23) from the time of surgery.Results. Serum sVCAM-1 concentrations are positively correlated to ascites sVCAM-1 concentrations. There was aweakly positive correlation of serum sVCAM-1 with tumour size and no correlation with inflammatory tumour markers,FIGO stage or grade. Higher concentrations of sVCAM-1 were associated with poor disease outcome (death fromovarian cancer) in almost all cases before chemotherapy was started.Conclusion. This is the first study demonstrating that serum concentrations of sVCAM-1 in advanced ovarian cancerpatients correlate with sVCAM-1 concentrations in ascites, thus expressing the biologic potential of malignant diseaseto metastasis, rather than systemic inflammation. Higher serum and ascites sVCAM-1 concentrations might have predictivepotential for different biologic behaviour.

Effect of ionizing radiation on human skeletal muscle precursor cells

Abstract: Background. Long term effects of different doses of ionizing radiation on human skeletal muscle myoblast proliferation, cytokine signalling and stress response capacity were studied in primary cell cultures.Materials and methods. Human skeletal muscle myoblasts obtained from muscle biopsies were cultured and irradiated with a Darpac 2000 X-ray unit at doses of 4, 6 and 8 Gy. Acute effects of radiation were studied by interleukin - 6 (IL-6) release and stress response detected by the heat shock protein (HSP) level, while long term effects were followed by proliferation capacity and cell death.Results. Compared with non-irradiated control and cells treated with inhibitor of cell proliferation Ara C, myoblast proliferation decreased 72 h post-irradiation, this effect was more pronounced with increasing doses. Post-irradiation myoblast survival determined by measurement of released LDH enzyme activity revealed increased activity after exposure to irradiation. The acute response of myoblasts to lower doses of irradiation (4 and 6 Gy) was decreased secretion of constitutive IL-6. Higher doses of irradiation triggered a stress response in myoblasts, determined by increased levels of stress markers (HSPs 27 and 70).Conclusions. Our results show that myoblasts are sensitive to irradiation in terms of their proliferation capacity and capacity to secret IL-6. Since myoblast proliferation and differentiation are a key stage in muscle regeneration, this effect of irradiation needs to be taken in account, particularly in certain clinical conditions.

Virome analysis of irrigation water sources provides extensive insights into the diversity and distribution of plant viruses in agroecosystems

Abstract: Plant viruses pose a significant threat to agriculture. Several are stable outside their hosts, can enter water bodies and remain infective for prolonged periods of time. Even though the quality of irrigation water is of increasing importance in the context of plant health, the presence of plant viruses in irrigation waters is understudied. In this study, we conducted a large-scale high-throughput sequencing (HTS)-based virome analysis of irrigation and surface water sources to obtain complete information about the abundance and diversity of plant viruses in such waters. We detected nucleic acids of plant viruses from 20 families, discovered several novel plant viruses from economically important taxa, like Tobamovirus and observed the influence of the water source on the present virome. By comparing viromes of water and surrounding plants, we observed presence of plant viruses in both compartments, especially in cases of large-scale outbreaks, such as that of tomato mosaic virus. Moreover, we demonstrated that water virome data can extensively inform us about the distribution and diversity of plant viruses for which only limited information is available from plants. Overall, the results of the study provided extensive insights into the virome of irrigation waters from the perspective of plant health. It also suggested that an HTS-based water virome surveillance system could be used to detect potential plant disease outbreaks and to survey the distribution and diversity of plant viruses in the ecosystem.

Gene immunotherapy of colon carcinoma with IL-2 and IL-12 using gene electrotransfer

Abstract: Gene immunotherapy has become an important approach in the treatment of cancer. One example is the introduction of genes encoding immunostimulatory cytokines, such as interleukin 2 and interleukin 12, which stimulate immune cells in tumours. The aim of our study was to determine the effects of gene electrotransfer of plasmids encoding interleukin 2 and interleukin 12 individually and in combination in the CT26 murine colon carcinoma cell line in mice. In the in vitro experiment, the pulse protocol that resulted in the highest expression of IL-2 and IL-12 mRNA and proteins was used for the in vivo part. In vivo, tumour growth delay and also complete response were observed in the group treated with the plasmid combination. Compared to the control group, the highest levels of various immunostimulatory cytokines and increased immune infiltration were observed in the combination group. Long-term anti-tumour immunity was observed in the combination group after tumour re-challenge. In conclusion, our combination therapy efficiently eradicated CT26 colon carcinoma in mice and also generated strong anti-tumour immune memory.

Numerical modelling of macrosegregation in three-dimensional continuous casting of steel billets

Abstract: Macrosegregation presents a considerable defect in the continuous casting of billets and can critically affect the final properties of the product. The numerical modelling can help to predict and better understand the segregation and flow patterns inside the mould. The process is modelled with a physical model described by a set of conservation equations describing the t heat transfer, turbulence, fluid flow, solidification and segregation. A two-equation low-Re k-epsilon model and Abe-Kondoh-Nagano closures are used to close governing equations in this incompressible fluid flow example. The Boussinesq approximation is applied to account for the thermo-solutal buoyancy effects, and the Darcy approximation is applied for the description of the flow through the porous mushy zone. On a microscale, a lever rule solidification model is used to couple liquid fraction, temperature and concentration. The three-dimensional model is solved with the method based on local collocation with multiquadric radial basis functions on seven-nodded subdomains. The aim of this contribution is to explore the three-dimensional macrosegregation patterns of 0.51 wt% carbon steel in the solidified shell of the steel in the mould.

Cell size dynamics and viability of cells exposed to hypotonic treatment and electroporation for electrofusion optimization

Abstract: Background. Various electrofusion parameters have to be adjusted to obtain theoptimal electrofusion efficiency. Based on published data, good electrofusion conditions can be achieved with the hypotonic treatment. However, the duration of the hypotonic treatment before electroporation and buffer hypoosmolarity have to be adjusted in order to cause cell swelling, to avoid regulatory volume decrease and to preserve cell viability. The aims of our study were to determine cell size dynamics and viability of four different cell lines in hypotonic buffer and to study the influence of the electroporation on the selected cell line in hypotonic buffer. Materials and methods. Cell size dynamics of different cell lines exposed to hypotonic buffer and electroporation were analyzed by time-resolvedcell size measurements. The viability of hypotonically treated oržand electroporated cells was determined 24 h after the experiment by a modified crystal violet (CV) viability assay. Results. In our experimental conditions the hypotonic treatment at 100 mOsm was efficient for CHO, V79 and B16-F1 cell lines. The optimal duration of the treatment was between two and five minutes. On the other hand the same hypotonic treatment did not cause cell swelling of NS1 cells. Cell swelling was also observed after electroporation of B16-F1 in isotonic buffer and it was amplified when hypotonic buffer was used. In addition, the regulatory volume decrease was successfully inhibited with electroporation. Conclusions. Cell size dynamicsin hypotonic conditions should be studied for each cell line since they differ in their sensitivity to the hypotonic treatment. The inhibition of cell regulatory volume decrease by electroporation may be beneficial in achieving higher electrofusion efficiency. (Abstract truncated at 2000 characters)
